Le but de cette méthode est de générer un modèle in vivo de l’angiogenèse tumorale en xénogreffer les cellules tumorales mammifères dans un embryon de poisson zèbre qui a des vaisseaux sanguins fluorescents étiquetés. En imagerie du xénogreffe et des vaisseaux associés, une mesure quantitative de la réponse angiogénique peut être obtenue.
L’angiogenèse tumorale est une cible clé de la thérapie anticancéreuse et cette méthode a été développée pour fournir un nouveau modèle pour étudier ce processus in vivo. Un xénogreffe de poisson zèbre est créé en implantant des cellules de tumeur de mammifères dans l’espace périvitelline de deux jours-post-fertilisation des embryons de poisson-zèbre, suivi par mesurer l’ampleur de la réponse angiogénique observée à un point final expérimental jusqu’à deux jours post-implantation. L’avantage clé de cette méthode est la capacité de quantifier avec précision la réponse angiogénique hôte de l’hôte de l’hôte zébré à la greffe. Cela permet un examen détaillé des mécanismes moléculaires ainsi que la contribution hôte vs tumeur à la réponse angiogénique. Les embryons xénogreffes peuvent être soumis à une variété de traitements, tels que l’incubation avec des médicaments anti-angiogenèse potentiels, afin d’étudier les stratégies pour inhiber l’angiogenèse tumorale. La réponse angiogénique peut également être en direct afin d’examiner des processus cellulaires plus dynamiques. La technique expérimentale relativement peu exigeante, les coûts d’entretien bon marché du poisson zèbre et la chronologie expérimentale courte rendent ce modèle particulièrement utile pour le développement de stratégies pour manipuler l’angiogenèse de tumeur.
L’angiogenèse est l’une des caractéristiques classiques du cancer et représente une cible de la thérapie anticancéreuse1,2. Pour étudier ce processus, des modèles de xénogreffe du cancer ontété créés en implantant des cellules de tumeur de mammifères dans des animaux tels que des souris 3. Un modèle de xénogreffe de poisson zèbre a également été développé, qui implique l’implantation des cellules de tumeur dans 2 joursaprès la fertilisation (dpi) le poisson zèbre qui a comme conséquence la croissance rapide des vaisseaux sanguins de poissons-zèbres dans le xénogreffe 4.
Ce protocole décrit un modèle in vivo de tumeur d’embryon de poisson-zèbre xénogreffe dans lequel la réponse angiogénique peut être exactement quantite à travers le xénogreffe entier. Cette méthode permet à l’investigateur d’examiner, in vivo, les mécanismes moléculaires qui sous-tendent la réponse angiogénique de tumeur. La tractabilité génétique du poisson zèbre permet d’interroger la contribution de l’hôte, tandis que la sélection de différentes lignées de cellules tumorales permet d’examiner la contribution tumorale à l’angiogenèse5,6,7. En outre, comme les larves de poissons zèbres sont perméables à de petites molécules, des inhibiteurs de voie spécifiques peuvent être utilisés ou les bibliothèques de médicaments peuvent être examinés pour identifier de nouveaux inhibiteurs de l’angiogenèse tumorale8,9,10, 11.
Le modèle de xénogreffe d’embryon de poisson zèbre présente des avantages uniques comparés à d’autres modèles de xénogreffe de mammifères. Les xénogreffes de poisson zèbre sont moins chères et plus faciles à exécuter, un grand nombre d’animaux peuvent être examinés et l’imagerie cellulaire vivante permet un examen détaillé du comportement cellulaire4. Contrairement à d’autres modèles in vivo, qui nécessitent jusqu’à plusieurs semaines pour observer une croissance significative des vaisseaux, l’angiogenèse chez les xénogreffes de poissons zèbres peut être observée dans les 24 h suivant l’implantation3,4. Cependant, l’absence d’un système immunitaire adaptatif chez le poisson zèbre embryonnaire, bien que bénéfique au maintien de la xénogreffe, signifie que la réponse immunitaire adaptative et sa contribution à l’angiogenèse tumorale ne peuvent pas être examinées. En outre, l’absence de cellules stromales tumorales, l’incapacité d’implanter orthotopiquement la tumeur et la différence de température d’entretien entre le poisson zèbre et les cellules mammifères sont des faiblesses potentielles de cette méthode. Néanmoins, l’agréabilité de ce modèle pour la formation image vivante et la capacité de quantifier avec précision la réponse angiogénique le rend particulièrement salutaire pour étudier les processus cellulaires qui règlent l’angiogenèse de tumeur in vivo.
La première étape critique du protocole est l’implantation de cellules tumorales. Il est essentiel que les cellules soient injectées dans un endroit qui permettra au xénogreffe de s’implanter avec succès dans l’embryon sans rendre l’embryon edematous.
