Het huidige protocol beschrijft een methode om extracellulaire blaasjes uit het perifere bloed en vaste weefsels te extraheren met daaropvolgende profilering van oppervlakteantigenen en eiwitladingen.
Circulerende en weefsel-residente extracellulaire blaasjes (EV’s) vertegenwoordigen veelbelovende doelen als nieuwe theranostische biomarkers, en ze komen naar voren als belangrijke spelers in het onderhoud van organismale homeostase en de progressie van een breed spectrum van ziekten. Terwijl het huidige onderzoek zich richt op de karakterisering van endogene exosomen met de endosomale oorsprong, hebben microvesicles die uit het plasmamembraan blossen steeds meer aandacht gekregen in gezondheid en ziekte, die worden gekenmerkt door een overvloed aan oppervlaktemoleculen die de membraansignatuur van oudercellen samenvatten. Hier wordt een reproduceerbare procedure gepresenteerd op basis van differentiële centrifugatie voor het extraheren en karakteriseren van EV’s uit het plasma en vaste weefsels, zoals het bot. Het protocol beschrijft verder de daaropvolgende profilering van oppervlakteantigenen en eiwitladingen van EV’s, die dus traceerbaar zijn voor hun afleidingen en geïdentificeerd met componenten die verband houden met de potentiële functie. Deze methode zal nuttig zijn voor correlatieve, functionele en mechanistische analyse van EV’s in biologische, fysiologische en pathologische studies.
Extracellulaire blaasjes (EV’s) zijn voorgesteld om cel-vrijgegeven lipide bilayer-omsloten extracellulaire structuren1 te definiëren, die een belangrijke rol spelen bij verschillende fysiologische en pathologische gebeurtenissen2. EV’s die door gezonde cellen worden vrijgegeven, kunnen grofweg worden onderverdeeld in twee hoofdcategorieën, namelijk exosomen (of kleine EV’s) gevormd door een intracellulaire endocytische handelsroute3 en microvesicles (of grote EV’s) ontwikkeld door de uitwendige ontluiking van het plasmamembraan van de cel4. Hoewel veel studies zich richten op de functie van EV’s verzameld uit gekweekte cellen in vitro5, zijn EV’s afgeleid van de circulatie of weefsels complexer en heterogener, die het voordeel hebben dat ze de ware toestand van het organisme in vivoweerspiegelen 6. Bovendien kunnen bijna alle soorten weefsels EV’s in vivo produceren, en deze EV’s kunnen fungeren als boodschappers in het weefsel of worden overgedragen door verschillende lichaamsvloeistoffen, vooral het perifere bloed, om systemische communicatie te vergemakkelijken7. EV’s in de bloedsomloop en weefsels zijn ook doelwitten voor ziektediagnose en -behandeling8.
Terwijl exosomen de afgelopen jaren intensief zijn bestudeerd, hebben microvesicles ook belangrijke biologische functies, die gemakkelijk kunnen worden geëxtraheerd zonder ultracentrifugatie, waardoor fundamenteel en klinisch onderzoek wordt bevorderd9. Met name een kritiek probleem met betrekking tot EV’s geïsoleerd uit de bloedsomloop en weefsels is dat ze zijn afgeleid van verschillende celtypen10. Aangezien microvesicles uit het plasmamembraan worden gespoeld en worden gekenmerkt door een overvloed aan celoppervlakmoleculen9, is het gebruik van oudercelmembraanmarkers om de cellulaire oorsprong van deze EV’s te identificeren haalbaar. Concreet kan de flowcytometrie (FC) techniek worden toegepast om membraanmarkers te detecteren. Bovendien kunnen onderzoekers de EV’s isoleren en verdere analyses maken op basis van de functionele ladingen.
Het huidige protocol voorziet in een grondige procedure voor het extraheren en karakteriseren van EV’s uit in vivo monsters. De EV’s worden geïsoleerd via differentiële centrifugatie en de karakterisering van EV’s omvat morfologische identificatie via nanodeeltjesvolganalyse (NTA) en transmissie-elektronenmicroscopie (TEM), oorsprongsanalyse via FC en eiwitladinganalyse via western blot. Het bloedplasma en het maxillaire bot van muizen worden gebruikt als vertegenwoordigers. Onderzoekers kunnen dit protocol raadplegen voor EV’s uit andere bronnen en overeenkomstige wijzigingen aanbrengen.
Bij het bestuderen van de kenmerken, het lot en de functie van EV’s is het cruciaal om EV’s met een hoge opbrengst en lage vervuiling te isoleren. Er bestaan verschillende methoden om EV’s te extraheren, zoals dichtheidsgradiëntcentrifugatie (DGC), grootte-uitsluitingschromatografie (SEC) en immunocapture-assays 4,20.
