Dit protocol biedt gedetailleerde instructies voor het vaststellen van muriene dunne darm organoïden, het isoleren van type-1 aangeboren lymfoïde cellen uit de muriene dunne darm lamina propria, en het vaststellen van 3-dimensionale (3D) co-culturen tussen beide celtypen om bi-directionele interacties tussen intestinale epitheelcellen en type-1 aangeboren lymfoïde cellen te bestuderen.
Complexe co-culturen van organoïden met immuuncellen bieden een veelzijdig hulpmiddel voor het onderzoeken van de bidirectionele interacties die de delicate balans van mucosale homeostase ondersteunen. Deze 3D, multicellulaire systemen bieden een reductionistisch model voor het aanpakken van multifactoriële ziekten en het oplossen van technische problemen die zich voordoen bij het bestuderen van zeldzame celtypen zoals weefselbewonende aangeboren lymfoïde cellen (LIC’s). Dit artikel beschrijft een muizensysteem dat dunne darmorganoïden en dunne darm lamina propria afgeleide helper-achtige type-1 ILCs (ILC1’s) combineert, die gemakkelijk kunnen worden uitgebreid naar andere ILC- of immuunpopulaties. ILCs zijn een weefsel-residente populatie die bijzonder verrijkt is in het slijmvlies, waar ze homeostase bevorderen en snel reageren op schade of infectie. Organoïde coculturen met ILCs zijn al begonnen met het werpen van licht op nieuwe epitheliaal-immuun signaleringsmodules in de darm, wat onthult hoe verschillende ILC-subsets de integriteit en regeneratie van intestinale epitheliale barrières beïnvloeden. Dit protocol zal verder onderzoek mogelijk maken naar wederzijdse interacties tussen epitheel- en immuuncellen, die het potentieel hebben om nieuwe inzichten te verschaffen in de mechanismen van mucosale homeostase en ontsteking.
Communicatie tussen het darmepitheel en het darm-residente immuunsysteem staat centraal in het onderhoud van intestinale homeostase1. Verstoringen van deze interacties worden geassocieerd met zowel lokale als systemische ziekten, waaronder inflammatoire darmziekte (IBD) en gastro-intestinale kankers2. Een opmerkelijk voorbeeld van een meer recent beschreven kritische regulator van homeostase komt uit de studie van aangeboren lymfoïde cellen (LIC’s), die naar voren zijn gekomen als belangrijke spelers in het intestinale immuunlandschap3. ILCs zijn een groep heterogene aangeboren immuuncellen die intestinale homeostase reguleren en ontstekingen grotendeels orkestreren via cytokine-gemedieerde signalering4.
Murine ILCs zijn grofweg onderverdeeld in subtypen op basis van transcriptiefactor, receptor en cytokine expressieprofielen5. Type-1 ILCs, waaronder cytotoxische Natural Killer (NK) cellen en helper-achtige type-1 ILCs (ILC1s), worden gedefinieerd door expressie van de transcriptiefactor (eomesodermin) Eomen en T-box eiwit uitgedrukt in T-cellen (T-bet)6, respectievelijk, en scheiden cytokines geassocieerd met T helper type-1 (TH1) immuniteit: interferon-γ (IFNγ) en tumor necrose factor (TNF), in reactie op interleukine (IL)-12, IL-15 en IL-187. Tijdens homeostase scheiden weefselbewonende ILC1’s Transforming Growth Factor β (TGF-β) uit om epitheliale proliferatie en matrixremodellering8 te stimuleren. Type-2 ILCs (ILC2’s) reageren voornamelijk op worminfectie via secretie van T-helper type-2 (TH2) geassocieerde cytokines: IL-4, IL-5 en IL-13, en worden gekenmerkt door de expressie van retinoïnezuur-gerelateerde weesreceptor (ROR) α (ROR-α)9 en GATA Binding Protein 3 (GATA-3)10,11,12 . Bij muizen worden intestinale “inflammatoire” ILC2’s verder gekenmerkt door expressie van Killer cell lectine-achtige receptor (subfamilie G-lid 1, KLRG)13 waar ze reageren op epitheliale plukcel afgeleide IL-2514,15. Ten slotte zijn type-3 ILCs, waaronder lymfoïde weefselinductorcellen en helperachtige type-3 ILCs (ILC3s), afhankelijk van de transcriptiefactor ROR-γt16 en clusteren ze zich in groepen die Granulocyte Macrophage Colony Stimulating Factor (GM-CSF), IL-17 of IL-22 afscheiden als reactie op lokale IL-1β- en IL-23-signalen17. Lymfoïde weefselinductorcellen clusteren in Peyer’s patches en zijn cruciaal voor de ontwikkeling van deze secundaire lymfoïde organen tijdens de ontwikkeling18, terwijl ILC3’s het meest voorkomende ILC-subtype zijn in de volwassen muriene dunne darm lamina propria. Een van de vroegste muriene intestinale organoïde co-kweeksystemen met ILC3’s werd gebruikt om de impact van het cytokine IL-22 op signaaltransducer en activator van transcriptie 3 (STAT-3) gemedieerde Leucine-Rich Repeat Containing G Protein Coupled Receptor 5 (Lgr5) + intestinale stamcelproliferatie19 uit elkaar te halen, een krachtig voorbeeld van een regeneratieve ILC-epitheliale interactie. ILCs vertonen imprint-heterogeniteit tussen organen20,21 en vertonen plasticiteit tussen subsets als reactie op polariserende cytokines22. Wat deze weefselspecifieke indrukken en plasticiteitsverschillen drijft, en welke rol ze spelen bij chronische ziekten zoals IBD23, blijven spannende onderwerpen die kunnen worden aangepakt met behulp van organoïde co-culturen.
Intestinale organoïden zijn naar voren gekomen als een succesvol en betrouwbaar model om het darmepitheelte bestuderen 24,25. Deze worden gegenereerd door het kweken van intestinale epitheliale Lgr5 + stamcellen, of hele geïsoleerde crypten, waaronder Paneth-cellen als een endogene bron van Wnt Family Member 3A (Wnt3a). Deze 3D-structuren worden onderhouden in synthetische hydrogels26 of in biomaterialen die de basale lamina propria nabootsen, bijvoorbeeld Thermal-crosslinking Basal Extracellular Matrix (TBEM), en worden verder aangevuld met groeifactoren die de omliggende niche nabootsen, met name Epithelial Growth Factor (EGF), de Bone Morphogenetic Protein (BMP)-inhibitor Noggin en een Lgr5-ligand en Wnt-agonist R-Spondin127 . Onder deze omstandigheden handhaven organoïden epitheliale apico-basale polariteit en recapituleren ze de crypt-villi-structuur van het darmepitheel met ontluikende stamcelcrypten die terminaal differentiëren in absorberende en secretoire cellen in het midden van de organoïde, die vervolgens door anoikis 28 in het interne pseudolumenworden afgeworpen. Hoewel intestinale organoïden alleen al enorm voordelig zijn geweest als reductionistische modellen van epitheliale ontwikkeling en dynamiek in isolatie29,30, hebben ze een enorm toekomstig potentieel om te begrijpen hoe dit gedrag wordt gereguleerd, beïnvloed of zelfs verstoord door het immuuncompartiment.
In het volgende protocol wordt een methode van co-kweek tussen muriene dunne darm organoïden en lamina propria afgeleide ILC1’s beschreven, die onlangs werd gebruikt om te identificeren hoe deze populatie onverwacht intestinale signaturen van ontsteking vermindert en in plaats daarvan bijdraagt aan verhoogde epitheliale proliferatie via TGF-β in dit systeem8.
Dit protocol beschrijft de methoden voor het vaststellen van muriene dunne darm organoïden, het isoleren van zeldzame ILC1 door het minimaliseren van het verlies van lymfocyten tijdens het intestinale dissociatieprotocol en het vaststellen van co-culturen tussen deze twee compartimenten. Er zijn veel stappen in dit protocol, en hoewel sommige specifiek zijn voor ILC1’s, kan deze aanpak worden toegepast op andere intestinale immuunceltypen en kunnen co-kweekopstellingen modulair worden aangepast aan individuele onderzoek…
