This manuscript provides the detailed procedure of intra-iliac artery (IIA) injection, a technique to deliver cancer cells specifically to hind limb tissues including bones to establish experimental bone metastases. Although initially established with breast tumor models, this protocol can be easily extended to other cancer types.
Intra-iliac artery (IIA) injection is an efficient approach to introduce metastatic lesions of various cancer cells in animals. Compared to the widely used intra-cardiac and intra-tibial injections, IIA injection brings several advantages. First, it can deliver a large quantity of cancer cells specifically to hind limb bones, thereby providing spatiotemporally synchronized early-stage colonization events and allowing robust quantification and swift detection of disseminated tumor cells. Second, it injects cancer cells into the circulation without damaging the local tissues, thereby avoiding inflammatory and wound-healing processes that confound the bone colonization process. Third, IIA injection causes very little metastatic growth in non-bone organs, thereby preventing animals from succumbing to other vital metastases, and allowing continuous monitoring of indolent bone lesions. These advantages are especially useful for the inspection of progression from single cancer cells to multi-cell micrometastases, which has largely been elusive in the past. When combined with cutting-edge approaches of biological imaging and bone histology, IIA injection can be applied to various research purposes related to bone metastases.
Metastasen goed voor meer dan 90% van de sterfgevallen als gevolg van solide tumoren. Bot is de meest voorkomende orgaan beïnvloed door metastasen van verscheidene soorten kanker, in het bijzonder borstkanker en prostaatkanker. Wanneer de diagnose in de kliniek, meestal botmetastasen al hebt ingevoerd gevorderde stadia met ofwel osteolytische of osteoblastische veranderingen in bot, vaak gepaard met neurologische symptomen.
Eerdere studies voornamelijk gericht op de openlijke osteolytische botmetastasen 1-3, maar we momenteel beperkte begrip van micrometastasen in botten voor het begin van het proces osteolytische. Dit is althans gedeeltelijk te wijten aan gebrek aan geschikte experimentele modellen en benaderingen. Genetisch gemanipuleerde muis modellen van borstkanker metastaseren vaak longen, maar veel minder efficiënt botten 4. Ook de orthotopically getransplanteerde tumoren ontwikkelen zelden spontaan botmetastasen, met een aantal bot-tropische 4T1 borstklier carcinoomeen sub-klonen en MSP overexpressie PyMT transgene muismodel als uitzonderingen 5-7. Intra-scheenbeen boren kan kankercellen te leveren aan het bot 8-10, maar zij maakt ook schade en ontstekingen aan de lokale weefsels. Momenteel intracardiale injectie van borstkanker cellijnen belangrijke benadering bot kolonisatie 11-13 onderzoeken geweest. Echter, na kankercellen worden ingebracht in linker ventrikel slechts een beperkt deel zal uiteindelijk bij bot en beenmerg, waardoor het moeilijk microscopische uitzaaiingen volgen op een meetbare manier.
In deze studie hebben we vastgesteld techniek, namelijk intra-iliacale arterie (IA) injectie 14, om kankercellen selectief leveren aan achterpoot weefsels, waardoor kankercellen in bot en beenmerg verrijken zonder schade aan lokale weefsels. Door het bot specificiteit, deze benadering ook voldoende voor indolent kankercellen uiteindelijk koloniseren alvorens het eenij dieren bezwijken voor primaire tumoren en uitzaaiingen in andere vitale organen. In combinatie met verschillende andere technieken, zoals bioluminescentie, immunofluorescentie kleuring en bothistomorfometrie, IIA injectie is potentieel nuttig voor een breed scala aan onderzoeksdoeleinden verband met botmetastasen, vooral voor de overgang van één kankercellen meerdere cellen volgen micrometastasen. In het bijzonder hebben we aangetoond dat IIA injectie kunnen we de interacties tussen kankercellen en diverse omringende cellen in het bot micro visualiseren.
Hoewel slechts de iliacale slagader is het doelwit van injectie voor kankercellen, adviseren wij de scheiding van zowel ader iliacale slagader en uit de omliggende weefsels, en om ze samen te heffen als een bundel. Dit komt omdat de ader en slagader intensief contact met elkaar, en de veneuze vaatwand is dun en gemakkelijk te breken. Daarom is voor een succesvolle injectie, bespaart tijd en moeite bij elkaar houden van de twee vaten, hoewel kankercellen worden geïnjecteerd alleen de slagader. Een 4-0 zijden hechtdraad …
