Dit protocol biedt een efficiënte collagenase-verteringsmethode voor isolatie van levensvatbare adipocyten en stromale vasculaire fractie-SVF-cellen uit menselijk visceraal vet in één enkel proces, inclusief methodologie om hoogwaardig RNA uit adipocyten te verkrijgen en fenotypificatie van SVF-macrofagen door kleuring van meerdere membraangebonden markers voor analyse door middel van flowcytometrie.
Visceraal vetweefsel (VAT) is een actief metabolisch orgaan dat voornamelijk bestaat uit rijpe adipocyten en stromale vasculaire fractie (SVF)-cellen, die verschillende bioactieve moleculen afgeven die metabole, hormonale en immuunprocessen regelen; Momenteel is het onduidelijk hoe deze processen in het vetweefsel worden gereguleerd. Daarom is de ontwikkeling van methoden die de bijdrage van elke celpopulatie aan de pathofysiologie van vetweefsel evalueren, cruciaal. Dit protocol beschrijft de isolatiestappen en biedt de nodige richtlijnen voor het oplossen van problemen voor een efficiënte isolatie van levensvatbare rijpe adipocyten en SVF uit menselijke VAT-biopsieën in één enkel proces, met behulp van een collagenase enzymatische verteringstechniek. Bovendien is het protocol ook geoptimaliseerd om macrofaagsubsets te identificeren en volwassen adipocyten-RNA-isolatie uit te voeren voor genexpressiestudies, waardoor studies kunnen worden uitgevoerd om de interactie tussen deze celpopulaties te ontleden. In het kort worden btw-biopsieën gewassen, mechanisch gehakt en verteerd om een eencellige suspensie te genereren. Na centrifugatie worden rijpe adipocyten geïsoleerd door flotatie uit de SVF-pellet. Het RNA-extractieprotocol zorgt voor een hoge opbrengst van totaal RNA (inclusief miRNA’s) uit adipocyten voor stroomafwaartse expressietests. Tegelijkertijd worden SVF-cellen gebruikt om macrofaagsubsets (pro- en anti-inflammatoir fenotype) te karakteriseren door middel van flowcytometrie-analyse.
Wit vetweefsel bestaat niet alleen uit vetcellen of adipocyten, maar ook uit een niet-vette celfractie die bekend staat als stromale vasculaire fractie (SVF), die een heterogene celpopulatie bevat die bestaat uit macrofagen, andere immuuncellen als regulerende T-cellen (Tregs) en eosinofielen, preadipocyten en fibroblasten, omgeven door vasculair en bindweefsel 1,2. Vetweefsel (AT) wordt nu beschouwd als een orgaan dat fysiologische processen reguleert die verband houden met metabolisme en ontsteking door middel van adipokines, cytokines en microRNA’s die door verschillende cellen worden geproduceerd en in het weefsel worden afgegeven, met autocriene, paracriene en endocriene effecten 3,4. Bij mensen omvat wit vetweefsel het onderhuidse vetweefsel (SAT) en het viscerale vetweefsel (VAT), met belangrijke anatomische, moleculaire, cellulaire en fysiologische verschillen tussen hen 2,5. SAT vertegenwoordigt tot 80% van de menselijke AT, terwijl VAT zich in de buikholte bevindt, voornamelijk in het mesenterium en het omentum, en metabolisch actiever is6. Bovendien is VAT een endocrien orgaan dat mediatoren afscheidt met een substantiële invloed op het lichaamsgewicht, de insulinegevoeligheid, het lipidenmetabolisme en ontstekingen. Bijgevolg leidt btw-accumulatie tot abdominale obesitas en aan obesitas gerelateerde ziekten zoals diabetes type 2, metabool syndroom, hypertensie en het risico op hart- en vaatziekten, wat een betere voorspeller is van obesitas-gerelateerde mortaliteit 6,7,8,9.
In homeostatische omstandigheden werken adipocyten, macrofagen en de andere immuuncellen samen om het VAT-metabolisme in stand te houden door de afscheiding van ontstekingsremmende mediatoren10. Buitensporige btw-verhoging bevordert echter de rekrutering van geactiveerde T-cellen, NK-cellen en macrofagen. In feite is de verhouding van de macrofagen bij magere btw 5%, terwijl deze verhouding oploopt tot 50% bij obesitas, waarbij macrofaagpolarisatie van ontstekingsremmend naar pro-inflammatoir fenotype ontstaat, waardoor een chronische ontstekingsomgeving ontstaat10,11.
Als gevolg van de obesitaspandemie is er een verbazingwekkend aantal rapporten verschenen over verschillende btw-onderzoeksonderwerpen, waaronder adipocytenbiologie, epigenetica, ontsteking, endocriene eigenschappen en opkomende gebieden zoals extracellulaire blaasjes, onder andere 8,10,12,13. Hoewel de btw-omgeving wordt bepaald door overspraak tussen adipocyten en de residente of aankomende macrofagen, hebben de meeste onderzoeken zich gericht op slechts één celpopulatie en is er weinig informatie over de interactie van deze cellen in btw en hun pathofysiologische gevolgen11,14. Bovendien werden waardevolle studies naar het adipocyt-macrofaag-samenspel in AT uitgevoerd met behulp van cellijnen, zonder de in vivo priming-omstandigheden 11,14,15. Een geschikte strategie om de interactie of de specifieke bijdrage van deze cellen in VAT te ontleden, vereist de isolatie van beide celtypen uit dezelfde vetbiopsie om in vitro assays uit te voeren die de in vivo eigenschappen die het VAT-metabolisme reguleren zo veel mogelijk weerspiegelen.
Hoewel de niet-enzymatische dissociatiemethoden op basis van mechanische krachten om de AT te breken minimale manipulatie garanderen, kunnen deze methoden niet worden gebruikt als het doel is om SVF-cellen te bestuderen, omdat ze een lagere efficiëntie hebben bij celherstel en een lage cellevensvatbaarheid in vergelijking met enzymatische methoden, en er een groter weefselvolume nodig is16,17. Enzymatische vertering met behulp van collagenase is een zachte methode die een adequate vertering van collageen en extracellulaire matrixeiwitten van vezelachtige weefsels zoals WAT18 mogelijk maakt en wordt vaak gebruikt wanneer trypsine niet effectief of schadelijk is19. Het protocol biedt fundamentele richtlijnen voor het oplossen van problemen voor efficiënte isolatie van levensvatbare rijpe adipocyten en SVF-cellen uit menselijke VAT-biopsieën in een enkel proces, met behulp van een collagenase-enzymatische verteringstechniek, en geeft informatie om hoge opbrengsten (hoeveelheid, zuiverheid en integriteit) van totaal RNA uit rijpe adipocyten, inclusief microRNA’s, te garanderen voor stroomafwaartse expressietoepassingen. Tegelijkertijd is het protocol geoptimaliseerd om macrofaagsubsets van SVF-cellen te identificeren door middel van de kleuring van meerdere membraangebonden markers voor verdere analyse door middel van flowcytometrie20.
BTW speelt een cruciale rol bij metabole regulatie en ontstekingen. Toenemende belangstelling voor de rol van adipocyten en immuuncellen bij de chronische ontsteking die gepaard gaat met obesitas heeft geleid tot de ontwikkeling van verschillende technieken om de SVF en vetcellen die aanwezig zijn in AT te scheiden. De meeste technieken maken het echter niet mogelijk om deze twee verschillende sets cellen die levensvatbaar zijn voor stroomafwaartse toepassingen uit dezelfde btw-biopsie in één enkele procedure te verkri…
