We beschrijven een methode voor het gebruik van Multi parameter flow cytometrie te detecteren mitochondriale reactieve zuurstof soorten (ROS) in Murine gezonde hematopoietische stam en voorlopercellen (HSPCs) en leukemie cellen van een muismodel van acute myeloïde leukemie (AML) gedreven door MLL-AF9.
We presenteren een flowcytometrische benadering voor het analyseren van mitochondriale ROS in diverse levende beenmerg (BM)-afgeleide stam-en voorlopercellen van gezonde muizen en muizen met AML aangedreven door MLL-AF9. Specifiek beschrijven we een tweestapskleurings proces, waarbij gezonde of leukemie BM-cellen eerst worden gekleurd met een fluorogene kleurstof die mitochondriale Super oxiden detecteert, gevolgd door kleuring met met fluorochrome verbonden monoklonale antilichamen die worden gebruikt verschillende gezonde en kwaadaardige hematopoietische voorlopercellen te onderscheiden. We bieden ook een strategie voor het verwerven en analyseren van de monsters door flow Cytometry. Het gehele protocol kan worden uitgevoerd in een tijdsbestek van slechts 3-4 h. We markeren ook de belangrijkste variabelen om te overwegen, evenals de voordelen en beperkingen van het bewaken van ROS productie in het mitochondriale compartiment van levende hematopoietische en leukemie stam en voorlopercellen subpopulaties met behulp van fluorogenic kleurstoffen doorstroming cytometrie . Bovendien presenteren we gegevens die mitochondriale ROS overvloed varieert tussen verschillende gezonde HSPC subpopulaties en leukemie voor ouders en bespreken de mogelijke toepassingen van deze techniek in hematologisch onderzoek.
Reactieve zuurstof soorten (ROS) zijn zeer reactieve moleculen afgeleid van moleculaire zuurstof. De meest goed gedefinieerde cellulaire locatie van ROS productie is de mitochondriën, waar elektronen die passeren de elektronentransport keten (ETC) tijdens oxidatieve fosforylering (OXPHOS) worden geabsorbeerd door moleculaire zuurstof die leidt tot de vorming van een specifieke type ROS genaamd Super oxiden1. Door de acties van een reeks enzymen, genaamd superoxide dismutases of sod’s, worden super oxiden omgezet in waterstof peroxiden, die vervolgens worden geneutraliseerd in water door enzymen zoals katalase of glutathion peroxiasen (GPX). Verstoringen in ROS-regelgevende mechanismen kunnen leiden tot de overtollige productie van ROS, vaak aangeduid als oxidatieve stress, die schadelijke en potentieel dodelijke cellulaire gevolgen zoals Macromolecuul schade hebben (dat wil zeggen, DNA, eiwitten, lipiden). Bovendien is oxidatieve stress gerelateerd aan verschillende pathologieën, zoals diabetes, ontstekingsziekten, veroudering en tumoren2,3,4. Te handhaven redox homeostase en oxidatieve stress te voorkomen, cellen bezitten een verscheidenheid van ROS-regulerende mechanismen5.
Fysiologische niveaus van bepaalde ROS zijn noodzakelijk voor goede embryonale en volwassen Hematopoiesis6. Echter, overtollige ROS wordt geassocieerd met DNA-schade, cellulaire differentiatie en uitputting van het hematopoietische stam en voorlopercellen pool. Er is ook bewijs dat veranderingen in redox biologie kunnen verschillen tussen leukemie en gezonde cellen. Bijvoorbeeld, Ros niveaus neigen te zijn hoger in acute myeloïde leukemie (AML) cellen ten opzichte van hun gezonde tegenhangers en andere studies hebben gesuggereerd dat leukemie stamcellen een lage steady-state niveau van Ros voor overleving7,8behouden. Belangrijk, strategieën voor therapeutisch kapitaliseren op deze redox verschillen hebben aangetoond belofte in verschillende menselijke kanker instellingen9,10. Daarom, assays die het mogelijk maken voor de beoordeling van ROS niveaus in muismodellen kunnen verbeteren ons begrip van hoe deze soorten bijdragen aan cellulaire fysiologie en ziekte pathogenese, alsmede mogelijk bieden een platform voor de beoordeling van de effectiviteit van nieuwe redox-gerichte anti-kankertherapieën.
Fluorogene kleurstoffen die zijn ontwikkeld voor de detectie van Ros worden vaak geëvalueerd in vaste cellen door microscopie of in levende cellen door flow flowcytometrieonderzoeken22.
