We beschrijven een protocol om de belangrijkste taken van gastheer signaalmoleculen in lytische replicatie van een model herpesvirus, gamma herpesvirus 68 (γHV68) te identificeren. Gebruik te maken van genetisch gemodificeerde muizenstammen en embryonale fibroblasten voor γHV68 lytische replicatie, het protocol maakt het mogelijk zowel de fenotypische karakterisatie en moleculaire ondervraging van virus-gastheer interacties in virale lytische replicatie.
