Om nieuwe regelgevers van transcriptiefactoren te identificeren, ontwikkelden we een benadering om arrayed lentivirale of retrovirale RNAi-bibliotheken te screenen met behulp van een dual-luciferase-gebaseerde transcriptie-verslaggever test. Deze aanpak biedt een snelle en relatief goedkope manier om honderden kandidaten in één experiment te screenen.
Transcriptie factoren kunnen veranderen de expressie van tal van doelgenen die een verscheidenheid van downstream processen waardoor ze goede doelen voor anti-kanker therapieën beïnvloeden. Echter, direct gericht transcriptie factoren is vaak moeilijk en kan leiden tot nadelige bijwerkingen als de transcriptie factor nodig is in een of meer volwassen weefsels. Het identificeren van upstream regulatoren die afwijkend activeren transcriptie factoren in kankercellen biedt een meer haalbaar alternatief, vooral als deze eiwitten zijn gemakkelijk te drogeren. Hier beschrijven we een protocol dat kan worden gebruikt om arrayed medium-scale lentivirale bibliotheken en een dual-luciferase-gebaseerde transcriptie verslaggever assay te combineren om nieuwe regelgevers van transcriptie factoren in kankercellen te identificeren. Onze aanpak biedt een snelle, eenvoudige en goedkope manier om honderden genen in één experiment te testen. Om het gebruik van deze aanpak aan te tonen, hebben we een scherm uitgevoerd van een arrayed lentivirale RNAi-bibliotheek met verschillende regelgevers van Ja-geassocieerdeiwit (YAP) en transcriptieco-activator met PDZ-bindend motief (TAZ), twee transcriptieco-activators die de downstream-effectoren van het Hippo-pad zijn. Deze benadering kan echter worden aangepast om te screenen op regelgevers van vrijwel elke transcriptiefactor of cofactor en kan ook worden gebruikt om CRISPR/CAS9-, cDNA- of ORF-bibliotheken te screenen.
Het doel van deze test is om virale bibliotheken te gebruiken om regelgevers van transcriptiefactoren op een relatief snelle en goedkope manier te identificeren. Afwijkende transcriptieactiviteit wordt geassocieerd met kanker en metastase1,,2,3,4,5,6, dus gericht op transcriptiefactoren in kankercellen is een veelbelovende therapeutische benadering. Echter, transcriptie factoren zijn vaak moeilijk te richten farmacologisch7 en velen zijn nodig voor de normale cellulaire functie in volwassen weefsels8,9,10. Gericht op de kanker-geassocieerde trajecten die aberrantly activeren transcriptie factoren om ziekte te rijden is een meer haalbare aanpak met het potentieel om minder ernstige bijwerkingen hebben. De commerciële beschikbaarheid van arrayed lentivirale en retrovirale RNAi, CRISPR/CAS9, cDNA, of ORF bibliotheken stelt onderzoekers in staat om het belang van talrijke genen te testen in een enkel experiment. Er is echter een betrouwbare uitlezing voor gewijzigde transcriptieactiviteit vereist.
Hier beschrijven we het gebruik van een dual-luciferase-gebaseerde transcriptie reporter test en arrayed lentivirale bibliotheken om eiwitten die transcriptie factoren in kankercellen te reguleren identificeren. In deze test worden shRNAs die zich richten op kanker-geassocieerde genen geleverd aan zoogdierkankercellen via lentivirale transductie en cellen worden geselecteerd voor stabiele integratie met behulp van puromycine. De cellen worden vervolgens getransfecteerd met een verslaggever constructie die firefly luciferase gedreven door een promotor die specifiek is voor de transcriptie factor die wordt onderzocht en een controle constructie die Renilla luciferase uitdrukt van een constitutief actieve promotor die niet reageert op de transcriptie factor wordt onderzocht. We demonstreren deze aanpak met een proof-of-concept scherm voor regelgevers van YAP en TAZ, de kritische downstream-effectors van het Hippo-pad8,10,11. Abnormale activiteit van YAP en TAZ bevordert verschillende stappen van de uitgezaaide cascade11 en wordt waargenomen bij veel vormen van kanker11,12,13. Echter, hoe YAP en TAZ worden aberrantly geactiveerd in sommige kankercellen is nog niet volledig begrepen. YAP en TAZ binden geen DNA, maar worden in plaats daarvan aangeworven om promotors door andere transcriptiefactoren. Leden van de TEA-domein (TEAD)-familie van transcriptiefactoren zijn de belangrijkste bindende partners voor YAP en TAZ en zijn van cruciaal belang voor de meeste YAP- en TAZ-afhankelijke functies. Onze verslaggever constructie drukt firefly luciferase van een YAP / TAZ-TEAD-responsieve promotor en eerdere studies hebben aangetoond dat het getrouw veranderingen in YAP-TEAD en TAZ-TEAD transcriptie activiteit2,14,15detecteert .15
Onze aanpak is snel, medium-throughput, en vereist geen screening faciliteiten, geautomatiseerde robots, of diepe volgorde van gepoolde bibliotheken. De kosten zijn relatief laag en er zijn tal van commercieel beschikbare bibliotheken om uit te kiezen. De benodigde apparatuur en reagentia zijn ook relatief standaard in de meeste laboratoria. Het kan worden gebruikt om te screenen voor regelgevers van vrijwel elke transcriptie factor als een luciferase-gebaseerde verslaggever bestaat of wordt gegenereerd. We gebruiken deze benadering om shRNAs in kankercellen te screenen, maar elke cellijn die met redelijke efficiëntie kan worden getransfecteerd, kan worden gebruikt met elk type arrayed-bibliotheek.
In deze studie tonen we een aanpak voor medium-throughput screening van arrayed virale bibliotheken in combinatie met een dual-luciferase-gebaseerde transcriptie reporter test die kan worden gebruikt om te identificeren en testen van nieuwe toezichthouders van transcriptie factoren.
