Ce manuscrit décrit un protocole pour déterminer si l’exposition à l’ozone, un polluant atmosphérique de critères, altère l’efferocytose de macrophage alvéolaire in vivo. Ce protocole utilise des réactifs et des techniques couramment utilisés et peut être adapté à plusieurs modèles de lésions pulmonaires pour déterminer les effets sur l’efférocytose macrophage alvéolaire.
L’ozone (O3) est un polluant atmosphérique qui exacerbe et augmente l’incidence des maladies pulmonaires chroniques. O3 l’exposition est connue pour induire l’inflammation pulmonaire, mais peu est connu concernant la façon dont l’exposition modifie les processus importants pour la résolution de l’inflammation. L’eférocytose est un processus de résolution, par lequel les macrophages phagocytize cellules apoptotiques. Le but de ce protocole est de mesurer l’efferocytosis alvéolaire de macrophage suivant o3-blessure et inflammation de poumon d’O 3-induite. Plusieurs méthodes ont été décrites pour mesurer l’efferocytose ; cependant, la plupart exigent des manipulations ex vivo. Décrit en détail voici un protocole pour mesurer in vivo alvéolaire macrophage efferocytosis 24 h après l’exposition O3, qui évite la manipulation ex vivo des macrophages et sert de technique simple qui peut être utilisé pour représenter avec précision les perturbations dans processus de résolution. Le protocole est une méthode techniquement non intensive et relativement peu coûteuse qui implique l’inhalation de l’Ensemble du corps O3 suivie par l’aspiration oropharyngée des cellules apoptotiques (c.-à-d., les lymphocytes T Jurkat) sous anesthésie générale. L’efferocytose de macrophage alvéolaire est alors mesurée par l’évaluation légère de microscopie des macrophages rassemblés du lavage bronchoalveolar (BAL). L’eférocytose est enfin mesurée par le calcul d’un indice efférocytique. Collectivement, les méthodes décrites quantifient l’activité efférocytique dans le poumon in vivo tout en servant également à analyser les effets négatifs sur la santé de O3 ou d’autres insultes inhalées.
Le poumon est constamment exposé à des insultes environnementales, y compris les particules d’air, les virus, les bactéries et les gaz oxydants qui déclenchent l’inflammation pulmonaire1,2,3. Ces insultes peuvent compromettre l’échange de gaz et induire des lésions tissulaires irréversibles4,5. Les macrophages alvéolaires, qui constituent environ 95% des cellules immunitaires présentes dans les poumons murins et humains à l’homéostasie, sont des régulateurs critiques de l’inflammation pulmonaire après des insultes environnementales1,2, 3,4,5. Les macrophages alvéolaux sont essentiels pendant la défense hôte en phagocytant et en éliminant les agents pathogènes. Récemment, les macrophages alvéolaires ont été montrés pour favoriser l’homéostasie de tissu et la résolution de l’inflammation par l’efferocytose6,7. L’eférocytose est un processus phagocytique dans lequel les macrophages engloutissent et éliminent les cellules apoptotiques8,9,10. L’eférocytose entraîne également la production de médiateurs (c.-à-d. IL-10, TGF-mD, PGE2, et oxyde nitrique) qui augmentent encore le processus, ce qui entraîne la résolution de l’inflammation9,10,11 ,12,16,18. Ce processus est nécessaire pour prévenir la nécrose secondaire et promouvoir l’homéostasie tissulaire12,13,14. Plusieurs études ont établi un lien entre l’eférocytose altérée et diverses maladies pulmonaires chroniques, y compris l’asthme, la maladie pulmonaire obstructive chronique et la fibrose pulmonaire idiopathique8,9,15, 16,17.
O3 est un polluant atmosphérique de critères qui exacerbe et augmente l’incidence des maladies pulmonaires chroniques19,20,21. O3 induit une inflammation pulmonaire et des blessures et est connu pour altérer la phagocytose macrophage alvéolaire des agents pathogènes bactériens22,23. Cependant, on ne sait pas si O3 altère l’efferocytose macrophage alvéolaure. L’étude des altérations o3-induitesdans l’efferocytosis alvéolaire de macrophage fournira un aperçu potentiel de la façon dont l’exposition peut mener à l’incidence et à l’exacerbation chroniques de la maladie pulmonaire. Décrit ci-dessous est une méthode simple pour évaluer l’efferocytosis alvéolifolité de macrophage dans les poumons des souris femelles après exposition aigue d’O3.
La méthode décrite présente plusieurs avantages par rapport à d’autres protocoles d’eférocytose couramment utilisés sur le terrain en éliminant l’utilisation de colorants fluorescents coûteux, de mesures de cytométrie à débit étendu et de manipulation ex vivo des macrophages alvéolaires24 ,25. En outre, ce protocole mesure l’efférocytose alvéolaire de macrophage dans le contexte du microenvironnement de poumon, qui peut influencer la fonction de macrophage.
