Rabbits are widely used to study the pharmacokinetics of intraocular drugs. We describe a method for conducting pharmacokinetic studies of intraocular drugs using rabbit eyes.
Het intra-oculaire route van toediening van geneesmiddelen kan de levering van hoge concentraties van therapeutische geneesmiddelen, terwijl het minimaliseren van hun systemische absorptie. Verschillende geneesmiddelen worden toegediend in de voorste kamer of glasvocht, en de intraoculaire injectie effectief in het genezen van diverse ziekten intraoculaire geweest. Konijnenogen zijn op grote schaal gebruikt voor oogheelkundig onderzoek, het dier gemakkelijk te hanteren en goedkoop in vergelijking met andere zoogdieren, en de grootte van een konijn oog is vergelijkbaar met die van een menselijk oog. Met behulp van een 30 G naald, kunnen medicijnen worden geïnjecteerd in de intracamerale en intravitreale ruimten van konijn ogen. De oogbollen worden vervolgens bevroren tot analyse, en kan worden verdeeld in het kamerwater, glasvocht en retina / choroidea. Het glasvocht en retina / choroidea monsters worden gehomogeniseerd en oplosbaar gemaakt vóór de analyse. Vervolgens kunnen immunoassays worden uitgevoerd om de concentraties van intraoculaire drugs meten elk compartiment. Geschikte farmacokinetische modellen kunnen wordengebruikt om verschillende parameters, zoals de halfwaardetijd en maximale concentratie van het geneesmiddel te berekenen. Konijnenogen kan een goed model voor farmacokinetische studies van intraoculaire drugs.
Voor de komst van intraoculaire geneesmiddelafgifte, de belangrijkste zorg van medische therapie voor intraoculaire aandoeningen was de efficiëntie waarmee het geneesmiddel in het oog kan binnendringen. De bloed-oculaire barrière voorkomt veel stoffen, zoals geneesmiddelen, diffundeert in het oog. Daarom concentraties van geneesmiddelen die boven therapeutische niveaus kunnen niet gemakkelijk worden verkregen. De intraoculaire toediening werkwijze, waaronder intracamerale en intravitreale injecties, direct kunnen omzeilen de bloed-oculaire barrière 1-3, zodat therapeutische concentraties van geneesmiddelen kunnen worden bereikt in het oog 4,5.
Daarom heeft intravitreale drug delivery uitgegroeid tot een populaire methode van de behandeling van verschillende intra-oculaire ziekten 5,6. Zo wordt intravitreale injectie wijd uitgevoerd leeftijdsgebonden maculaire degeneratie, diabetische retinopathie, retinale ader occlusies en intraoculaire infecties 7-10. Aangezien met namede introductie van anti-VEGF medicatie is de frequentie van intravitreale injecties opmerkelijk verhoogd voor de behandeling van retinale ziekten. Daarom is het belangrijk om de intraoculaire farmacokinetiek van dergelijke geneesmiddelen begrijpelijk voor het evalueren van de werkzaamheid en veiligheid van de medische behandeling.
Hoewel de intraoculaire toediening van geneesmiddelen wordt beschouwd als een belangrijke doorbraak in de medische therapie voor oculaire ziekten, bewaken van de geneesmiddelconcentratie in de oogbol is technisch veeleisend. Omdat menselijke ogen slechts kleine hoeveelheden waterige humor (ongeveer 200 ui) en kunstmatige (ongeveer 4,5 ml, tabel 1) bevatten, is het technisch moeilijk om voldoende hoeveelheden oogvloeistof de geneesmiddelconcentratie meten verkrijgen. Bovendien methoden die worden gebruikt om de oogvloeistof te verkrijgen, zoals glasachtige tikken of voorste kamer paracentesis kan het oogweefsel beschadigen en ernstige complicaties, zoals cataract, endoftalmitis, ofnetvliesloslating 11,12. Dienovereenkomstig worden diermodellen gebruikt in farmacokinetische studies van de meest gebruikte geneesmiddelen intraoculaire 13. Onder deze diermodellen, konijnen of apen zijn de meest gebruikte dieren.
Konijnen, die kleine zoogdieren van de orde Lagomorpha in de familie Leporidae zijn, worden gevonden in verschillende delen van de wereld. Omdat konijnen zijn niet agressief, ze zijn gemakkelijk te hanteren, te gebruiken in een experiment, en observeren. Lagere kosten, gemakkelijke beschikbaarheid van het dier, vergelijkbaar grootteoogschaduw mens, en een grote database van informatie vergelijking gunst uitvoeren farmacokinetische studies met konijnenogen. In dit artikel wordt een protocol voor farmacokinetisch onderzoek van de intra-oculaire drugs bij konijnen ogen beschreven.
