This report provides a detailed description of transplanting murine thymi from different aged donor mice under the kidney capsule of immunodeficient mouse recipients. The goal of this approach is to model T cell development and thymic selection events in vivo.
De mechanismen die de werkzaamheid van de thymus selectie reguleren blijft slecht gedefinieerd. De hier gepresenteerde methode maakt in vivo analyse van de ontwikkeling en selectie van T-cellen specifiek voor zichzelf en vreemde antigenen. De aanpak omvat implantatie van thymus transplantaten afkomstig van verschillende leeftijd van muizen in immunodeficiënte scid ontvangers. Over een relatief korte tijdsperiode de ontvangers volledig gereconstitueerd met T-cellen verkregen uit het geïmplanteerde transplantaat thymus. Slechts thymocyten enten de zwezerik bij de isolatie onderworpen selectie en ontwikkelen tot rijpe T-cellen. Als zodanig kunnen veranderingen in de aard en specificiteit van het getransplanteerde T-cellen als functie van de leeftijd afhankelijke thymus events worden beoordeeld. Hoewel technische expertise vereist is voor succesvolle transplantatie thymus, deze werkwijze verschaft een unieke strategie in vivo te bestuderen uiteenlopende aandoeningen die als gevolg van of berusten op afwijkende thymus functie en / of zijn homeostasis.
De thymus is een orgaan waarin kritieke gebeurtenissen in T cel ontwikkeling plaatsvinden 1. Resident thymocyten na herschikking van de T-celreceptor (TCR) α en β genen ondergaat aantal interacties met lymphostromal en antigen presenterende cellen (APC) in corticale en medullaire gebieden van de thymus 2. Thymus positieve selectie wordt gemedieerd door ritmische thymus epitheelcellen (TEC) om thymocyten die antigene peptiden herkennen in de context van ontvangst major histocompatibility complex (MHC) klasse I en II moleculen 2-3 opleveren. Daaropvolgende thymus negatieve selectie leidt zuivering van autoreactieve thymocyten, aangedreven door een interactie met medullaire TEC of dendritische cellen (DC) aanwezige peptiden afgeleid van zelf-eiwitten die door MHC klasse I en II moleculen 3 gebonden. Het resultaat van deze processen is het opzetten van een pool van volwassen CD4 + en CD8 + T-cellen kunnen reageren op een breed spectrumvan vreemde antigenen, terwijl vertonen een minimale reactiviteit om zichzelf eiwitten 4.
De efficiëntie van thymus selectie gebeurtenissen wordt beïnvloed door een groot aantal factoren, waaronder thymus rijping, frequentie medullaire en corticale TEC subset samenstelling thymus DC, en de bron van thymus precursors 3. Met name kunnen afwijkende thymus selectie resulteren in autoimmune 5 of immunodeficiënte pathologieën die voortkomen uit verminderde negatieve of positieve selectie, resp. De moleculaire gebeurtenissen reguleren thymus selectie worden echter slecht begrepen. In vitro benaderingen zoals reaggregate thymus orgaankweken (RTOC) 6, bleken nuttig voor het analyseren basic afspraken verbonden thymus keuze zijn, maar niet om de dynamiek van lopende volledig recapituleren in vivo evenementen. Hierdoor was dit thymus transplantatie benadering vastgesteld beter onderzoek thymocyte selectie gebeurtenissen in vivo 7 </sup>.
Dit protocol beschrijft het transplanteren thymi vanaf de geboorte en volwassen donor muizen in immunodeficiënte scid ontvangende muizen. Deze techniek maakt de studie van mechanismen die positieve en negatieve selectie thymus, en thymus productie van verschillende T cel subsets gedurende ontogenie regelen. Onlangs heeft deze benadering toegepast om aan te tonen dat de efficiëntie van thymus selectie is beperkt vroeg na geboorte in muizen leidt tot toegenomen ontwikkeling van autoreactieve T-cellen en een verminderde T-cel repertoire specifiek voor vreemde antigenen 7.
Events regulating thymic selection are poorly understood. Recent studies have demonstrated ontogenic changes in the efficiency of selection. Coupling this approach with different transgenic thymus donor and recipient mice will further facilitate studies to identify the events and parameters that regulate thymic selection.
