Het doel van dit artikel is bedoeld als een gedetailleerde beschrijving van het protocol voor de pulmonaire metastase assay (PuMA). Dit model staat onderzoekers bestuderen metastatische Osteosarcoom (OS) celgroei in longweefsel met behulp van een widefield fluorescentie- of confocale laser scanning microscoop.
De pulmonaire metastase assay (PuMA) is een ex vivo Long explant en gesloten celcultuur waarmee onderzoekers om te bestuderen van de biologie van longkanker kolonisatie in Osteosarcoom (OS) door fluorescentie microscopie. Dit artikel geeft een gedetailleerde beschrijving van het protocol, en voorbeelden van het verkrijgen van afbeeldingsgegevens op metastatische groei met widefield of confocal fluorescentie microscopie platformen. De flexibiliteit van het model van de PuMA toelaat onderzoekers bestuderen van niet alleen de groei van OS cellen in de longen-communicatie, maar ook voor het beoordelen van de effecten van anti-metastatische therapeutics na verloop van tijd. Confocale microscopie zorgt voor ongekende, high-resolution beeldvorming van OS cel interacties met de Long parenchym. Bovendien, wanneer de PuMA model wordt gecombineerd met fluorescente kleurstoffen of TL proteïne genetische verslaggevers, onderzoekers kunnen bestuderen de Long-communicatie, cellulaire en subcellular structuren, genfunctie en promotor activiteit in uitgezaaide cellen van het OS. De PuMA-model biedt een nieuw hulpmiddel voor Osteosarcoom onderzoekers om te ontdekken van nieuwe metastase biologie en de activiteiten beoordelen van nieuwe anti-gemetastaseerd, gerichte therapieën.
Verbeterde resultaten voor pediatrische patiënten met metastatische Osteosarcoom (OS) blijft een kritische unmet klinische behoefte 1. Dit onderstreept het belang van de ontwikkeling van nieuwe moleculair-gerichte therapieën. Conventionele chemotherapeutica dat doel tumor celproliferatie hebben niet bewezen effectief bij de behandeling van uitgezaaide ziekte, en zo nieuwe strategieën moeten richten op het metastatische proces zelf 2. Het huidige artikel behandelt de praktische aspecten van een relatief nieuw soort ex vivo Long metastase model, de pulmonaire metastase assay (PuMA) ontwikkeld door Mendoza en collega’s3, waarmee een nuttig instrument in het ontdekken van nieuwe moleculaire bestuurders in Long metastase progressie in OS 4,5. Voordat u verdergaat, echter, zou het verstandig zijn kort ingaan op de verschillende huidige modellen van metastase, en hoe de PuMA model biedt diverse voordelen ten opzichte van conventionele in vitro testen.
Meest experimentele modellen gebruikt bij het bestuderen van metastase bestaan uit in vitro en in vivo systemen die een specifieke handeling of verschillende stappen van de metastatische cascade recapituleren. Deze stappen omvatten: 1) de tumorcellen migreren uit de buurt van de primaire tumor, 2) intravasation in nabijgelegen vaartuigen (bloed- of lymfestelsel) en doorvoer in omloop, 3) arresteren op het secundaire site, de 4) extravasation en de overleving op de secundaire site, 5) vorming micrometastasen, en 6) groei in gevacuoliseerd metastasen (Figuur 1). In vitro modellen van metastase 2-dimensionale (2D) migratie kunnen opnemen en 3-dimensionale (3D) Matrigel invasie testen die zijn herzien in detail elders 6. Voor in vivo modellen, de twee meest gebruikte modelsystemen omvatten: 1) de spontane metastase model is waarbij een tumorcellen orthotopically geïnjecteerd in een specifieke weefseltype te vormen van een lokale tumor die spontaan uitgezaaide cellen werpt naar verre locaties; 2) het experimentele metastase model is waar tumorcellen worden ingespoten in het bloedvat stroomopwaarts van de doel-orgel. Bijvoorbeeld, een staart veneuze injectie van tumor cellen leidt tot de ontwikkeling Long metastasen5,7,8. Andere experimentele metastase modellen zijn injectie van tumorcellen in de milt of lymfklieren ader wat in de ontwikkeling van lever metastasen9,10 resulteert. Praktische overwegingen van deze in vivo modellen worden in detail besproken door Welch 11. Een andere in vivo model gebruikt bij het bestuderen van metastase in pediatrische sarcomen is het renale nier subcapsular tumor implantatie model, wat in de vorming van lokale tumor en spontane metastase voor de longen 12,13 resulteert. Een meer technisch veeleisende techniek zoals intravital videomicroscopy kan direct visualiseren, in real-time, interacties tussen metastatische kankercellen en de microvasculature van een metastatische site (dwz. longen of lever) zoals beschreven door MacDonald14 en Entenberg15, of kanker cel extravasation in het chorion membraan zoals beschreven door Kim, 16.
De PuMA-model is een ex vivo, Long weefsel explant, gesloten cultuur systeem waar de groei van fluorescerende tumorcellen lengterichting waarneembaar via fluorescentie microscopie gedurende een periode van een maand (Zie figuur 2A). Dit model recapituleert de aanvankelijke stadia van longkanker kolonisatie (stappen 3 tot en met 5) in de gemetastaseerde cascade. Enkele belangrijke voordelen van de PuMA model ten opzichte van conventionele in vitro modellen zijn: 1) het biedt een kans om de lengterichting meten metastatische kanker-celgroei in een 3D communicatie die heeft veel kenmerken van de Long communicatie behouden vivo 3; 2) puMA kan de onderzoeker om te beoordelen of het knockdown van een kandidaat-gen of drug behandeling anti-metastatische activiteit in het kader van een 3D Long communicatie heeft; 3) de PuMA model is flexibel met vele soorten fluorescentie microscopie platforms (figuur 2B) zoals widefield fluorescentie microscopie of laser-scanning confocal microscopie, worden voorbeelden van elk weergegeven in figuur 2C & O, respectievelijk. Dit artikel zal bespreken hoe de PuMA model kunt verkrijgen van longitudinale imaging gegevens over de metastatische groei van verbeterde groen fluorescent proteïne (eGFP)-uiten, menselijke hoge en lage metastatische Osteosarcoom cellen (cellen met MNNG en HOS, respectievelijk) met behulp van lage-vergroting widefield fluorescentie. Voorbeelden van imaging een fluorescente kleurstof waarmee de Long parenchym de labels, en een rood-fluorescerende eiwit genetische verslaggever waarmee labels mitochondriën in OS cellen in het model van de PuMA met behulp van confocale laser scanning microscopie worden ook besproken.
Het volgende technische artikel beschrijft enkele praktische aspecten van het model van de PuMA in het bestuderen van de Long kolonisatie in OS. Sommige kritische stappen in het protocol waar onderzoekers extra zorg moeten omvatten het volgende:
