Clathrin-mediated endocytosis, a rapid and highly dynamic process internalizes many proteins, including signaling receptors. The protocol described here directly visualizes the kinetics of individual endocytic events. This is essential for understanding how core members of the endocytic machinery coordinate with each other, and how protein cargo influence this process.
Many important signaling receptors are internalized through the well-studied process of clathrin-mediated endocytosis (CME). Traditional cell biological assays, measuring global changes in endocytosis, have identified over 30 known components participating in CME, and biochemical studies have generated an interaction map of many of these components. It is becoming increasingly clear, however, that CME is a highly dynamic process whose regulation is complex and delicate. In this manuscript, we describe the use of Total Internal Reflection Fluorescence (TIRF) microscopy to directly visualize the dynamics of components of the clathrin-mediated endocytic machinery, in real time in living cells, at the level of individual events that mediate this process. This approach is essential to elucidate the subtle changes that can alter endocytosis without globally blocking it, as is seen with physiological regulation. We will focus on using this technique to analyze an area of emerging interest, the role of cargo composition in modulating the dynamics of distinct clathrin-coated pits (CCPs). This protocol is compatible with a variety of widely available fluorescence probes, and may be applied to visualizing the dynamics of many cargo molecules that are internalized from the cell surface.
O processo de endocitose mediada por clatrina (CME) é dependente da chegada oportuna dos diversos componentes da maquinaria de endocitose mediada por clatrina para recolher a carga e manipular a membrana plasmática para as vesículas 1-3. CME é iniciada por proteínas deformantes membrana e do adaptador de carga que se juntam em locais de nascentes de endocitose 1. Estas proteínas recrutar o clathrin capa protéica, que reúne em uma estrutura de gaiola que forma o poço revestido de clatrina (CCP) 4. Uma vez que o PCC está totalmente montados em uma forma esférica, cisão membrana, principalmente através da ação do grande GTPase, dynamin, gera vesículas revestidas por clatrina livres (ccvs) 5,6. A descoberta e caracterização das proteínas envolvidas no CME foi enraizada na Bioch tradicionalemical, genéticos e técnicas de microscopia 4-6,8. Estes ensaios têm elucidado as funções e pontos de interação destes componentes endocíticas. Embora muito útil para a definição de componentes essenciais de máquinas tráfico, estes ensaios são muito limitados em capturar o comportamento dinâmico de componentes CME ou concentração de carga. Esta é uma limitação fundamental, uma vez CME é impulsionada pela montagem coreografada por conjuntos de módulos de proteína em etapas definidas, e uma vez que pequenas alterações na dinâmica dos eventos endocíticas individuais podem ter grandes conseqüências cumulativas sobre endocitose. Além disso, dados recentes indicam que as CPCs individuais podem diferir tanto na composição como no comportamento, o que sugere que a regulação fisiológica do presente processo é altamente espacialmente e temporalmente restrita 9-14. Aqui descreve-se a utilização de reflexão interna total microscopia de fluorescência (TIRFM) para observar o CME ao nível da dinâmica de CPCs individuais em células vivas. TIRFM baseia-se na diferença de índice de refracção entre a lamela de vidro e para o ambiente fluido de células 15,16. Quando a luz de excitação é dirigida para as células em mais do que o ângulo crítico, é reflectida internamente, gerando uma onda evanescente, que mantém uma área de iluminação fina que se estende, aproximadamente, de 100 nm acima da lamela. Isso garante que apenas as moléculas fluorescentes dentro desse estreito campo está animado. Na prática, isto permite a excitação de moléculas fluorescentes em ou perto da membrana do plasma, e minimiza a fluorescência a partir das partes interiores da célula. Isto proporciona uma resolução significativamente mais elevada relação sinal-ruído e do eixo z para visualizar os eventos na membrana plasmática, compared para modos mais habitualmente utilizados tais como epifluorescência convencional ou microscopia de fluorescência confocal. Descrevemos também, a um nível de introdução e prático, a utilização de uma plataforma de análise de imagem utilizada para analisar e quantificar características morfológicas simples e dinâmica dos fenómenos endocíticos cargas individuais.
