Pour étudier le mutualisme entre<em> Xenorhabdus</em> Les bactéries et<em> Steinernema</emNématodes>, des méthodes ont été développées pour contrôler la présence de bactéries et nématodes emplacement à l'intérieur. L'approche expérimentale, qui peut être appliquée à d'autres systèmes, ingénierie implique bactéries pour exprimer la protéine fluorescente verte et de visualisation, en utilisant des bactéries de microscopie par fluorescence dans le nématode transparent.
Symbioses, le vivre ensemble de deux ou plusieurs organismes, sont répandues dans tous les règnes du vivant. Comme deux des organismes les plus répandus sur la terre, les nématodes et les bactéries forment un large éventail d'associations symbiotiques qui vont de bénéfique pour les pathogènes 1-3. Une telle association est la relation mutuellement bénéfique entre les bactéries et nématodes Steinernema Xenorhabdus, qui a émergé en tant que système modèle de symbiose 4. Nématodes entomopathogènes Steinernema sont, en utilisant leur symbiote bactérien pour tuer les insectes 5. Pour la transmission entre les hôtes d'insectes, les bactéries colonisent l'intestin de l'étape du nématode infectieux juvénile 6-8. Récemment, plusieurs autres espèces de nématodes a été démontré que les bactéries utilisent pour tuer les insectes 9-13, et les enquêtes ont commencé à étudier les interactions entre les nématodes et les bactéries dans ces systèmes <sup> 9.
Nous décrivons une méthode pour la visualisation d'un symbiote bactérien dans ou sur un hôte de nématodes, profitant de la transparence optique de nématodes lorsqu'ils sont vus au microscope. Les bactéries sont modifiées pour exprimer une protéine fluorescente, ce qui permet leur visualisation par microscopie de fluorescence. Il existe de nombreux plasmides qui portent des gènes codant pour des protéines qui sont fluorescents à différentes longueurs d'onde (c.-vert ou rouge), et la conjugaison des plasmides provenant d'une souche d'Escherichia coli donateurs dans un symbiote bactérien destinataire est positive pour une large gamme de bactéries. Les méthodes décrites ont été développés pour étudier l'association entre Steinernema carpocapsae et Xenorhabdus nematophila 14. Des méthodes similaires ont été utilisées pour étudier d'autres nématodes bactérie associations 9, 15-18 et l'approche est donc généralement applicable.
Le rencontréhod permet de caractériser la présence de bactéries et de localisation au sein de nématodes à des stades de développement différents, donnant un aperçu de la nature de l'association et le processus de la colonisation 14, 16, 19. L'analyse microscopique révèle la fréquence de colonisation à l'intérieur d'une population et la localisation des bactéries aux tissus de l'hôte 14, 16, 19-21. Il s'agit d'un avantage sur les autres méthodes de surveillance des bactéries au sein des populations de nématodes, tels que la sonication 22 ou meulage 23, qui peuvent fournir des niveaux moyens de la colonisation, mais ne peut pas, par exemple, distinguer les populations avec une fréquence élevée de faibles charges symbiotes des populations une basse fréquence de charges élevées symbiotes. Discriminer la fréquence et la charge de bactéries colonisant peut être particulièrement important lors du dépistage ou de caractérisation de mutants bactériens des phénotypes colonisation 21, 24. En effet, la microscopie à fluorescence a été utilisé dans criblage à haut débit de mutants bactériens pour les défauts de la colonisation 17, 18, et est moins pénible que les autres méthodes, y compris sonication 22, 25-27 et dissection des nématodes individuelle 28, 29.
Le protocole décrit ici fournit une méthode pour la détection optique de bactéries dans un hôte de nématodes (Figure 1). Cette méthode tire avantage de la transparence optique des nématodes et la capacité de bactéries étiquettes par fluorescence, permettant l'analyse in vivo des bactéries à l'intérieur de l'hôte de nématodes (Figure 3).
