Este estudio revela el mecanismo de Trichosanthes-Fritillaria thunbergii en el tratamiento del adenocarcinoma de pulmón basado en la farmacología de red y la verificación experimental. El estudio también demuestra que la vía de señalización PI3K / AKT juega un papel vital en la acción de Trichosanthes-Fritillaria thunbergii en el tratamiento del adenocarcinoma de pulmón.
Nuestro objetivo fue estudiar el mecanismo de Trichosanthes-Fritillaria thunbergii en el tratamiento del adenocarcinoma de pulmón (LUAD) basado en la farmacología de la red y la verificación experimental. Los componentes efectivos y los objetivos potenciales de Trichosanthis y Fritillaria thunbergii se recopilaron mediante experimentos de alto rendimiento y bases de datos guiadas por referencia (HERB) de medicina tradicional china y una base de datos de enfoque de conjunto de similitud (SEA), y los objetivos relacionados con LUAD fueron consultados por las bases de datos GeneCards y Online Mendelian Inheritance in Man (OMIM). El software Cytoscape construyó una red de fármacos-componentes-enfermedad-diana. Se realizaron análisis de enriquecimiento de la vía de la red de interacción proteína-proteína (PPI), la función de la ontología génica (GO) y la enciclopedia de genes y genomas de Kyoto (KEGG) para obtener objetivos centrales y vías clave. Para la validación experimental posterior se utilizó un extracto acuoso de células de Trichosanthes-Fritillaria thunbergii y A549. A través de la base de datos HERB y la búsqueda bibliográfica, se examinaron 31 compuestos efectivos y 157 genes diana potenciales de Trichosanthes-Fritillaria thunbergii, de los cuales 144 eran objetivos reguladores de Trichosanthes-Fritillaria thunbergii en el tratamiento del adenocarcinoma de pulmón. El análisis de enriquecimiento funcional GO mostró que el mecanismo de acción de Trichosanthes-Fritillaria thunbergii contra el adenocarcinoma de pulmón es principalmente la fosforilación de proteínas. El análisis de enriquecimiento de la vía KEGG sugirió que el tratamiento del adenocarcinoma de pulmón por Trichosanthes-Fritillaria thunbergii implica principalmente la vía de señalización PI3K / AKT. La validación experimental mostró que un extracto acuoso de Trichosanthes-Fritillaria thunbergii podría inhibir la proliferación de células A549 y la fosforilación de AKT. A través de la farmacología en red y la validación experimental, se verificó que la vía de señalización PI3K / AKT juega un papel vital en la acción de Trichosanthes-Fritillaria thunbergii en el tratamiento del adenocarcinoma de pulmón.
El cáncer de pulmón se refiere a tumores malignos que se originan en la mucosa bronquial pulmonar, incluidos el carcinoma de células escamosas, el adenocarcinoma, el carcinoma de células grandes y el carcinoma de células pequeñas1. El adenocarcinoma de pulmón (LUAD) es el tipo más común de cáncer de pulmón, que representa alrededor del 40% del total de casos de cáncer de pulmón2. La mayoría de los pacientes son diagnosticados en una etapa avanzada o tienen metástasis remotas y, por lo tanto, pierden la oportunidad de la cirugía3. En el tratamiento clínico actual, la quimiorradioterapia concurrente es la estrategia más común para el tratamiento de la LUAD, pero su aplicación es limitada debido a reacciones adversas graves4.
La medicina tradicional china (MTC) puede aliviar eficazmente los síntomas clínicos de los pacientes con LUAD y reducir las reacciones adversas causadas por la radioterapia y la quimioterapia y, por lo tanto, se ha convertido en un punto caliente de investigación 5,6,7. En la medicina tradicional china, el cáncer de pulmón pertenece a la categoría de “acumulación pulmonar” y “petroso pulmonar”. La deficiencia de Qi y la interacción de flema, estasis y veneno son importantes en la patogénesis del cáncer de pulmón. Por lo tanto, tonificar el Qi y eliminar la flema y la estasis sanguínea son el principal tratamiento clínico8 métodos para el cáncer de pulmón según la teoría de la MTC9. Trichosanthes kirilowii Maxim (Gualou) y Fritillaria thunbergii Miq (Zhebeimu) representan un par de fármacos comunes en el tratamiento del cáncer de pulmón, y esta combinación tiene los efectos de eliminar el calor y reducir la flema10,11,12. Sin embargo, su mecanismo de acción aún no está claro, y es necesario realizar más investigaciones.
La farmacología de redes es un método integral basado en la teoría de la biología de sistemas y la farmacología multidireccional que tiene como objetivo revelar relaciones complejas de red entre múltiples fármacos y enfermedades13. Las prescripciones tradicionales chinas tienen las características de ser multicomponente y multiobjetivo, por lo que son muy adecuadas para el estudio de la farmacología de red14,15. Recientemente, la farmacología en red ha surgido como un enfoque poderoso en el estudio de las fórmulas de MTC y se ha convertido en un punto caliente de investigación16,17.
Sin embargo, hasta donde sabemos, todas las investigaciones sobre farmacología de redes se presentan como texto. Presentar esta tecnología a través de video reducirá en gran medida el umbral de aprendizaje y facilitará la promoción de esta tecnología, que es una de las ventajas de este artículo. En este estudio, tomamos Trichosanthes-Fritillaria thunbergii contra adenocarcinoma de pulmón como ejemplo para llevar a cabo la predicción de farmacología en red y la validación experimental.
En general, un estudio completo de farmacología de red incluye la identificación de componentes activos a partir de bases de datos, la adquisición de objetivos correspondientes a componentes activos y enfermedades, la construcción de una red de fármacos-componentes-enfermedad-diana, y la predicción de objetivos y vías centrales.
