Het doel van dit manuscript is om het KitV558Δ/+ muismodel en technieken voor succesvolle dissectie en verwerking van muismonsters te beschrijven.
Gastro-intestinale stromale tumor (GIST) is het meest voorkomende menselijke sarcoom en wordt meestal aangedreven door een enkele mutatie in de KIT-receptor. Voor alle tumortypen zijn talloze muismodellen ontwikkeld om de volgende generatie kankertherapieën te onderzoeken. In GIST gebruiken de meeste in vivo studies echter xenograft muismodellen die inherente beperkingen hebben. Hier beschrijven we een immunocompetent, genetisch gemanipuleerd muismodel van gastro-intestinale stromale tumor met een KitV558Δ / + -mutatie. In dit model wordt mutant KIT, het oncogen dat verantwoordelijk is voor de meeste GISTs, aangedreven door zijn endogene promotor die leidt tot een GIST die het histologische uiterlijk en immuuninfiltraat nabootst dat wordt gezien in menselijke GISTs. Bovendien is dit model met succes gebruikt om zowel gerichte moleculaire als immuuntherapieën te onderzoeken. Hier beschrijven we het fokken en onderhouden van een KitV558Δ/+ muizenkolonie. Daarnaast beschrijft dit artikel de behandeling en verkrijging van GIST, drainerende mesenteriale lymfeklier en aangrenzende blindedarm in KitV558Δ / + -muizen, evenals monstervoorbereiding voor moleculaire en immunologische analyses.
GIST is het meest voorkomende sarcoom bij mensen met een incidentie van ongeveer 6.000 gevallen in de Verenigde Staten van Amerika1. GIST lijkt afkomstig te zijn van de gastro-intestinale pacemakercellen die de interstitiële cellen van Cajal worden genoemd en wordt meestal aangedreven door een enkele mutatie in de tyrosinekinase KIT of PDGFRA2. Chirurgie is de steunpilaar van de behandeling voor GIST en kan curatief zijn, maar patiënten met een gevorderde ziekte kunnen worden behandeld met de tyrosinekinaseremmer (TKI), imatinib. Sinds de introductie meer dan 20 jaar geleden heeft imatinib het behandelingsparadigma in GIST getransformeerd, waardoor de overleving bij gevorderde ziekte is verbeterd van 1 naar meer dan 5 jaar 3,4,5. Helaas is imatinib zelden curatief vanwege verworven KIT-mutaties, dus nieuwe behandelingen zijn nodig voor deze tumor.
Muismodellen zijn een belangrijk onderzoeksinstrument bij het onderzoek naar nieuwe therapieën bij kanker. Meerdere subcutane xenograft- en patiënt-afgeleide xenograftmodellen zijn ontwikkeld en onderzocht in GIST 6,7. Immunodeficiënte muizen vertegenwoordigen echter niet volledig menselijke GIST, omdat GISTs differentiële immuunprofielen hebben, afhankelijk van hun oncogene mutatie, en het veranderen van de gastro-intestinale tumormicro-omgeving verbetert de effecten van TKI-therapie 8,9. De KitV558Δ/+ muis heeft een heterozygote kiembaandeletie in Kit exon 11, die codeert voor het juxtamembrane domein, de meest gemuteerde plaats in menselijke GIST10. KitV558Δ/+ muizen ontwikkelen een enkele cecale GIST met 100% penetrantie, en tumoren hebben vergelijkbare histologie, moleculaire signalering, immuuninfiltratie en respons op therapie als menselijke GIST 8,11,12,13. Het KitV558Δ/+ muismodel is een krachtig onderzoeksinstrument in de moleculaire en immunologische analyse van GIST. Hoewel de fokstrategie een enkele kruising vereist, vereist het gebruik van KitV558Δ / + muiscohorten in experimenten die de tumorrespons analyseren, uitgebreide fokkerij.
