This protocol describes the quick enrichment of leukocytes from small blood samples for a subsequent specific determination of the halogenating peroxidase activity within the cells. The method can be applied to human and non-human material and may contribute to the evaluation of new inflammatory markers.
In dit artikel een protocol voor de snelle en gestandaardiseerde verrijking van leukocyten uit kleine hele bloedmonsters wordt beschreven. Deze procedure is gebaseerd op de hypotonische lysis van erytrocyten en kan worden toegepast op menselijke monsters en bloed van niet-menselijke oorsprong. De aanvankelijke kleine monstervolume van ongeveer 50 tot 100 gl maakt deze methode van toepassing op periodieke bloedmonsters van kleine proefdieren. Bovendien is leukocyten verrijking bereikt binnen enkele minuten en met een laag materiaal inspanningen ten aanzien van chemische stoffen en instrumentatie, het maken van deze methode niet van toepassing in meerdere laboratorium omgevingen.
Gestandaardiseerde zuivering van leukocyten wordt gecombineerd met een zeer selectieve kleuring methode halogeneringsmiddel peroxidase activiteit van het heem peroxidasen evalueren, myeloperoxidase (MPO) en eosinofiel peroxidase (EPO), dat wil zeggen de vorming van hypochloorzuur en onderbromig zuur (HOCl en HOBr). Terwijl MPO is sterk uitgedrukt in neutrophils, het meest voorkomende celtype immuunsysteem in menselijk bloed als in monocyten, de bijbehorende enzym EPO uitsluitend tot expressie gebracht in eosinofielen. Halogeneringsmiddel activiteit van deze enzymen wordt behandeld met het bijna HOCl- en HOBr specifieke kleurstof aminofenyl fluoresceïne (APF) en de primaire peroxidase substraat waterstofperoxide. Bij daaropvolgende flowcytometrieanalyse alle peroxidase-positieve cellen (neutrofielen, monocyten, eosinofielen) zijn onderscheiden en hun halogenerend peroxidase activiteit kan worden gekwantificeerd. Aangezien APF kleuring kan worden gecombineerd met de toepassing van celoppervlak markers, kan dit protocol worden uitgebreid om meer bepaald leukocyt subfracties. De methode is toepasbaar op te sporen HOCl en HOBr productie zowel in de mens en in knaagdieren leukocyten.
Gezien het wijd en divers besproken immunologische rol van deze enzymatische produkten chronische ontstekingsziekten kan dit protocol bijdragen aan een beter begrip van deimmunologische relevantie van leukocyt afgeleide heem peroxidasen.
Polymorfonucleaire leukocyten (PMN's, ook wel granulocyten) en monocyten vormen belangrijke cellulaire componenten van het aangeboren immuunsysteem in het bloed 1,2. Zij bijdragen aan de primaire afweer tegen ziekteverwekkers alsook de activering van het verworven immuunsysteem en het initiëren van een systemische ontstekingsreactie 2-4. Maar vooral neutrofielen, de meest voorkomende vorm van granulocyten en monocyten ook aanzienlijk bijdragen tot de regulering en de beëindiging van acute ontstekingen gebeurtenissen 5. Daarom zijn deze cellen kunnen ook een belangrijke rol bij chronische ontstekingsziekten zoals reumatoïde artritis 6,7 spelen. In feite, astma, een pathologie wordt gekenmerkt door een verminderde apoptose van eosinofielen, de tweede soort granulocyten in het bloed 8. Toch is de apoptose van granulocyten en hun snelle verwijdering door macrofagen zijn twee essentiële stappen tijdens de cellulaire beëindiging9-11 van ontsteking.
In de genoemde immuuncellen twee nauw verwante enzymen, namelijk myeloperoxidase (MPO, neutrofielen en monocyten) en eosinofiel peroxidase (EPO, eosinofielen) kan worden gevonden 12,13. Deze heem peroxidasen zijn klassiek gerelateerd aan de humorale immuunrespons mocht twee elektronisch oxideren (pseudo-) halogeniden in de overeenkomstige hypo (pseudo) halous zuren die bekend staan om hun bactericide 14-16. Onder fysiologische omstandigheden MPO voornamelijk vormen hypochloorzuur (HOCl) en hypothiocyaniet (- OSCN) terwijl de laatste en onderbromig acid (HOBr) gevormd door EPO 17-19. Nieuwe resultaten suggereren dat de (pseudo-) halogeneringsmiddel enzymactiviteit kan bijdragen aan de regulering van ontstekingsreacties en de beëindiging van immuunreacties 20,21. In feite is de productie van HOCl MPO en afgeleide producten werden naar T-cellen gebaseerde adaptieve immuunreacties onderdrukken 22-24.
Om meer inzicht in de immunologische rol leukocyten profiteren van het aangeboren immuunsysteem bij chronische ontstekingsziekten en het bepalen van de bijdrage van MPO en EPO deze fysiologische functie hebben we een methode om snel te verrijken leukocyten uit kleine bloedmonsters voor een volgende specifieke bepaling van het halogeneringsmiddel peroxidase activiteit in deze cellen. Voor erytrocyt depletie kozen we een gestandaardiseerde werkwijze omvattende twee opeenvolgende hypotone lysis stappen met gedestilleerd water, wat leidt tot een snelle leukocyten verrijking tegen lage materiaalkosten. Voor de volgende bepaling van de halogenerend MPO en EPO activiteit van de HOCl- en HOBr specifieke kleurstof aminofenyl fluoresceïne (APF) werd gebruikt 25-27. In tegenstelling tot de toepassing van niet-specifieke peroxidase kleuringen 28,29, deze benadering de selectieve detectie van het halogeneringsmiddel peroxidase activiteit, die vaak ernstig is aangetast inflammation 30,31.
Als neutrofielen zijn de meest voorkomende leukocyten in menselijk bloed de isolatie van peroxidase-positieve cellen vaak alleen gericht op deze cellen en omvat een scheiding van neutrofielen uit andere leukocyten door dichtheidsgradiënt centrifugatie 38. Nog neutrofielen veel minder overvloedig in muizen 39 bloedmonsters voor deze ingewikkelder methoden worden gebruikt 40.
