We demonstreren eencellige cultuur van bacteriën in gigantische blaasjes (GVS). Gv’s met bacteriële cellen werden bereid door de druppel overdrachtsmethode en werden geïmmobiliseerd op een ondersteund membraan op een glas substraat voor directe observatie van bacteriële groei. Deze aanpak kan ook worden aangepast aan andere cellen.
We ontwikkelden een methode voor het kweken van bacteriële cellen op het eencellige niveau binnen gigantische blaasjes (GVS). Bacteriële celcultuur is belangrijk voor het begrijpen van de functie van bacteriële cellen in de natuurlijke omgeving. Vanwege de technologische vooruitgang kunnen verschillende bacteriële celfuncties op het eencellige niveau in een besloten ruimte worden onthuld. Gv’s zijn sferische micro-sized compartimenten samengesteld uit amfifiele lipide moleculen en kan verschillende materialen, met inbegrip van cellen houden. In deze studie werd een enkele bacteriële cel ingekapseld in 10 – 30 μm GVs door de druppel overdrachtmethode en werden de Gv’s die bacteriële cellen bevatten geïmmobiliseerd op een ondersteund membraan op een glas substraat. Onze methode is nuttig voor het observeren van de real-time groei van enkele bacteriën binnen GVs. We gekweekte Escherichia coli (E. coli) cellen als een model binnen GVS, maar deze methode kan worden aangepast aan andere celtypen. Onze methode kan worden gebruikt in de wetenschap en industriële gebieden van microbiologie, biologie, biotechnologie, en synthetische biologie.
De cultuur van bacteriële cellen op het eencellige niveau heeft steeds meer aandacht gekregen. Het kweken van bacteriële cellen op het eencellige niveau in een besloten ruimte kan een verheldende werking van bacteriële functies zoals fenotypische variabiliteit1,2,3,4, Celgedrag5, 6 , 7 , 8 , 9, en antibioticaresistentie10,11. Door de recente ontwikkelingen in cultuur technieken kan de cultuur van enkele bacteriën worden bereikt in een besloten ruimte, zoals in een goed-chip4,7,8, gel druppel12,13 en water-in-olie (W/O) druppel5,11. Om het begrip of gebruik van enkele bacteriële cellen te bevorderen, zijn verdere technische ontwikkelingen van teelttechnieken nodig.
Blaasjes die de biologische celmembraan nabootsen zijn sferische compartimenten bestaande uit amfifiele moleculen en kunnen verschillende materialen vasthouden. Blaasjes zijn geclassificeerd op grootte en omvatten kleine blaasjes (SVs, diameter < 100 nm), grote blaasjes (LVs, 1 μm). SVs of Lv’s worden vaak gebruikt als medicijn dragers vanwege hun affiniteit met het biologische celmembraan14. Gv’s zijn ook gebruikt als een reactor systeem voor de bouw van protocellen15 of kunstmatige cellen16. Inkaping van biologische cellen in GVS is gemeld17,18, en dus GVS tonen potentieel als een celkweek systeem in combinatie met het reactor systeem.
Hier beschrijven we samen met een video van experimentele procedures hoe Gv’s kunnen worden gebruikt als nieuwe celkweek schepen19. Gv’s met bacteriën werden gemaakt door de druppel overdrachtsmethode20 en werden vervolgens geïmmobiliseerd op een ondersteund membraan op een afdekglas. We gebruikten dit systeem om bacteriële groei op het eencellige niveau binnen GVs in real-time te observeren.
