Directe stochastische optische reconstructiemicroscopie (dSTORM) wordt gebruikt om de typische diffractielimiet van lichtmicroscopie te omzeilen en exosomen op nanometerschaal te bekijken. Het kan worden gebruikt in zowel twee als drie dimensies om exosomen te karakteriseren.
Extracellulaire blaasjes (EV’s) worden door alle celtypen vrijgegeven en spelen een belangrijke rol bij celsignalering en homeostase. De visualisatie van EV’s vereist vaak indirecte methoden vanwege hun kleine diameter (40-250 nm), die onder de diffractielimiet van typische lichtmicroscopie ligt. We hebben een op superresolutiemicroscopie gebaseerde visualisatie van EV’s ontwikkeld om de diffractielimiet in zowel twee als drie dimensies te omzeilen. Met behulp van deze aanpak kunnen we de driedimensionale vorm van EV’s oplossen tot binnen +/- 20 nm resolutie op de XY-as en +/- 50 nm resolutie langs de Z-as. Concluderend stellen we voor dat superresolutiemicroscopie wordt beschouwd als een karakteriseringsmethode van EV’s, inclusief exosomen, evenals omhulde virussen.
Extracellulaire blaasjes (EV’s) zijn membraangebonden blaasjes die door alle celtypen worden afgegeven. Ze bevatten lipiden, eiwitten, metabolieten en nucleïnezuren en dragen deze materialen lokaal over tussen cellen en distaal tussen weefsels en organen. Er zijn drie primaire subtypen van EV’s: apoptotische lichamen, microvesicles en exosomen1,2. Hier richten we onze discussie op exosomen en hun bijbehorende eiwitten.
Exosomen zijn uitgescheiden blaasjes afkomstig van de inwaartse ontluiking van vroege endosomen in het multivesiculaire lichaam (MVB). De MVB smelt vervolgens met het plasmamembraan, waardoor de exosomen in de extracellulaire ruimte vrijkomen om naar andere cellen te reizen3,4. Exosomen bestaan op een spectrum van groottes variërend van 40 tot 150 nm en zijn verrijkt met endosomale transmembraaneiwitten die bekend staan als tetraspaninen (CD9, CD63, CD81), membraangebonden endosomale sorteercomplex dat nodig is voor het transport (ESCRT) en lipide vlot-geassocieerde eiwitten1,2,5,6,7.
Het karakteriseren van de biochemische samenstelling van exosomen is een populair veld geworden voor onderzoekers om hun functionele aard beter te begrijpen. Er bestaan veel methoden voor het visualiseren en karakteriseren van exosomen, waaronder flowcytometrie op nanoschaal, nanodeeltjesvolganalyse (NTA), scanning- en transmissie-elektronenmicroscopie (TEM), oppervlakteplasmonresonantie, resistieve pulsdetectie en traditionele lichtmicroscopie, die elk intrinsieke voor- en nadelen bevatten8,9. TEM en cryo-EM kunnen een op nanometer gebaseerde resolutie bereiken, maar vereisen vaak uitdrogings- en vriesbreekstappen, waardoor EV’s10, 11krimpen of liggen. NTA vertrouwt op lichtverstrooiing, waardoor de karakterisering van honderden EV’s tegelijk mogelijk is, maar is een indirecte meting van de deeltjesgrootte en kan niet gemakkelijk onderscheid maken tussen EV’s, virussen en eiwitaggregaten12,13,14,15,16. Nanoschaal flowcytometrie maakt gebruik van lichtverstrooiing van een excitatiepad, dat vervolgens kan worden vertaald in groottemetingen, maar is een opkomende technologie, en er is weinig consensus over welke grootte van deeltjes zich binnen het lineaire detectiebereik bevinden voor verschillende instrumenten12,17,18.
Traditionele lichtmicroscopie met behulp van fluorescerende eiwitten of kleurstoffen is een van de meest gebruikte technieken voor het visualiseren van subcellulaire compartimenten, eiwitcomplexen en signaleringsmachines in een cel. Hoewel deze techniek nuttig blijkt bij het visualiseren van de lokalisatie van complexen, voorkomt de diffractielimiet van traditionele lichtmicroscopie (ongeveer 250-400 nm) de duidelijke resolutie van eiwitten of structuren in het typische groottebereik van een exosoom (40-150 nm)12,19,20.
Superresolutiemicroscopie, namelijk directe stochastische optische reconstructiemicroscopie (dSTORM), onderscheidt zich van conventionele lichtmicroscopie door gebruik te maken van de fotoschakelbare eigenschappen van specifieke fluoroforen en deze knipperende gebeurtenissen te detecteren om beelden tot nanometerprecisie te reconstrueren21. Fotoswitchinggebeurtenissen worden verzameld met behulp van een detectiecamera met hoge framesnelheid in de loop van tienduizenden individuele belichtingen, en een puntspreidingsfunctie wordt gebruikt om met hoge betrouwbaarheid de exacte locatie van de fotoswitching fluorofoor19,20,22in kaart te brengen. Hierdoor kan dSTORM de diffractielimiet van lichtmicroscopie omzeilen. Verschillende groepen hebben het gebruik van superresolutietechnieken gemeld voor het visualiseren en volgen van exosomen en hun bijbehorende eiwitten22,23,24,25. De uiteindelijke resolutie hangt af van de biofysische eigenschappen van de fluorofoor, maar varieert vaak van +/-10-100 nm langs de XY-as, waardoor een resolutie van één molecuul mogelijk is.
Het vermogen om individuele fluoroforen op deze schaal op de XY-as op te lossen, heeft een revolutie teweeggebracht in de microscopie. Er zijn echter weinig gegevens over de driedimensionale (3D) dSTORM van een exosoom. Daarom hebben we geprobeerd een standaard operationele procedure (SOP) vast te stellen voor dSTORM-gebaseerde visualisatie en karakterisering van gezuiverde EV’s, inclusief exosomen tot nanometerprecisie in 3D.
EV’s zijn een populair studiegebied geworden vanwege hun belangrijke rol in veel intracellulaire processen en cel-naar-cel signalering1,30. Hun visualisatie blijkt echter moeilijk te zijn omdat hun kleine formaat onder de diffractielimiet van lichtmicroscopie valt.
