Dit protocol beschrijft het enten van C57BL / 6J muizen met de EGD stam van Listeria monocytogenes (L. monocytogenes) en interferon-γ (IFN-γ) responsen door natuurlijke killer (NK) cellen, natuurlijke killercellen (NKT) cellen te meten, en adaptieve T-lymfocyten na de infectie. Dit protocol beschrijft ook hoe om te overleven onderzoek bij muizen na infectie met een aangepaste LD 50 dosis van de ziekteverwekker.
L. monocytogenes is een gram-positieve bacterie die een oorzaak van voedsel overgedragen ziekte bij mensen. Experimentele infectie van muizen met deze ziekteverwekker zeer informatief over de rol van het aangeboren en adaptieve immuunsysteem cellen en specifieke cytokinen in gastheer immuniteit tegen intracellulaire pathogenen zijn. Productie van IFN-γ door aangeboren cellen tijdens subletale infectie met L. monocytogenes is belangrijk voor het activeren van macrofagen en snelle bestrijding van het pathogeen 1-3. Bovendien, IFN-γ productie door adaptieve geheugen lymfocyten is belangrijk voor het vullen van de activering van het aangeboren cellen na 4 herinfectie. L. monocytogenes infectie model vormt dus een groot hulpmiddel voor het onderzoeken of er nieuwe therapieën die zijn ontworpen om IFN-γ verhogen van invloed zijn op IFN-γ responsen in vivo en zijn productieve biologische effecten zoals het verhogen van bacteriële verwijdering en overleving verbeteren muis vaninfectie. Hier beschreven is een eenvoudig protocol voor hoe intraperitoneale infectie van C57BL / 6J muizen met de EGD stam van L. monocytogenes te voeren en IFN-γ productie door NK-cellen, NKT-cellen en adaptieve lymfocyten door flowcytometrie gemeten. Bovendien worden procedures beschreven: (1) groeien en voorbereiden van de bacteriën voor inoculatie (2) meten bacteriële verontreiniging in de milt en lever, en (3) overleving maatregel dier eindpunten. Representatieve gegevens worden ook verschaft om te illustreren hoe deze infectie model kan worden gebruikt om het effect van specifieke middelen die IFN-γ reacties op L. monocytogenes en overleving van muizen uit deze infectie te testen.
IFN-γ is een cytokine die cruciaal is voor het mediëren van immuniteit tegen intracellulaire pathogenen en voor het regelen tumorgroei 5. Het belang van deze cytokine in bacteriële resistentie blijkt uit de observatie dat de mens met mutaties in de IFN-γ signaalweg zijn zeer gevoelig voor infectie met mycobacteriën en Salmonella 6. Evenzo muizen die zowel IFN-γ of IFN-γ receptor vertonen defecten in weerstand tegen mycobacteriën 7-9 en andere intracellulaire pathogenen waaronder L. monocytogenes 10,11, Leishmania major 12, Salmonella typhimurium 13 en 11 bepaalde virussen. Naast bestrijding pathogenen, IFN-γ speelt een cruciale rol in gastheer-afweer tegen tumoren 14. Hoewel hogere productie van IFN-γ gunstig in verband met infectie of kanker is langdurige productie van dit cytokine verbonden to de ontwikkeling van systemische auto-immuniteit 15-17 en de versnelling van diabetes type I in de niet-obese diabetische muismodel 18.
De belangrijkste bronnen van IFN-γ omvatten NK-cellen, NKT-cellen, γδ T-cellen, T helper 1 (Th1) cellen en cytotoxische T-lymfocyten (CTL) 5,19,20. IFN-γ verhoogt zowel aangeboren als adaptieve immuniteit door: (1) opreguleren major histocompatibility complex (MHC) klasse I en II expressie, (2) het verhogen van de expressie van co-stimulatoire moleculen op antigeen presenterende cellen, (3) verbetering macrofaag fagocytose en de productie van pro-inflammatoire cytokines en microbiële factoren (bijvoorbeeld stikstofoxide en reactieve zuurstofsoorten), (4) het bevorderen van de differentiatie van naïeve CD4 + T-cellen in Th1 effectorcellen, (5) het bevorderen antilichaamklasse omschakeling immunoglobuline ( ig) 2a en IgG3 (in muizen), (6) het induceren van de productie van chemokinen immuuncellen rekruteren naar sites infection, en (7) verbetering van NK-cellen en CTL-reacties 5,19. Gezien het grote belang van IFN-γ in de gastheer respons op pathogenen en tumoren is recombinant IFN-γ getest voor de behandeling van verschillende infecties en maligniteiten (besproken in 19). Vanwege systemische toediening van IFN-γ of Th1 bevorderen cytokine interleukine-12 (IL-12) is geassocieerd met bijwerkingen en dosis-gerelateerde toxiciteit 19,21 is er belangstelling voor de ontwikkeling van alternatieve strategieën voor IFN-γ-productie door verhogen immuuncellen. Ontwikkeling van nieuwe biologische middelen en kleine moleculen nodig vivo screening hulpmiddelen om te testen of dergelijke middelen verhogen IFN-γ productie tijdens een immuunrespons en of dit neer zinvolle biologische effecten, zoals toename in overleving van dieren.
Experimentele infectie van muizen met de gram-positieve bacterie L. monocytogenes is een instrumentale model geweestontcijferen van de rol van IFN-γ in gastheer-immuniteit tegen intracellulaire pathogenen 1,22. Infectie van muizen met het pathogeen intraveneus of intraperitoneaal (ip) leidt tot de snelle verspreiding van de bacteriën aan de milt en lever, waar ze raken geïnternaliseerd door macrofagen en hepatocyten met piek bacteriële belasting in de milt die tussen 3 en 4 dagen na de infectie 1,3,22. Productie van IFN-γ NK-cellen is belangrijk voor macrofaagactivering en vroege resistentie tegen het pathogeen 3; echter bij hoge infectieuze doses productie van IFN-γ kan ook schadelijk pathogeen speling 23 zijn. NKT cellen zijn ook een bron van IFN-γ in de milt en lever tijdens de vroege beheersing van pathogenen 2,24 en deze productie is aangetoond dat amplificeren IFN-γ productie door andere celtypen, waaronder NK-cellen 2. Anderzijds, later werkende adaptieve T-lymfocyten, CD8 + T-cellen in partiname op, zijn belangrijk voor het bemiddelen van de goedkeuring van de ziekteverwekker en bescherming tegen herinfectie 1,4,22.
Deze infectie model voor onderzoekers om een aantal redenen (besproken in 1) is. Eerste infectie met het pathogeen is zeer reproduceerbaar en induceert een sterke Th1 en cellulaire immuunrespons. Anderzijds in subletale infecties, bacteriële verontreiniging wordt geconcentreerd in de lever en milt die gemakkelijk kan worden gemeten. Ten derde kan de ziekteverwekker veilig worden behandeld onder Biosafety Level 2 (BSL2) omstandigheden. Ten vierde, het organisme en de immuunrespons die wordt gegenereerd zijn uitgebreid gekarakteriseerd. Tenslotte zijn verschillende mutant en genetisch gemodificeerde stammen ontwikkeld die beschikbaar zijn voor gebruik.
Hier beschreven is een eenvoudig protocol voor inoculatie van C57BL / 6J muizen met de EGD stam van L. monocytogenes en 25 voor het meten van IFN – γ reresponsies door NK, NKT en adaptieve lymfocyten na infectie. Ook beschreven is hoe bacteriële verontreiniging in de milt en lever meten nadat subletale infectie en overleving studies na infectie uit te voeren met een gewijzigde LD 50 dosis van de ziekteverwekker. Tenslotte zijn representatieve gegevens weergegeven hoe dit protocol kan worden gebruikt om het effect van nieuwe behandelingen IFN-γ reacties en muis overleving screenen van L. monocytogenes infectie.
