Cystic fibrosis transmembrane conductance regulator (CFTR), een chloride kanaal voornamelijk gevestigd zijn op de apicale membranen van epitheliale cellen, speelt een cruciale rol in transepitheliaal vloeistof homeostase 1-3. CFTR is betrokken bij twee belangrijke ziekten: cystische fibrose (CF) 4 en secretoire diarree 5. In CF, is de synthese of functionele activiteit van het CFTR Cl-kanaal verminderd. Deze aandoening treft ongeveer 1 op de 2.500 blanken in de Verenigde Staten 6. Overmatige CFTR-activiteit is ook betrokken in geval van toxine-geïnduceerde secretoire diarree (bijvoorbeeld door cholera toxine en warmte stabiele E. coli enterotoxine), die cAMP of cGMP productie in de darm 7 stimuleert.
Opstapelen van bewijsmateriaal dat suggereert het bestaan van fysieke en functionele interacties tussen CFTR en een groeiend aantal andere eiwitten, waaronder vervoerders, ionkanalen, receptoren, kinasen, fosfatasen, signaaling moleculen en cytoskelet elementen en deze interacties tussen CFTR en bindende eiwitten aangetoond kritisch bij de regulering CFTR-gemedieerde transepitheliaal ionentransport in vitro en in vivo 8-19. In dit protocol richten we ons alleen op de methoden die helpen bij de studie van de interacties tussen CFTR carboxyl-terminale staart, die een eiwit-bindend motief bezit [aangeduid als PSD95/Dlg1/ZO-1 (PDZ) motief], en een groep van raamwerk eiwitten die een specifieke binding module bevatten genoemd PDZ domeinen. Tot nu toe zijn verschillende PDZ raamwerk eiwitten is gerapporteerd dat de carboxyl terminal staart van CFTR binden met verschillende affiniteiten zoals Nherf1, Nherf2, PDZK1, PDZK2, CAL (CFTR-geassocieerde ligand), Shank2 en 20-27 bevatten. De PDZ motief in CFTR dat wordt herkend door PDZ steiger eiwitten is de laatste vier aminozuren aan de C-terminus (dat wil zeggen, 1477-Dtrl-1480 in de menselijke CFTR) 20. InteressantCFTR kunnen binden meer dan een PDZ-domein van zowel NHERFs en PDZK1, zij het met verschillende affiniteiten 22. Meerdere biochemische testen zijn ontwikkeld om te studeren CFTR-met eiwit interacties, zoals co-immunoprecipitatie, pull-down test, paarsgewijs bindingstest, colorimetrische paarsgewijs bindingstest, en macromoleculaire complexe assemblage-test 16-19,28,29 . Hier richten we ons op de gedetailleerde procedures voor het samenstellen van een PDZ motief-afhankelijke CFTR-bevattende macromoleculaire complex in vitro, die op grote schaal wordt gebruikt door ons laboratorium om eiwit-eiwit of domein-domain interacties van CFTR 16-19,28,29 te bestuderen.
In dit protocol we aangetoond dat een werkwijze voor in vitro montage en detectie van een CFTR met macromoleculaire signaleringscomplex met gezuiverde eiwitten (of eiwitfragmenten) en / of cellysaten Zoals eerder 16-19,29,30.
