Deze studie beschrijft een methode om exosoom-verrijkte extracellulaire blaasjes te isoleren die immuunstimulerende granulocyten macrofaagkolonie-stimulerende factoren dragen uit embryonale stamcellen.
Embryonale stamcellen (SER’s) zijn pluripotente stamcellen die in staat zijn tot zelfvernieuwing en differentiatie in alle soorten embryonale cellen. Net als veel andere celtypen geven SER’s kleine membraanblaasjes, zoals exosomen, af aan de extracellulaire omgeving. Exosomen dienen als essentiële bemiddelaars van intercellulaire communicatie en spelen een basisrol in veel (patho)fysiologische processen. Granulocyte-macrofaag koloniestimulerende factor (GM-CSF) functioneert als een cytokine om de immuunrespons te moduleren. De aanwezigheid van GM-CSF in exosomen heeft het potentieel om hun immuunregulerende functie te stimuleren. Hier werd GM-CSF stabiel overexpressie in de muriene ESC-cellijn ES-D3. Er werd een protocol ontwikkeld om hoogwaardige exosoom-verrijkte extracellulaire blaasjes (EV’s) te isoleren uit ES-D3-cellen die GM-CSF overexpressie geven. Geïsoleerde exosoom-verrijkte EV’s werden gekenmerkt door een verscheidenheid aan experimentele benaderingen. Belangrijk is dat aanzienlijke hoeveelheden GM-CSF aanwezig bleken te zijn in exosoom-verrijkte EV’s. Over het algemeen kunnen GM-CSF-dragende exosoom-verrijkte EV’s van SER’s functioneren als celvrije blaasjes om hun immuunregulerende activiteiten uit te oefenen.
SER’s zijn afgeleid van het blastocyststadium van een pre-implantatie-embryo1. Als pluripotente stamcellen hebben SER’s het vermogen om zichzelf te vernieuwen en te differentiëren in elk type embryonale cel. Vanwege hun opmerkelijke ontwikkelingspotentieel en proliferatieve capaciteit op lange termijn zijn SER’s uiterst waardevol voor biomedisch onderzoek1. De huidige onderzoeksinspanningen hebben zich grotendeels gericht op het therapeutisch potentieel van SER’s voor een verscheidenheid aan belangrijke pathologische aandoeningen, waaronder diabetes, hartaandoeningen en neurodegeneratieve ziekten2,3,4.
Van zoogdiercellen, waaronder SER’s, is bekend dat ze blaasjes met variabele grootte afgeven aan de extracellulaire omgeving, en deze EV’s bezitten veel fysiologische en pathologische functies vanwege hun rol in intercellulaire communicatie5. Onder verschillende subtypen van EV’s zijn exosomen kleine membraanblaasjes die vrijkomen uit verschillende celtypen in de extracellulaire ruimte bij fusie van intermediaire endocytische compartimenten, multivesiculaire lichamen (MWB’s), met het plasmamembraan6. Van exosomen is gemeld dat ze intercellulaire communicatie bemiddelen en zijn kritisch betrokken bij veel (patho)fysiologische processen7,8. Exosomen erven sommige biologische functies van hun eigen ouderlijke cellen, omdat exosomen biologische materialen bevatten die uit het cytosol zijn verkregen, waaronder eiwitten en nucleïnezuren. De geassocieerde antigenen of factoren die de immuunrespons specifiek voor een bepaalde ziekte stimuleren, zijn dus ingekapseld in de exosomen van bepaalde soorten cellen9. Dit maakte de weg vrij voor klinische onderzoeken naar tumor-afgeleide exosomen als een anti-kankervaccin10.
GM-CSF is een cytokine uitgescheiden door verschillende soorten immuuncellen11. Opkomend bewijs toont aan dat GM-CSF het immuunsysteem activeert en reguleert en een essentiële rol speelt in het antigeen-presenterende proces12. Een klinisch rapport suggereert bijvoorbeeld dat GM-CSF de immuunrespons op tumoren stimuleert als een vaccinadjuvans13. Verschillende gm-csf-gebaseerde kanker immunotherapie strategieën om de krachtige immuunstimulerende activiteit van GM-CSF te benutten zijn onderzocht in klinische onderzoeken14. Onder deze, een kankervaccin samengesteld uit bestraalde GM-CSF-afscheidende tumorcellen heeft enige belofte getoond bij gevorderde melanoompatiënten door cellulaire en humorale antitumorreacties en daaropvolgende necrose in uitgezaaide tumoren te induceren15.
Omdat de exosomen afgeleid van SER’s vergelijkbare biologische activiteiten hebben als de oorspronkelijke SER’s, kunnen GM-CSF-dragende exosomen van SER’s misschien functioneren als celvrije blaasjes om de immuunrespons te reguleren. In dit artikel wordt een gedetailleerde methode beschreven om hoogwaardige exosoom-verrijkte EV’s te produceren van SER’s die GM-CSF tot expressie geven. Deze exosoom-verrijkte EV’s hebben het potentieel om te dienen als immuunregulerende blaasjes om de immuunrespons te moduleren.
Deze studie toont een zeer efficiënte methode voor het produceren van exosoom-verrijkte EV’s met het immuunstimulerende eiwit GM-CSF, dat kan worden gebruikt om de immuunmodulerende effecten van exosoom-verrijkte EV’s te bestuderen. Verschillende studies suggereren dat exosomen immuunregulerende en antitumorale functies vertonen22.
