Immunostaining is an effective technique for visualizing specific cell types and proteins within tissues. By utilizing sonication, the protocol described here alleviates the need to dissect Drosophila melanogaster tissues from late-stage embryos and larvae before immunostaining. We provide an efficient methodology for the immunostaining of formaldehyde-fixed whole mount larvae.
Studies uitgevoerd in Drosophila melanogaster embryo's en larven leveren cruciale inzicht in ontwikkelingsprocessen zoals lot van de cel specificatie en organogenese. Immunokleuring maakt de visualisatie van ontwikkelende weefsels en organen. Echter, een beschermende cuticula die zich vormt aan het eind van embryogenese voorkomt permeatie van antilichamen in late-embryo's en larven. Terwijl dissectie voor immunokleuring wordt regelmatig gebruikt om Drosophila larvale weefsels analyseren inefficiënt sommige analyses blijkt omdat kleine weefsels moeilijk kan zijn te lokaliseren en te isoleren. Sonicatie biedt een alternatief voor dissectie in larvale Drosophila immunostaining protocollen. Het maakt een snelle, gelijktijdige verwerking van grote aantallen late-embryo's en larven en onderhoudt in situ morfologie. Na fixatie in formaldehyde, wordt een monster gesonificeerd. Monster wordt vervolgens onderworpen aan immunokleuring met antigen-specifiek primair antibodies en fluorescent gelabelde secundaire antilichamen aan doelcel types en specifieke proteïnen te visualiseren via fluorescentiemicroscopie. Tijdens het proces van sonicatie, juiste plaatsing van een sonicatie sonde boven het monster, alsmede de duur en intensiteit van sonicatie, kritisch. Additonal kleine wijzigingen standaard immunokleuring protocollen die noodzakelijk zijn voor hoge kwaliteit vlekken. Op antilichamen met een lage signaal-ruisverhouding, langere incubatietijden zijn typisch noodzakelijk. Als een proof of concept voor deze-geluidsgolven gefaciliteerd protocol, laten we immunostains van drie soorten weefsel (testikels, eierstokken, en zenuwweefsel) op een afstand van ontwikkelingsstadia.
Drosophila embryo's en larven bieden een uitstekend model om ontwikkelingsprocessen te studeren in veel organen en weefsels. Beeldvorming van afzonderlijke cellen is vaak noodzakelijk in deze studies om de complexe omgevingen waarin cellen ontwikkelen bepalen. Visualisatie van cellen in weefsels kunnen worden bereikt door immunokleuring. Goed beschreven immuunkleuring protocollen bestaan voor embryonale Drosophila weefsels <17 uur na de eileg (AEL) 1-3. Echter, een beschermende cuticula vormt tegen het einde van embryogenese voorkomende effectieve antilichaam permeatie. Aldus zijn deze immunokleuring protocollen inefficiënt in de analyse van weefsels in late-stadium embryo en in latere fasen van larvale ontwikkeling (1e instar (L1), 2e instar (L2) en 3e instar (L3)). Deze inefficiëntie legt een barrière voor ons begrip van de dynamische processen die optreden tijdens deze uitgebreide ontwikkelingsperiode 4. Tissue dissectie is een uitgebreid toegepast techniek om deze barrière 5-7 omzeilen. Echter, dissectie ondoeltreffend. Extractie kan worden bezwaard door de moeilijkheden bij het lokaliseren of het isoleren van embryonale en larvale weefsels. Verder kan de fysieke verwijdering van doelwitweefsels schade veroorzaken door scheuren hen of door niet uitpakken in hun geheel.
Sonicatie is een methode die geluidsgolven maakt gebruik van intermoleculaire interacties te verstoren. Het is gebruikt om de integriteit van de Drosophila larven cuticula verstoren om immunostain ontwikkelen neurale celtypes 6. Dit protocol is aangepast om late-fase embryonale en larvale geslachtsklieren, van slechts 50 urn diameter 8-10 kan immunostain. Door middel van deze studies, is het proces van de mannelijke kiembaan stamcellen (GSC) niche-formatie werd gekenmerkt in een vergevorderd stadium Drosophila embryo's 8-10 en mechanismen tot regeling van stamcellen ontwikkeling en differentiatie in een vergevorderd stadium embryonale gonaden en larven zijn opgehelderd 9-12. Aldus sonicatie biedt een efficiënt alternatief voor weefsel dissectie die kan moeilijk zijn vanwege weefselgrootte. Bovendien maakt immunokleuring van Drosophila weefsels in situ, waardoor cellen in de context van het hele organisme en handhaven situ morfologie. Hier is een stap-voor-stap protocol voor fluorescentie immunokleuring van late-fase embryonale beschrijven we door vroege / midden-L3 weefsels in situ. Analyse van Drosophila gonadale en neuraal weefsel in de Representatieve resultaten voor de werkzaamheid van dit protocol aangetoond. Bovendien kan dit immunokleuring protocol worden aangepast aan andere Drosophila weefsels en weefsels in andere organismen te analyseren met een buitenste cuticula.
Dit protocol verschaft een werkwijze succesvol immunostain richten Drosophila embryonale en larvale weefsels in situ, waardoor de noodzaak voor dissectie. Als per eerdere protocollen voor het kleuren van de vroege embryo's 1,2,3, is het chorion membraan verwijderd met behulp van 50% bleekwater (NaOCl). Monsters worden gefixeerd in formaldehyde en methanol. Omdat de larvale cuticula veroorzaakt oudere monster drijven, wordt het gehele monster vervolgens door een cel-zeef om larvale retent…
