Immunohistochemische methoden zijn nuttig in honingbijenonderzoek om het niveau van apoptose en necrose in de midgut- en hypofaryngeale klieren van volwassen bijen te detecteren en te beoordelen.
Honingbijen (Apis mellifera L.) in de korf (verpleegsters en andere bijenkorfbijen) en buiten de korf (foerageerders) worden blootgesteld aan klimaat- en weersveranderingen, verschillende pesticiden, pathogenen en ondervoeding, voornamelijk via de mond en voornamelijk van invloed op het spijsverteringskanaal van volwassen bijen. Om de effecten van dergelijke externe en interne stressoren op honingbijen te begrijpen en te voorkomen, is een nuttige onderzoeksmethode de immunohistochemische methode. Een basisprotocol wordt beschreven om de midgut (ventriculus) en hypofaryngeale klieren (HPG’s) van volwassen bijen voor te bereiden op histologische analyse. Een gedetailleerde methodologie wordt beschreven om het niveau van celbeschadiging te beoordelen en necrose te onderscheiden van geprogrammeerde celdood (apoptose) als een natuurlijk proces van weefselregeneratie. De resultaten van volwassen honingbijenbehandeling met oxaalzuur en pesticiden (insecticide en acaricide) en de bepaling van celdood in de ventriculus en HPG’s worden gepresenteerd. Ook de voor- en nadelen van de methodiek komen aan bod.
Honingbijen (Apis mellifera L.) zijn, naast andere wilde bestuivers, de belangrijkste bestuivers van landbouwgewassen. Gedurende duizenden jaren heeft de veranderende omgeving bijen beïnvloed om hun morfologie, fysiologie, gedrag en tolerantie aan te passen aan verschillende pathogenen en parasieten. Daarom hebben honingbijen een zeer divers scala aan soorten en ondersoorten over de hele wereld ontwikkeld1. Deze resultaten komen overeen met eerdere bevindingen, dat er genetische variatie is in de structuur van het spijsverteringskanaal van de honingbij, maar suggereren ook dat veranderingen van het midgut te wijten zijn aan omgevingsfactoren 2,3.
Het spijsverteringskanaal van de honingbij bestaat uit drie hoofdonderdelen: foregut, midgut (ventriculus) en hindgut4. De ventriculus is een essentieel orgaan voor de vertering van stuifmeel en nectar/honing; in het achterbeen vindt osmotische controle plaats door absorptie van water en ionen2. De hypofaryngeale klieren (HPG’s) van honingbijwerkers bevinden zich in het hoofd en synthetiseren en scheiden koninginnengeleicomponenten af om het broed, de koningin en leden van de kolonie te voeden. Hun grootte verandert met leeftijd en taken en is afhankelijk van de juiste voeding (kwaliteit stuifmeel). Verpleegsters van 6 tot 18 dagen voeren broedopfok uit en de grootte van HPG’s neemttoe met 5,6. Bij foerageerbijen degenereren de HPG’s en scheiden ze alleen enzymen af die belangrijk zijn om de complexe suikers om te zetten in eenvoudige (α-glucosidases, leucine arylamidase, invertase) in honing7.
Honingbijen worden blootgesteld aan verschillende biotische en abiotische stressoren8, en het spijsverteringskanaal kan worden beïnvloed door verschillende negatieve stimulerende middelen. De eerste barrière die het organisme beschermt tegen pathogenen is het peritrofe membraan in het midgut, dat bestaat uit darmslijmvlies om te beschermen tegen pathogenen4. De ontwikkeling en functie van HPG’s zijn afhankelijk van dieet, leeftijd en kolonieconditie9 en worden beïnvloed door insecticiden, acariciden10 en pathogenen 11,12,13. Acaricideresiduen in de korf als gevolg van varroabestrijding en pesticiden uit het milieu beïnvloeden foerageerbijen en verpleegbijen14,15. De grootste bedreiging voor honingbijenkolonies is de mijt Varroa-destructor, zowel als vector van virussen die bijdragen aan kolonieverliezen16 als als consument van het vetlichaam van de gastheer (een belangrijk vitaal orgaan in honingbijen), wat bijgevolg het lichaam en de koloniefuncties van het individu beïnvloedt17.
Intensieve landbouwgrondhabitats kunnen echter op korte termijn zorgen voor een voedselvoorziening voor honingbijen. Daarom moeten agromilieuregelingen de beschikbaarheid van honingbloemen in landbouwlandschappen vergroten18. Om de morfologie van verschillende ondersoorten 6,19,20,21 of subletale effecten van deze factoren op cel- of weefselniveau, met name midgut en HPG’s, te beoordelen, zijn histologische en immunohistochemische methoden praktisch en voldoende nauwkeurig om te worden gebruikt in histologisch onderzoek bij honingbijen.
In levende organismen wordt celdood gedefinieerd als apoptose of necrose25 en kan gepaard gaan met autofagie26. Het verschil tussen apoptotische en necrotische cellen is dat apoptose een vorm van geprogrammeerde celdood is en voorkomt in normale cellen, terwijl necrose optreedt als gevolg van dodelijke aandoeningen (bijv.
