We present a rapid and flexible protocol for a single T cell receptor (TCR) retroviral-based in vivo expression system. Retroviral vectors are used to transduce bone marrow progenitor cells to study T cell development and function of a single TCR in vivo as an alternative to TCR transgenic mice.
T cell receptor (TCR) signaling is essential in the development and differentiation of T cells in the thymus and periphery, respectively. The vast array of TCRs proves studying a specific antigenic response difficult. Therefore, TCR transgenic mice were made to study positive and negative selection in the thymus as well as peripheral T cell activation, proliferation and tolerance. However, relatively few TCR transgenic mice have been generated specific to any given antigen. Thus, studies involving TCRs of varying affinities for the same antigenic peptide have been lacking. The generation of a new TCR transgenic line can take six or more months. Additionally, any specific backcrosses can take an additional six months. In order to allow faster generation and screening of multiple TCRs, a protocol for retroviral transduction of bone marrow was established with stoichiometric expression of the TCRα and TCRβ chains and the generation of retrogenic mice. Each retrogenic mouse is essentially a founder, virtually negating a founder effect, while the length of time to generate a TCR retrogenic is cut from six months to approximately six weeks. Here we present a rapid and flexible alternative to TCR transgenic mice that can be expressed on any chosen background with any particular TCR.
T-cel receptor (TCR) repertoire van mensen en muizen wordt geschat op 1 x 10 8 en 2 x 10 6 unieke TCR's respectievelijk 1,2. Deze veelzijdigheid maakt T-cellen om een breed scala aan antigen epitopen afgeleid van zelf-peptiden alsook van pathogenen door de major histocompatibility complex (MHC) op antigeenpresenterende cellen (APC's) herkennen. De subtiele verschillen in de interactie van het TCR's met unieke peptide-MHC complexen bepalen of een T-cel apoptose, anergie, activatie, differentiatie, cytokine productie of cytotoxiciteit zal ondergaan. Vanwege de grote TCR repertoire, analyse hoe een specifieke TCR zal reageren op een bepaald antigeen vereist het gebruik van één TCR systemen.
Verschillende TCR transgene muizen werden gegenereerd om de functie van een TCR bestuderen in een in vivo model 3-9. Er zijn echter restricties transgene muizen oa TCRkosten, de tijdsduur om een transgene muis en de zogenaamde stichterseffect willekeurige transgene insertie in kiemlijn DNA 10 genereren. Daarom hebben relatief weinig TCR transgene muizen gegenereerd voor een bepaald antigeen en de functionele implicaties van hoge en lage affiniteit TCR voor dezelfde epitoop zelden aangepakt. De noodzaak van een snelle aanpak scherm pakken en meerdere TCR bestuderen of een mengsel daarvan, hebben retrogenic ( "retro" uit retrovirus en "gene" uit transgene) muizen werden gebruikt als alternatief voor TcR transgene muizen 11-13.
2A peptide consensus motief binnen enkele virussen bestaan uit een 2A-Asp-Val / Ile-Glut-X-Asn-Pro-Gly-2B-Pro, waarbij klieving optreedt tussen de glycine van de 2A en de proline van de 2B van cis-werkende hydrolase activiteit, wat resulteert in ribosomaal overslaan tijdens de vertaling 10,14-16. Voor een gedetailleerd diagram dat de cleavage van de verschillende peptiden 2A (F2A, E2A, T2A en P2A) zie referenties 10 – 12. Op deze wijze 2 cistrons (TCR TCR alfa en bèta) kunnen worden gekoppeld waardoor stoichiometrische kopie in een enkele vector. Met behulp van deze aanpak zijn we in staat om uit te drukken en meerdere antigeenspecifieke TCRs direct vergelijken in vivo.
In het protocol, we detail een aantal kritische stappen om te zorgen voor een optimale beenmerg gezondheid, transductie efficiëntie en reconstructie. Eerste belangrijke stap is de productie en het juiste onderhoud van de GP + E86 virus producerende cellen. Gebruik vroege passage producerende cellijnen en handhaven op 80% confluentie of lager voor gebruik. Bij het maken van verse GP + E86 virale productiecellen, zorgen voor de 293T cellen zijn vroege passage en groeien in cultuur voor 24 – 48 uur. Plating teveel GP + E8…
