Marginating-hepatic leukocytes exhibit unique characteristics and distinct immunological functions compared to other leukocyte populations. Here we describe a method for selective harvesting of this specific hepatic cell population, through forced perfusion of the liver of rats or mice. Marginating-hepatic leukocytes seem critical in determining susceptibility to hepatic-related diseases and metastases.
Marginating-hepatic (MH) leukocytes (leukocytes adhering to the sinusoids of the liver), were shown to exhibit unique composition and characteristics compared to leukocytes of other immune compartments. Specifically, evidence suggests a distinct pro- and anti-inflammatory profile of the MH-leukocyte population and higher cytotoxicity of liver-specific NK cells (namely, pit cells) compared to circulating or splenic immunocytes in both mice and rats. The method presented herein enables selective harvesting of MH leukocytes by forced perfusion of the liver in mice and rats. In contrast to other methods used to extract liver-leukocytes, including tissue grinding and biological degradation, this method exclusively yields leukocytes from the liver sinusoids, uncontaminated by cells from other liver compartments. In addition, the perfusion technique better preserves the integrity and the physiological milieu of MH leukocytes, sparing known physiological responses to tissue processing. As many circulating malignant cells and infected cells are detained while passing through the liver sinusoids, physically interacting with endothelial cells and resident leukocytes, the unique MH leukocyte population is strategically located to interact, identify, and react towards aberrant circulating cells. Thus, selective harvesting of MH-leukocytes and their study under various conditions may advance our understanding of the biological and clinical significance of MH leukocytes, specifically with respect to circulating aberrant cells and liver-related diseases and cancer metastases.
De lever sinusoïden bevatten talrijke leukocyt subtypen van verschillende immuun activiteiten kritisch voor het organisme. Zo worden marginating lever (MH) natural killer (NK) cellen, ook bekend als pitcelen kenmerk morfologisch als grote granulaire lymfocyten (LGLs) en functioneel als leukocyten spontane cytotoxische capaciteit dat hepatische weerstand tegen de vestiging van bloed- gedragen tumor uitzaaiingen. Het doel van de werkwijze die hierin wordt selectief oogsten van MH leukocyten, om deze belangrijke en unieke celpopulatie (en immuuncompartiment) bestuderen inschakelen en het effect van verschillende manipulaties (bijv immuunactivatie) de volgende specifieke cellen te helderen.
Ondanks het feit dat immuniteit voor een zich ontwikkelende primaire tumor te elimineren, bewijsmateriaal kankerpatiënten en diermodellen geven aan dat het immuunsysteem regelen circulerende tumorcellen, micrometastasen en residual disease gedugh-cel gemedieerde immuniteit (CMI). Er bestaat echter een duidelijke inconsistentie tussen de in vivo capaciteit en in vitro studies bij mensen en bij dieren die aantonen dat de meeste autologe tumorcellen resistent tegen cytotoxiciteit van circulerende leukocyten of milt 1,2. Deze inconsistentie kan worden toegeschreven, ten minste gedeeltelijk, om de in vivo er een afzonderlijke leukocyten subpopulatie, namelijk de marginating-lever (sinusoïden) leukocyten en de subpopulatie van geactiveerde NK-cellen, namelijk pitcelen 3. Inderdaad, ongepubliceerde gegevens uit ons laboratorium aangetoond dat in F344 ratten syngene tumorcellen (MADB106), die bestand zijn tegen circulerende leukocyten en milt werden gevonden, werden gelyseerd door MH-NK-cellen 4. Aldus kunnen tumorcellen die zogenaamd "NK-resistent" circulerende leukocyten worden gecontroleerd door MH-NK-cellen. Opmerkelijk, in muizen de verhoogde activiteit van de MH-NK is duidelijk alleen na in vivo immuunstimulatie (bijvoorbeeld door gebruikmaking van Poly I: C of CpG-C) 5.
Lever-specifieke NK-cellen (MH-NK) bevinden zich binnen de sinusvormige lumen, die vastzit aan de endotheelcellen en Kupffer cellen. MH-NK-cellen uitsluitend gekenmerkt door dichte bolvormige korrels en-rod uitgeboord vesicles 6, waarbij zuurfosfatase als lysosomale enzymen en perforine en granzymen bevatten bioactieve stoffen 7,8. Vergeleken met circulerende NK-cellen, NK-MH-cellen vertonen een grotere en kleinere granules en blaasjes 9-11. Onder ontstekingsaandoeningen werden MH-NK-cellen aangetoond hogere expressie vertonen van LFA-1 12 ten opzichte van circulerende NK cellen. Deze verhoogde expressie kan een mechanisme waardoor MH-NK-cellen zijn cytotoxischer tegen sommige tumorcellen dan circulerende NK cellen 13,14 vormen. nterestingly, na in vitro incubatie met interleukine (IL) -2, MH-NK-cellen worden vergroot, enhet aantal en de grootte van de korrels te vergroten, die stroken met een fi le of lymfokine geactiveerde killercellen (LAK) cellen 15.
Actieve MH leukocyten kan niet alleen worden verkregen door standaard lever leukocyten oogstmethodes, die gebaseerd zijn op het slijpen en biologische afbraak van het weefsel. Onze aanpak perfusie hierin beschreven heeft twee belangrijke voordelen ten opzichte van de standaard methoden. Ten eerste, de perfusie benadering oogst MH selectief leukocyten voorkomen van besmetting door andere leukocyten uit andere lever compartimenten. Ten tweede, de perfusie techniek beter behoudt de integriteit, de activiteit en de fysiologische milieu van MH leukocyten, anders dan het weefsel verwerking benaderingen die cellen beschadigen of hun morfologie te veranderen en door weefselbeschadiging, het induceren van de afgifte van verschillende factoren die duidelijk moduleren immuunsysteem activiteit.
De lever is een belangrijk doelwit orgaan voor kanker metastase eennd voor verschillende infecties 16. Zoals MH cellen unieke kenmerken is het belangrijk om deze specifieke populatie onder verschillende omstandigheden te bestuderen met betrekking tot deze pathologieën. Zo is het waard op te merken dat systemische activering van het immuunsysteem door verschillende BRM (bijvoorbeeld poly I: C of CpG-C) bleken te activeren MH-leukocyten meer dan 5 circulerende leukocyten.
De lever perfusie methode die hierin maakt een selectief oogsten en het bestuderen van de unieke populatie van marginating lever leukocyten. Lever- NK-cellen, ook wel pit cellen 3, vormen een onderscheidende NK cel populatie die in de lever sinusoïden wonen. Ze zijn te vinden in ratten, muizen 17 en bij de mens 18,19.
