Affinitätsreinigung von markierten Proteinen in Kombination mit der Massenspektrometrie (APMS) ist eine leistungsfähige Methode für die systematische Kartierung von Protein-Wechselwirkung Netzwerken und zur Untersuchung der mechanistischen Grundlagen von biologischen Prozessen. Hier beschreiben wir eine optimierte sequentielle Peptid Affinität (SPA) APMS Verfahren für das Bakterium entwickelt<em> Escherichia coli</em> Das verwendet werden kann, zu isolieren und zu charakterisieren stabilen Multi-Protein-Komplexen zur Homogenität, auch ausgehend von einer geringen Kopienzahl pro Zelle ist.

Dieser Ansatz ist erfolgreich in evolutionär diversen Organismen, einschließlich Hefe, Fliegen, Würmern, Säugetierzellen, Bakterien und 1-6 aufgebracht. Insbesondere haben wir eine Carboxy-terminale Peptid Sequentielle Affinity (SPA) dualen Markierungssystem für Affinitäts-Reinigungskatalysator native Protein-Komplexen aus kultivierten gramnegativen Escherichia coli erzeugt wird, unter Verwendung genetically Spann Gastlabor Stämme, die für genomweite Untersuchungen der grundlegenden Biologie und konservierte Prozesse der Prokaryoten 1, 2, 7 gut geeignet.
