Rabbits are widely used to study the pharmacokinetics of intraocular drugs. We describe a method for conducting pharmacokinetic studies of intraocular drugs using rabbit eyes.
A via intra-ocular de administração da droga permite a entrega de concentrações elevadas de drogas terapêuticas, enquanto minimiza a sua absorção sistémica. Vários fármacos são administrados dentro da câmara anterior ou vítrea, e a injecção intra-ocular tem sido eficaz na cura de várias doenças intra-oculares. Coelho olhos têm sido amplamente utilizadas para a investigação oftálmica, tal como o animal é fácil de manusear e económica em comparação com outros mamíferos, e o tamanho do olho de coelho é semelhante ao de um olho humano. Usando uma agulha G 30, a droga pode ser injectado para dentro dos espaços intracamerais e intravítrea de olhos de coelho. Os globos oculares são então congeladas até à análise e pode ser dividido em humor aquoso e do humor vítreo e retina / coróide. Antes do advento de entrega intra-ocular de droga, a principal preocupação de terapia médica para doenças intra-oculares foi a eficiência com a qual o fármaco pode penetrar no interior do olho. A barreira hemato-ocular impede muitas substâncias, incluindo drogas, de difundir para dentro do olho. Portanto, as concentrações dos fármacos que são acima dos níveis terapêuticos pode não ser facilmente obtido. O método de administração de droga intra-ocular, incluindo injecções intravítreas e intracamerais, é possível ignorar directamente a barreira hemato-ocular 1-3, de modo que concentrações terapêuticas de drogas pode ser conseguida no olho 4,5.
Por conseguinte, a entrega de drogas intravítreas tornou-se um método popular de tratamento para várias doenças intra-oculares 5,6. Por exemplo, injecção intravítrea é amplamente realizada para a degeneração macular relacionada com a idade, retinopatia diabética, oclusão venosa retiniana, e infecções intra-oculares 7-10. Em particular, uma veza introdução de medicamentos anti-VEGF, a frequência das injecções intravítreas foi notavelmente aumentada para o tratamento de doenças da retina. Portanto, é importante compreender a farmacocinética intraoculares de tais drogas para avaliar a eficácia e segurança da terapia médica.
Embora a administração intra-ocular de drogas é considerado um grande avanço na terapia médica para doenças oculares, o monitoramento da concentração da droga dentro do globo ocular é tecnicamente exigente. Uma vez que os olhos humanos contêm apenas pequenas quantidades de humor aquoso (cerca de 200 uL) e vidro (cerca de 4,5 ml, Tabela 1), é tecnicamente difícil obter uma quantidade suficiente de fluido ocular para medir a concentração da droga. Além disso, os métodos que são usados para se obter o líquido do olho, como bater vítreo ou paracentese da câmara anterior, podem danificar o tecido ocular e resultar em complicações graves, tais como cataratas, endoftalmite, oudescolamento de retina 11,12. Por conseguinte, os modelos animais são utilizados em estudos farmacocinéticos de fármacos comumente utilizados intraoculares 13. Entre estes modelos animais, coelhos ou macacos são os animais mais frequentemente utilizados.
Coelhos, que são pequenos mamíferos da ordem Lagomorpha na família dos leporídeos, são encontrados em várias partes do mundo. Porque os coelhos não são agressivos, eles são fáceis de manusear, usar em um experimento, e observar. Baixo custo, disponibilidade imediata do animal, de tamanho semelhante ao olho humano, e uma grande base de dados de informação para a comparação favorável realizar estudos farmacocinéticos usando olhos de coelhos. Neste trabalho, um protocolo para estudos de farmacocinética dos medicamentos intra-ocular em olhos de coelhos é descrito.
With the increasing use of intraocular drugs, such as anti-vascular endothelial growth factor (VEGF) agents, for the treatment of diverse ocular diseases, knowledge of the tissue distribution and clearance of the drug after the intraocular injection is important.
