Neurale circuits topografisch georganiseerd in functionele compartimenten specifieke moleculaire profielen. Hier bieden wij de praktische en technische stappen voor het openbaren van globaal brein topografie met behulp van een veelzijdige wholemount immunohistochemische kleuring aanpak. We zien het nut van de methode met behulp van de goed begrepen morfologie en de circuits van cerebellum.
De herhaalde en goed begrepen cellulaire architectuur van de kleine hersenen maken het een ideaal model systeem voor het verkennen van de hersenen topografie. Onderliggende zijn relatief uniforme morfologie is een complexe serie van parasagittal domeinen van gen-en eiwitexpressie. De moleculaire compartimentering van de kleine hersenen wordt weerspiegeld door de anatomische en functionele organisatie van afferente vezels. Om ten volle bewust van de complexiteit van cerebellaire organisatie hebben we eerder een verfijnd wholemount vlekken aanpak voor hoge doorvoer analyse van de patronen gebreken in de muis cerebellum. Dit protocol beschrijft in detail de reagentia, hulpmiddelen en praktische stappen die nuttig zijn om met succes eiwit expressie patronen laten zien in de volwassen muis kleine hersenen met behulp van wholemount immunokleuring. De stappen die hier aandacht aan te tonen het nut van deze methode gebruik van de uitdrukking van zebrinII / aldolaseC als een voorbeeld van hoe de fijne topografie van de hersenen kan worden onthuld in zijnnatieve driedimensionale conformatie. Ook beschreven zijn aanpassingen aan het protocol die het mogelijk maken voor de visualisatie van de expressie van eiwitten in afferente projecties en grote cerebella voor vergelijkende studies van moleculaire topografie. Om deze toepassingen te illustreren, worden de gegevens van afferente kleuring van de rat cerebellum inbegrepen.
We hebben de technische details die nodig zijn voor een succesvolle wholemount kleuring met behulp van een veelzijdige immunohistochemische aanpak voor het openbaren van eiwitexpressie in de ontwikkelende en volwassen hersenen. Door deze aanpak kan complexe moleculaire expressiepatronen geanalyseerd en hersenen topografie gewaardeerd zonder bewerkelijk en tijdrovend weefsel snijden procedures.
