Dit protocol beschrijft de methodologie om het retinale pigmentepitheel (RPE) genetisch te aborteren met behulp van een transgeen zebravismodel. Het aanpassen van het protocol om signaleringsroutemodulatie met behulp van farmacologische verbindingen op te nemen, is uitgebreid gedetailleerd. Een MATLAB-platform voor het kwantificeren van RPE-regeneratie op basis van pigmentatie werd ontwikkeld en wordt gepresenteerd en besproken.
Het retinale pigmentepitheel (RPE) bevindt zich aan de achterkant van het oog en vervult functies die essentieel zijn voor het behoud van de gezondheid en integriteit van aangrenzende retinale en vasculaire weefsels. Op dit moment heeft de beperkte herstelcapaciteit van RPE bij zoogdieren, die beperkt is tot kleine verwondingen, de vooruitgang in het begrijpen van in vivo RPE regeneratieve processen belemmerd. Hier wordt een gedetailleerde methodologie gegeven om de studie van in vivo RPE-reparatie met behulp van de zebravis te vergemakkelijken, een gewerveld model dat in staat is tot robuuste weefselregeneratie. Dit protocol beschrijft een transgeen nitroreductase/metronidazol (NTR/MTZ)-gemedieerd letselparadigma (rpe65a:nfsB-eGFP), dat resulteert in ablatie van de centrale tweederde van de RPE na 24 uur behandeling met MTZ, met daaropvolgend weefselherstel. De nadruk wordt gelegd op RPE-ablaties in larvale zebravissen en methoden voor het testen van de effecten van farmacologische verbindingen op RPE-regeneratie worden ook geschetst. Generatie en validatie van RpEGEN, een MATLAB-script dat is gemaakt om de kwantificering van RPE-regeneratie op basis van pigmentatie te automatiseren, wordt ook besproken. Naast actieve RPE-reparatiemechanismen kan dit protocol worden uitgebreid naar studies van RPE-degeneratie en letselresponsen, evenals de effecten van RPE-schade op aangrenzende retinale en vasculaire weefsels, naast andere cellulaire en moleculaire processen. Dit zebravissysteem is veelbelovend bij het identificeren van genen, netwerken en processen die RPE-regeneratie en RPE-ziektegerelateerde mechanismen aansturen, met het langetermijndoel om deze kennis toe te passen op zoogdiersystemen en, uiteindelijk, in de richting van therapeutische ontwikkeling.
De hierin beschreven methodologie beschrijft een protocol om het retinale pigmentepitheel (RPE) genetisch te aborteren met behulp van larvale zebravissen. De RPE strekt zich uit over de achterkant van het oog en bevindt zich tussen de gelaagde lagen van het neurale netvlies en de laag vasculatuur die het vaatvlies vormt. Trofische ondersteuning, absorptie van fototoxisch licht en onderhoud van visuele cycluseiwitten zijn slechts enkele van de kritieke functies die de RPE uitvoert en die essentieel zijn voor het behoud van de gezondheid en integriteit van deze aangrenzende weefsels1. Schade aan RPE bij zoogdieren is te herstellen wanneer laesies klein zijn2; schade geleden door grotere verwondingen of progressieve degeneratieve ziekte is echter onomkeerbaar. Bij mensen leiden RPE-degeneratieve ziekten (bijv. leeftijdsgebonden maculaire degeneratie (AMD) en de ziekte van Stargardt) tot permanent verlies van het gezichtsvermogen en, met weinig beschikbare behandelingsopties, verminderde kwaliteit van leven van de patiënt. Het beperkte vermogen van RPE bij zoogdieren om zichzelf te herstellen heeft een kenniskloof gecreëerd op het gebied van RPE-regeneratieve processen. Gezien het robuuste regeneratieve vermogen van de zebravis in veel verschillende weefseltypen, is dit protocol ontwikkeld om een in vivo gewerveld systeem op te zetten om studies naar intrinsiek regenererende RPE te vergemakkelijken en mechanismen bloot te leggen die die respons stimuleren. Met behulp van het hier geschetste ablatieparadigma zijn de canonieke Wnt-signaleringsroute3, de mTOR-route4 en immuungerelateerde responsen5 geïdentificeerd als kritieke mediatoren van RPE-regeneratie, waarschijnlijk met overlappende functies.
In dit genetische ablatieparadigma brengen Tg(rpe65a:nfsB-eGFP)3 zebravissen het bacteriële nitroreductase (NTR/nfsB) gen6 tot expressie, gefuseerd met eGFP onder controle van het RPE-versterkerelement, rpe65a7. Ablatie wordt bereikt door de prodrug, metronidazol (MTZ), toe te voegen aan het systeemwater dat zebravissen huisvest. Intracellulaire activering van MTZ door nitroreductase resulteert in DNA-crosslinking en apoptose in NTR/nfsB-tot expressie brengende cellen 8,9. Deze technologie is veel gebruikt in zebravissen om cellen van het netvlies10,11,12,13 en andere weefsels te aborteren8. Samen maken deze elementen gerichte expressie (rpe65a) van een induceerbare celablatiemethodologie (NTR/MTZ)8,9 en een fluorescerende marker (eGFP) voor visualisatie mogelijk.
Er bestaan ook andere interessante in vivo modellen die kunnen worden gebruikt om het regeneratieve potentieel van de RPE14 te bestuderen. Deze zijn breed en omvatten RPE-naar-retina transdifferentiatie post-retinectomie bij amfibieën, waarbij RPE-cellen die verloren gaan aan retinale hergroei worden vervangen15,16; RPE restauratie na letsel in de “super healing” MRL/MpJ muis17; en exogene stimulatie van RPE-proliferatie in een rattenmodel van spontane RPE en retinale degeneratie18, onder anderen. Er zijn ook in vitro modellen ontwikkeld, zoals volwassen menselijke RPE-stamcellen (RPESCs)19. Deze modellen zijn allemaal waardevolle hulpmiddelen die werken aan het blootleggen van de cellulaire processen die verband houden met RPE-regeneratie (bijv. Proliferatie, differentiatie, enz.); de zebravis is echter uniek in zijn vermogen tot intrinsiek RPE-herstel na ablatie.
Hoewel de methodologie hier is geschreven om zich te concentreren op het begrijpen van de mechanismen die RPE-regeneratie aansturen, kunnen de Tg (rpe65a: nfsB-eGFP) – lijn en dit genetische ablatieprotocol worden gebruikt om andere cellulaire processen te bestuderen, zoals RPE-apoptose, RPE-degeneratie en het effect van RPE-letsel op aangrenzende retinale en vasculaire weefsels. Het ablatieprotocol kan ook worden gewijzigd om farmacologische manipulatie op te nemen, wat een handige voorlopige strategie is om signaalroutes van belang te screenen. Het blokkeren van de canonieke Wnt-route met behulp van Inhibitor of Wnt Response-1 (IWR-1)20, is bijvoorbeeld aangetoond dat het de RPE-regeneratie3 schaadt. Dit werd hier herhaald om gebruikers door een farmacologisch manipulatie-experiment te leiden en als proof-of-concept te dienen om een MATLAB-script (RpEGEN) te valideren dat is gemaakt om RPE-regeneratie te kwantificeren op basis van herstel van pigmentatie. Net als de transgene lijn en het ablatieprotocol zijn de RpEGEN-scripts aanpasbaar en kunnen ze worden gebruikt om andere markers / cellulaire processen binnen de RPE te kwantificeren.
Dit protocol beschrijft methodologie om de RPE genetisch te aborteren en bestudeert mechanismen van degeneratie en regeneratie bij larvale-oude zebravissen. Dit protocol is ook met succes uitgevoerd bij volwassen zebravissen3, maar met minder uitgebreide karakterisering, daarom zijn larven hier de focus.
