We beschrijven een humaan perifeer bloed mononucleaire cel (PBMC) — op basis van gehumaniseerd xenotransplantaatmodellen is muismodel voor translationeel immuno-oncologisch onderzoek. Dit protocol kan dienen als een algemene richtlijn voor het vaststellen en karakteriseren van vergelijkbare modellen voor I-O-therapie beoordeling.
De ontdekking en ontwikkeling van immuno-oncologie (I-O) therapie in de afgelopen jaren vormt een mijlpaal in de behandeling van kanker. Echter, behandeling uitdagingen blijven bestaan. Robuuste en ziektegerelateerde diermodellen zijn essentiële hulpmiddelen voor voortgezet preklinisch onderzoek en ontwikkeling om een reeks extra immuuncontrolepunten aan te pakken. Hier beschrijven we een humaan perifeer bloed mononucleaire cel (PBMC) — gebaseerd gehumaniseerd xenotransplantaatmodellen is model. BGB-A317 (Tislelizumab), een humaan anti-PD-1 antilichaam in de late fase van de klinische ontwikkeling, wordt gebruikt als een voorbeeld om de opstelling van het platform, de model karakterisatie en evaluaties van de geneesmiddelen werkzaamheid te bespreken. Deze gehumaniseerde muizen ondersteunen de groei van de meeste menselijke tumoren getest, waardoor de beoordeling van I-O therapieën in het kader van zowel menselijke immuniteit en menselijke kankers. Na de oprichting is ons model relatief tijd-en kostenbesparend en levert het meestal zeer reproduceerbare resultaten op. Wij stellen voor dat het protocol beschreven in dit artikel kan dienen als een algemene richtlijn voor het tot stand brengen van Muismodellen die zijn gereconstitueerd met menselijke PBMC en tumoren voor I-O onderzoek.
Immuno-oncologie (I-O) is een snel groeiend gebied van kankerbehandeling. Onderzoekers zijn onlangs begonnen te waarderen het therapeutische potentieel van het moduleren van functies van het immuunsysteem om tumoren aan te vallen. Immuun Checkpoint blokkades hebben aangetoond bemoedigende activiteiten in een verscheidenheid van kankersoorten, met inbegrip van melanoom, niercelcarcinoom, hoofd en nek, Long, blaas en prostaatkanker1,2. In tegenstelling tot gerichte therapieën die kankercellen direct doden, potentiëren I-O-therapieën het afweersysteem van het lichaam om tumoren3aan te vallen.
Tot op heden zijn tal van relevante I-O-diermodellen vastgesteld. Deze omvatten: 1) muis tumor cellijnen of tumor homo graft in syngene muizen; 2) spontane tumoren afgeleid van genetisch gemanipuleerde muis (GEM) of kankerverwekkend-inductie; 3) chimerische edelstenen met de knock-in van de drug target (s) in een functionele Murine immuunsysteem; en 4) muizen met gereconstitueerde menselijke immuniteit getransplanteerd met menselijke kankercellen of patiënt-afgeleide xenotransplantaten (pdxs). Elk van deze modellen hebben duidelijke voordelen, evenals beperkingen, die zijn beschreven en uitgebreid elders beoordeeld4.
Reconstitutie van de menselijke immuniteit in immunodeficiënte muizen is gegroend gewaardeerd als een klinisch relevante aanpak voor translationeel I-O-onderzoek. Dit wordt meestal bereikt door 1) engraftment van volwassen immuuncellen (bijv. perifere bloed mononucleaire cellen (pmbc))5,6of 2) engraftment van hematopoietische stamcellen (HSC) van, bijvoorbeeld, navelstreng bloed of foetale lever7,8. Deze gehumaniseerde muizen kunnen de groei van menselijke tumoren ondersteunen, waardoor de beoordeling van I-O-therapieën mogelijk is in het kader van zowel menselijke immuniteit als menselijke kankers. Ondanks de voordelen werden toepassingen van gehumaniseerde muizen in I-O-onderzoek meestal gehinderd door verschillende zorgen, zoals lange model ontwikkelingstijd en aanzienlijk hoge kosten.
Hier beschrijven we een menselijk PBMC-gebaseerd model dat op grote schaal kan worden toegepast voor translationele I-O-studies. Dit model is relatief tijd-en kosteneffectief met een hoge reproduceerbaarheid in werkzaamheidsstudies. Het is in eigen huis gebruikt voor de evaluaties van verschillende I-O-therapieën die momenteel onder preklinisch en klinische ontwikkeling vallen. BGB-A317 (Tislelizumab), een experimenteel gehumaniseerd anti-PD-1 antilichaam9 , wordt gebruikt als voorbeeld om de modelontwikkeling, karakterisering en mogelijke toepassingen voor anti-tumor werkzaamheids analyses te bespreken.
Onze kennis van de ontwikkeling van kanker en progressie is aanzienlijk gevorderd in de afgelopen jaren, met de focus op een uitgebreid begrip van zowel de tumorcellen en de bijbehorende stroma. Het benutten van de immuunmechanismen van de gastheer kan een grotere impact op kankercellen veroorzaken, wat een veelbelovende behandelingsstrategie vertegenwoordigt. Murine modellen met intact muis immuunsysteem, zoals syngene en GEM modellen, zijn op grote schaal gebruikt voor het bestuderen van Checkpoint-gemedieerde immunite…
