がん研究における創薬と開発は、3D形式の薬剤スクリーンに基づいてますます進んでいます。がん細胞の移動性および侵襲性を標的とする新規阻害剤、ひるま症などの高侵襲性癌における補完的な治療法として発見され、考えられている。したがって、薬剤の添加後の3D環境における細胞の詳細な分析を可能にするデータの生成が必要となる。ここで説明する方法論は、共焦点レーザー走査顕微鏡(CLSM)による高解像度画像キャプチャーとデータ解析とスフェロイド侵入アッセイを組み合わせることで、潜在的な抗渡走阻害剤の効果の詳細な特徴付けを可能にした。グリオーマ細胞上のMI-192.スフェロイドは、低付着性96ウェルプレートの侵入アッセイ用の細胞株から生成され、CLSM分析のために調製された。記載されたワークフローは、容易さと再現性の両方に起因する他の一般的に使用されるスフェロイド生成技術よりも好まれていました。これは、従来の広視野アプローチと比較して共焦点顕微鏡によって達成される高められた画像分解能と組み合わせることで、次の3D環境における移動細胞における明確な形態変化の同定と分析を可能にした。移動性薬物MI-192による治療。
