Dit protocol beschrijft de inductie van een ischemie-reperfusie (IR) model op de muis oor huid met behulp van magneet klemmen. Met behulp van een custom-built intravitale imaging model, bestuderen we in vivo ontstekingsreacties post-reperfusie. De reden voor de ontwikkeling van deze techniek is om te begrijpen hoe leukocyten reageren op huid IR schade verlengen.
Ischemia-reperfusion injury (IRI) occurs when there is transient hypoxia due to the obstruction of blood flow (ischemia) followed by a subsequent re-oxygenation of the tissues (reperfusion). In the skin, ischemia-reperfusion (IR) is the main contributing factor to the pathophysiology of pressure ulcers. While the cascade of events leading up to the inflammatory response has been well studied, the spatial and temporal responses of the different subsets of immune cells to an IR injury are not well understood. Existing models of IR using the clamping technique on the skin flank are highly invasive and unsuitable for studying immune responses to injury, while similar non-invasive magnet clamping studies in the skin flank are less-than-ideal for intravital imaging studies. In this protocol, we describe a robust model of non-invasive IR developed on mouse ear skin, where we aim to visualize in real-time the cellular response of immune cells after reperfusion via multiphoton intravital imaging (MP-IVM).
Ischemie-reperfusie schade (IRI) treedt op wanneer er een voorbijgaande hypoxie als gevolg van de obstructie van de bloedstroom (ischemie), gevolgd door een latere re-oxygenatie van weefsels (reperfusie). In de huid wordt ischemie-reperfusie (IR) beschouwd als een van de factoren die bijdragen aan de pathofysiologie van decubitus, waarbij langdurige bedrust predisponeert langdurige ziekenhuispatiënten om letsel. Bij deze patiënten wordt zowel de huid en de onderliggende spieren voortdurend blootgesteld aan gewicht uitgeoefende druk over gebieden van botuitsteeksel, waardoor gelokaliseerde verwondingen die, indien onbehandeld, kan necrotisch 1 worden.
De betrokken bij een IRI schade zijn tweeledig. Tijdens ischemie, de occlusie van bloedvaten leidt tot een drastische daling van zuurstofafgifte aan de weefsels. Dit resulteert in een daling van ATP en pH, die ATPasen betrokken celstofwisseling inactiveert. Op zijn beurt, de cellulaire calcium niveaus spike, en gestrest of beschadigde cells ondergaan apoptose of necrose 2. De afgifte van intracellulaire inhoud of schade geassocieerde moleculaire patronen (DAMP), zoals HMGB1 bijdraagt aan de ontstekingsreactie 3. De tweede belediging optreedt tijdens reperfusie. Hoewel zuurstof en pH-niveaus worden hersteld tijdens reperfusie resulteert in de generatie van reactieve zuurstofsoorten (ROS), die leidt tot de oxidatie van intracellulaire lipiden, DNA en eiwitten. Dientengevolge zijn pro-inflammatoire mediatoren geactiveerd, dat een secundair ontstekingsreactie die de rekrutering van immuuncellen naar de ontstekingsplaats 2 omvat vertrekt. Terwijl de cascade van biochemische gebeurtenissen die leidden tot de ontstekingsreactie is goed beschreven, worden de ruimtelijke en temporele regulatie van het immuunsysteem cel activiteiten niet goed begrepen.
Hier beschrijven we een robuuste IR-model op de muis oor huid met behulp van eenvoudige magneet klemmen. In combinatie met multifoton intravitale beeldvorming (MP-IVM), wewerd een model voor de in vivo ontstekingsreacties die na reperfusie plaatsvindt bestuderen. De grondgedachte achter de ontwikkeling en het gebruik van deze techniek is om te proberen te begrijpen hoe zowel de interstitiële en infiltrerende cellen reageren op IR in real time.
Bestaande modellen van IR behulp van de stel- techniek op de huid flank zijn sterk invasieve mocht de chirurgische implantatie van staalplaten in de huid flank vereist, waardoor ze minder dan ideaal voor immunologische studies 4. Een dergelijke niet-invasieve klemming techniek is beschreven in de muizenhuid flank 5,6. Vanwege de integratie van de intravitale beeldvorming component in deze werkwijze, maar wij kozen voor de oorhuid de beoogde IR site, zoals omzeilt bewegingen door ademhaling en stabiliteit biedt tijdens beeldvorming 7,8. Bovendien leukocyten subsets de interstitium overspannen zijn identiek tussen het oor huid en de huid flank, hoewel degetallen en verhoudingen kunnen enigszins variëren 9. Dus de oorhuid vertegenwoordigt een ideale beeldvorming plaats.
Daarnaast hebben de meeste gegevens uit deze IRI modellen zijn beperkt tot macroscopische evaluaties (grading van zweren) en microscopische analyses van endpoint inflammatoire indicatoren 10. Met behulp van dit model wordt real-time visualisatie van de cellulaire respons van neutrofielen na reperfusie in de huid van een tl-reporter muis ingeschakeld. Een eerder gepubliceerd intravitale oor imagingmodel wordt gebruikt 8 met extra modificaties (figuren 1, 2).
Betekenis
IR is een van de belangrijkste oorzaken van de huid decubitus. De vroege stadia (I en II) van decubitus beschrijft de toestand van de menselijke huid (ten opzichte van de onderliggende onderhuidse weefsels en spieren).
