Het doel van deze methode is het genereren van een in vivo model van tumor angiogenese door xenotransplanterende tumorcellen van zoogdieren tot een embryo van zebravis dat fluorescently gelabelde bloedvaten heeft. Door beeldvorming van de xenotransplantaatmodellen is en geassocieerde vaten kan een kwantitatieve meting van de angiogene respons worden verkregen.
Tumor angiogenese is een belangrijk doelwit van anti-kankertherapie en deze methode is ontwikkeld om een nieuw model te bieden om dit proces in vivo te bestuderen. Een zebravis xenotransplantaatmodellen is wordt gecreëerd door het implanteren van zoogdieren tumorcellen in de perivitelline ruimte van twee dagen-post-bemesting zebravis embryo’s, gevolgd door het meten van de mate van de angiogene respons waargenomen bij een experimenteel eindpunt tot twee dagen na implantatie. Het belangrijkste voordeel van deze methode is de mogelijkheid om de zebravis host angiogene reactie op het transplantaat nauwkeurig te Kwantificeer. Dit maakt gedetailleerd onderzoek van de moleculaire mechanismen en de bijdrage van gastheer versus tumor aan de angiogene respons mogelijk. De xenografted embryo’s kunnen worden onderworpen aan een verscheidenheid aan behandelingen, zoals incubatie met potentiële anti-angiogenese medicijnen, om strategieën voor de remming van de tumor angiogenese te onderzoeken. De angiogene respons kan ook live-imaged zijn om meer dynamische cellulaire processen te onderzoeken. De relatief onveeleisende experimentele techniek, goedkope onderhoudskosten van zebravis en korte experimentele tijdlijn maken dit model vooral nuttig voor de ontwikkeling van strategieën om de tumor angiogenese te manipuleren.
Angiogenese is een van de klassieke kenmerken van kanker en vertegenwoordigt een doelwit van anti-kankertherapie1,2. Om dit proces te bestuderen, xenotransplantaatmodellen is modellen van kanker zijn gemaakt door het implanteren van zoogdier tumorcellen in dieren zoals muizen3. Een zebravis xenotransplantaatmodellen is model is ook ontwikkeld, waarbij de implantatie van tumorcellen in 2 dagen post fertilisatie (dpi) zebravis die resulteert in een snelle groei van zebravis bloedvaten in de xenotransplantaatmodellen is4.
Dit protocol beschrijft een in vivo zebravis embryo tumor xenotransplantaatmodellen is model waarin de angiogene respons nauwkeurig kan worden gekwantificeerd over de gehele xenotransplantaatmodellen is. Deze methode stelt de onderzoeker in staat om in vivo de moleculaire mechanismen te onderzoeken die de tumor angiogene respons ondersteunen. De genetische traceerbaarheid van de zebravissen maakt het mogelijk de gastheer bijdrage te ondervragen, terwijl de selectie van verschillende tumor cellijnen toestaat dat de tumor bijdrage aan angiogenese ook wordt onderzocht5,6,7. Bovendien, als zebravis larven zijn permeabel voor kleine moleculen, specifieke pathway remmers kunnen worden gebruikt of drug bibliotheken kunnen worden gescreend om te identificeren van de nieuwe remmers van tumor angiogenese8,9,10, 11.
Het zebravis embryo xenotransplantaatmodellen is model presenteert unieke voordelen ten opzichte van andere zoogdier xenotransplantaatmodellen is modellen. Zebravis xenotransplantaten zijn goedkoper en gemakkelijker uit te voeren, grote aantallen dieren kunnen worden onderzocht en live-celbeeldvorming laat gedetailleerd onderzoek van het Celgedrag toe4. In tegenstelling tot andere in vivo-modellen, die tot enkele weken nodig hebben om significante bloedvat groei te observeren, kan angiogenese in zebravis xenotransplantaten worden waargenomen binnen 24 uur na implantatie3,4. Het ontbreken van een adaptief immuunsysteem in embryonale zebravis, terwijl het gunstig is voor het behoud van de xenograft, betekent echter dat de adaptieve immuunrespons en zijn bijdrage aan de tumor angiogenese niet kunnen worden onderzocht. Bovendien zijn het gebrek aan tumor stromale cellen, het onvermogen om de tumor orthotopisch te implanteren en het verschil in onderhouds temperatuur tussen zebravis en zoogdiercellen potentiële zwakke punten van deze methode. Niettemin, de geschiktheid van dit model voor Live beeldvorming en de mogelijkheid om de angiogene respons nauwkeurig te kwantificen maakt het uniek gunstig voor het bestuderen van de cellulaire processen die de tumor angiogenese in vivo reguleren.
De eerste kritieke stap in het protocol is de implantatie van tumorcellen. Het is essentieel dat cellen worden geïnjecteerd in een locatie die de xenotransplantaatmodellen is in staat stelt om met succes in het embryo te implanteren zonder het embryo edematous te maken. Een te anterieure injectie kan de cellen in de richting van het hart laten bewegen, de bloedbaan blokkeren en tot oedeem leiden, terwijl een injectie die te posterieure is, een slecht geïmplanteerde xenograft zal veroorzaken.
