De gepresenteerde methode combineert de kwantitatieve analyse van DNA Double-strand Breaks (DSBs), celcyclus verdeling en apoptosis om de celcycluspecifieke evaluatie van de inductie en reparatie van DSB mogelijk te maken, evenals de gevolgen van herstel fouten.
De gepresenteerde methode of licht gewijzigde versies zijn bedacht om specifieke behandelings responsen en bijwerkingen van verschillende anti-kankerbehandelingen te bestuderen, zoals gebruikt in de klinische oncologie. Het maakt een kwantitatieve en longitudinale analyse van de DNA-schade reactie mogelijk na genotoxische stress, zoals geïnduceerd door radiotherapie en een veelheid aan anti-kanker drugs. De methode dekt alle stadia van de DNA-schade reactie, het verstrekken van eindpunten voor inductie en reparatie van DNA dubbel strand Breaks (DSBs), arrestatie van de celcyclus en celdood door Apoptosis in geval van herstel falen. Het combineren van deze metingen geeft informatie over de celcyclus afhankelijke behandelingseffecten en maakt zo een grondige studie mogelijk van de wisselwerking tussen cellulaire proliferatie en het omgaan met DNA-beschadiging. Aangezien het effect van veel kankertherapieën, waaronder chemotherapeutische middelen en ioniserende straling, beperkt is tot of sterk varieert naargelang de specifieke celcyclus fasen, vertrouwen correlatieve analyses op een robuuste en haalbare methode om de behandelingseffecten te beoordelen op het DNA in een celcyclus-specifieke manier. Dit is niet mogelijk met assays met één eindpunt en een belangrijk voordeel van de gepresenteerde methode. De methode is niet beperkt tot een bepaalde cellijn en is grondig getest in een veelheid van tumor en normale weefsel cellijnen. Het kan op grote schaal worden toegepast als een uitgebreide genotoxiciteitstest op vele gebieden van oncologie naast radio-oncologie, waaronder milieueffectbeoordeling, geneesmiddelen screening en evaluatie van genetische instabiliteit in tumorcellen.
Het doel van oncologie is om kankercellen te doden of te deactiveren zonder de normale cellen te schaden. Veel therapieën induceren direct of indirect genotoxische stress in kankercellen, maar ook bij sommige zich uitstrekken in normale cellen. Chemotherapie of gerichte geneesmiddelen worden vaak gecombineerd met radiotherapie om de radio gevoeligheid van de bestraalde tumor1,2,3,4,5te verbeteren, wat een reductie van de stralingsdosis om normale weefselbeschadiging te minimaliseren.
Ioniserende straling en andere genotoxische middelen induceren verschillende soorten DNA-schade, waaronder basis modificaties, strand verknopingen en enkel-of dubbel strand pauzes. DNA Double-strand Breaks (DSBs) zijn de ernstigste DNA laesies en hun inductie is de sleutel tot het celdodende effect van ioniserende straling en diverse cytostatica in radio chemotherapie. Dsbs schaden niet alleen de integriteit van het genoom, maar bevorderen ook de vorming van mutaties op6,7. Daarom hebben verschillende DSB-reparatie trajecten en mechanismen om onherstelbaar beschadigde cellen zoals apoptosis te elimineren tijdens de evolutie ontwikkeld. De gehele DNA-schade respons (DDR) wordt gereguleerd door een complex netwerk van signalerings trajecten die reiken van DNA-schade herkenning en celcyclus arrestatie om DNA-herstel mogelijk te maken, tot geprogrammeerde celdood of inactivatie in geval van reparatie storing8.
De gepresenteerde Flow-cytometrische methode is ontwikkeld om de DDR na genotoxische stress te onderzoeken in een uitgebreide test die de inductie en reparatie van DSB dekt, evenals de gevolgen van herstel fouten. Het combineert de meting van de veeltoegepaste DSB marker γH2AX met analyse van de celcyclus en inductie van apoptosis, met behulp van klassieke subG1 analyse en specifiekere evaluatie van caspase-3 activering.
De combinatie van deze eindpunten in één assay vermindert niet alleen de tijd, arbeid en kosten uitgaven, maar maakt ook de celcyclus-specifieke meting van DSB inductie en reparatie, evenals caspase-3 activering. Dergelijke analyses zijn niet mogelijk met onafhankelijk uitgevoerde testen, maar ze zijn zeer relevant voor een uitgebreid begrip van de DNA-schade respons na genotoxische stress. Veel anti-kanker drugs, zoals cytostatische verbindingen, zijn gericht tegen het verdelen van cellen en hun efficiëntie is sterk afhankelijk van de fase van de celcyclus. De beschikbaarheid van verschillende DSB reparatie processen is ook afhankelijk van de celcyclus fase en traject keuze die essentieel is voor de reparatie nauwkeurigheid, en op zijn beurt bepaalt het lot van de cel9,10,11, 12. Bovendien is celcycle-specifieke meting van DSB-niveaus nauwkeuriger dan gepoolde analyse, omdat DSB-niveaus niet alleen afhankelijk zijn van de dosis van een genotoxische verbinding of straling, maar ook van het DNA-gehalte van de cel.
De methode is gebruikt om de werkzaamheid van verschillende radio therapieën te vergelijken om de resistentiemechanismen in multiform glioblastoom13 te overwinnen en het samenspel tussen ioniserende straling en gerichte geneesmiddelen in Osteosarcoom te ontleden14,15 en atypische teratoïde rhabdoïde kankers16. Bovendien is de beschreven methode op grote schaal gebruikt voor het analyseren van bijwerkingen van radio-en chemotherapie op mesenchymale stamcellen17,18,19,20,21, 22,23,24, die essentieel zijn voor de reparatie van de behandeling veroorzaakte normale weefselbeschadiging en een mogelijke toepassing in regeneratieve geneeskunde.
De aanbevolen methode is eenvoudig te gebruiken en biedt een snelle, nauwkeurige en reproduceerbare meting van de DNA-schade respons inclusief dubbele-strand break (DSB) inductie en reparatie, celcyclus effecten en apoptotische celdood. De combinatie van deze eindpunten biedt een vollediger beeld van hun onderlinge relaties dan individuele assays.
