Kolonisatie van de muriene nasopharynx met Streptococcus pneumoniae en de daaropvolgende extractie van aanhangende of gerekruteerde cellen wordt beschreven. Deze techniek omvat het spoelen van de nasopharynx en het opvangen van de vloeistof door de nares en is aanpasbaar voor verschillende uitlezingen, waaronder differentiële celkwantificering en analyse van mRNA-expressie in situ.
Nasofaryngeale kolonisatie door Streptococcus pneumoniae is een voorwaarde voor invasie in de longen of bloedbaan1. Dit organisme is in staat om het mucosale oppervlak van de nasopharynx te koloniseren, waar het kan verblijven, zich kan vermenigvuldigen en uiteindelijk gastheerverdediging kan overwinnen om andere weefsels van de gastheer binnen te dringen. Het vaststellen van een infectie in de normaal lagere luchtwegen resulteert in longontsteking. Als alternatief kunnen de bacteriën zich verspreiden in de bloedbaan die bacteriëmie veroorzaakt, wat gepaard gaat met hogesterftecijfers 2, of anders rechtstreeks leiden tot de ontwikkeling van pneumokokken meningitis. Het begrijpen van de kinetiek van en immuunresponsen op nasofaryngeale kolonisatie is een belangrijk aspect van S. pneumoniae-infectiemodellen.
Ons muismodel van intranasale kolonisatie is aangepast aan menselijke modellen3 en is gebruikt door meerdere onderzoeksgroepen in de studie van gastheer-pathogene reacties in de nasopharynx4-7. In het eerste deel van het model gebruiken we een klinisch isolaat van S. pneumoniae om een zelfbeperkende bacteriële kolonisatie vast te stellen die vergelijkbaar is met vervoergebeurtenissen bij menselijke volwassenen. De hierin beschreven procedure omvat de bereiding van een bacterieel entmateriaal, gevolgd door de vaststelling van een kolonisatiegebeurtenis door levering van het entmateriaal via een intranasale toedieningsweg. Ingezeten macrofagen zijn het overheersende celtype in de nasopharynx tijdens de steady state. Meestal zijn er weinig lymfocyten aanwezig in niet-geïnfecteerde muizen8, maar mucosale kolonisatie zal leiden tot laag- tot hoogwaardige ontsteking (afhankelijk van de virulentie van de bacteriële soort en stam) die zal resulteren in een immuunrespons en de daaropvolgende rekrutering van gastheerimmuuncellen. Deze cellen kunnen worden geïsoleerd door een lavage van de tracheale inhoud door de nares, en gecorreleerd met de dichtheid van kolonisatiebacteriën om de kinetiek van de infectie beter te begrijpen.
In deze studie presenteerden we gedetailleerde methoden voor de intranasale kolonisatie van muizen met behulp van een klinische isolaatstam van Streptococcus pneumoniae en de daaropvolgende isolatie en karakterisering van de immuuncellen die als reactie op de bacteriën worden gerekruteerd voor nasopharynx.
