Dit protocol beschrijft een techniek voor cybridegeneratie uit suspensie-groeiende kankercellen als een hulpmiddel om de rol van mitochondriën in het tumorogene proces te bestuderen.
In de afgelopen jaren is het aantal studies gewijd aan het vaststellen van het verband tussen mitochondriën en kanker aanzienlijk gestegen. Er zijn echter nog meer inspanningen nodig om het verband met veranderingen in mitochondriën en tumorigenese volledig te begrijpen, evenals om tumor-geassocieerde mitochondriale fenotypes te identificeren. Om bijvoorbeeld de bijdrage van mitochondriën in tumorigenese en metastaseprocessen te evalueren, is het essentieel om de invloed van mitochondriën van tumorcellen in verschillende nucleaire omgevingen te begrijpen. Voor dit doel bestaat een mogelijke benadering uit het overbrengen van mitochondriën naar een andere nucleaire achtergrond om de zogenaamde cybride cellen te verkrijgen. In de traditionele cybridisatietechnieken wordt een cellijn zonder mtDNA (ρ0, nucleaire donorcel) opnieuw bevolkt met mitochondriën die zijn afgeleid van enucleated cellen of bloedplaatjes. Het enucleatieproces vereist echter een goede celhechting aan de kweekplaat, een kenmerk dat in veel gevallen gedeeltelijk of volledig verloren gaat in invasieve cellen. Bovendien is een andere moeilijkheid die in de traditionele methoden wordt gevonden, het bereiken van volledige verwijdering van het endogene mtDNA uit de mitochondriale ontvangercellijn om zuivere nucleaire en mitochondriale DNA-achtergronden te verkrijgen, waarbij de aanwezigheid van twee verschillende mtDNA-soorten in het gegenereerde cybride wordt vermeden. In dit werk presenteren we een mitochondriaal uitwisselingsprotocol toegepast op suspensie-groeiende kankercellen op basis van de herbevolking van rhodamine 6G-voorbehandelde cellen met geïsoleerde mitochondriën. Deze methodologie stelt ons in staat om de beperkingen van de traditionele benaderingen te overwinnen en kan dus worden gebruikt als een hulpmiddel om het begrip van de mitochondriale rol in kankerprogressie en metastase uit te breiden.
Het herprogrammeren van het energiemetabolisme is een kenmerk van kanker1 dat voor het eerst werd waargenomen door Otto Warburg in de jaren 19302. Onder aerobe omstandigheden zetten normale cellen glucose om in pyruvaat, dat vervolgens acetyl-coA genereert, de mitochondriale machinerie voedt en cellulaire ademhaling bevordert. Niettemin toonde Warburg aan dat, zelfs onder normoxische omstandigheden, de meeste kankercellen pyruvaat verkregen uit het glycolyseproces omzetten in lactaat, waardoor ze hun weg verleggen om energie te verkrijgen. Deze metabole aanpassing staat bekend als het “Warburg-effect” en stelt sommige kankercellen in staat om te voorzien in hun energetische behoeften voor snelle groei en deling, ondanks het feit dat ATP minder efficiënt wordt gegenereerd dan het aërobe proces 3,4,5. In de afgelopen decennia hebben talrijke werken de implicatie van herprogrammering van het metabolisme in de progressie van kanker ondersteund. Vandaar dat tumor energetica wordt beschouwd als een interessant doelwit tegen kanker1. Als een centrale hub in het energetisch metabolisme en in de toevoer van essentiële voorlopers, spelen mitochondriën een sleutelrol in deze celaanpassingen die we tot op heden slechts gedeeltelijk begrijpen.
In overeenstemming met het bovenstaande zijn mitochondriale DNA (mtDNA) mutaties voorgesteld als een van de mogelijke oorzaken van deze metabole herprogrammering, die zou kunnen leiden tot een verminderde elektronentransportketen (ETC) prestaties6 en zou verklaren waarom sommige kankercellen hun glycolytische metabolisme verbeteren om te overleven. Er is inderdaad gemeld dat mtDNA mutaties in kankercellen accumuleert, aanwezig in ten minste 50% van de tumoren7. Een recente studie uitgevoerd door Yuan et al. meldde bijvoorbeeld de aanwezigheid van hypergemuteerde en afgeknotte mtDNA-moleculen in nier-, colorectale en schildklierkankers8. Bovendien hebben veel werken aangetoond dat bepaalde mtDNA-mutaties geassocieerd zijn met een agressiever tumorfenotype en met een toename van het metastatische potentieel van kankercellen 9,10,11,12,13,14,15,16.
Ondanks de schijnbare relevantie van het mitochondriale genoom in de progressie van kanker, is de studie van deze mutaties en hun bijdrage aan de ziekte een uitdaging geweest vanwege beperkingen in de experimentele modellen en technologieën die momenteel beschikbaar zijn17. Er zijn dus nieuwe technieken nodig om de echte impact van mitochondriaal DNA in de ontwikkeling en progressie van kankerziekten te begrijpen. In dit werk introduceren we een protocol voor transmitochondriale cybridegeneratie uit suspensie-groeiende kankercellen, gebaseerd op de herbevolking van rhodamine 6G-voorbehandelde cellen met geïsoleerde mitochondriën, dat de belangrijkste uitdagingen van traditionele cybridisatiemethoden overwint18,19. Deze methodologie maakt het gebruik van elke kerndonor mogelijk, ongeacht de beschikbaarheid van hun overeenkomstige ρ0-cellijn en de overdracht van mitochondriën van cellen die, volgens de traditionele technieken, moeilijk te enucleeren zouden zijn (d.w.z. niet-aanhangende cellijnen).
Sinds Otto Warburg meldde dat kankercellen hun metabolisme verschuiven en “aerobe glycolyse” versterken3,4 terwijl ze mitochondriale ademhaling verminderen, is de interesse in de rol van mitochondriën in kankertransformatie en -progressie exponentieel gegroeid.
