Hier wordt een protocol gepresenteerd om een snel en niet-destructief systeem te bouwen voor het meten van cel- of kerncompressie op basis van acoustofluidic microdevice. Veranderingen in mechanische eigenschappen van tumorcellen na epitheliaal-mesenchymale overgang of ioniserende straling werden onderzocht, wat het toepassingsperspectief van deze methode in wetenschappelijk onderzoek en klinische praktijk aantoont.
Celmechanica speelt een belangrijke rol bij tumormetastase, kwaadaardige transformatie van cellen en radiosensitiviteit. Tijdens deze processen is het bestuderen van de mechanische eigenschappen van de cellen vaak een uitdaging. Conventionele meetmethoden op basis van contact, zoals compressie of stretching, kunnen celbeschadiging veroorzaken, wat de meetnauwkeurigheid en de daaropvolgende celcultuur beïnvloedt. Metingen in hechtende toestand kunnen ook de nauwkeurigheid beïnvloeden, vooral na bestraling, omdat ioniserende straling cellen zal afvlakken en de hechting zal verbeteren. Hier is een celmechanisch meetsysteem ontwikkeld op basis van een acoustofluidische methode. De samendrukbaarheid van de cel kan worden verkregen door het bewegingstraject van de cel te registreren onder invloed van de akoestische kracht, die snelle en niet-destructieve metingen in zwevende toestand kan realiseren. Dit artikel rapporteert in detail de protocollen voor chipontwerp, monstervoorbereiding, trajectregistratie, parameterextractie en -analyse. De samendrukbaarheid van verschillende soorten tumorcellen werd gemeten op basis van deze methode. Meting van de samendrukbaarheid van de kern werd ook bereikt door de resonantiefrequentie van het piëzo-elektrische keramiek en de breedte van het microkanaal aan te passen. In combinatie met de moleculaire niveauverificatie van immunofluorescentie-experimenten werd de celcompressie-voor en na geneesmiddel-geïnduceerde epitheliale naar mesenchymale overgang (EMT) vergeleken. Verder werd de verandering van de samendrukbaarheid van cellen na röntgenbestraling met verschillende doses onthuld. De celmechanica meetmethode die in dit artikel wordt voorgesteld, is universeel en flexibel en heeft brede toepassingsperspectieven in wetenschappelijk onderzoek en de klinische praktijk.
Celmechanische eigenschappen spelen een belangrijke rol bij tumormetastase, kwaadaardige transformatie van cellen en radiogevoeligheid 1,2. Om een diepgaand inzicht te krijgen in de rol van celmechanische eigenschappen in het bovenstaande proces, is nauwkeurige meting van cellulaire mechanica van cruciaal belang en mag de meting geen schade aan de cellen veroorzaken voor daaropvolgende cultuur en analyse. Het meetproces moet zo snel mogelijk verlopen, anders kan de levensvatbaarheid van cellen worden aangetast als cellen voor een lange tijd uit de kweekomgeving worden verwijderd.
Bestaande celmechanica meetmethoden hebben te maken met enkele beperkingen. Sommige methoden, zoals magnetische draaiende cytometrie, magnetisch pincet en deeltjesvolgende microrheologie, veroorzaken celschade door de introductie van deeltjes in cellen 3,4,5. Methoden die meten door contact met cellen, zoals atoomkrachtmicroscoop (AFM), micropipette-aspiratie, microvernauwing en parallelplaattechniek, zijn ook gevoelig voor celbeschadiging en de doorvoer is moeilijk te verhogenmet 6,7,8. Bovendien zal ioniserende straling cellen afvlakken en hun hechting verhogen9; het is daarom noodzakelijk om de hele celmechanica in suspensie te meten.
Als antwoord op de bovenstaande uitdagingen is een celmechanisch meetsysteem ontwikkeld op basis van acoustofluidische methode10,11,12,13,14. De kanaalbreedte is afgestemd op de akoestische halve golflengte, waardoor een staande golfknoop ontstaat op de middellijn van het microkanaal. Onder invloed van akoestische stralingskracht kunnen de cellen of standaardparels naar de akoestische drukknoop bewegen. Omdat de fysische eigenschappen van de standaard kralen (grootte, dichtheid en samendrukbaarheid) bekend zijn, kan de akoestische energiedichtheid worden bepaald. Vervolgens kan de samendrukbaarheid van de cel worden verkregen door de bewegingstrajecten van cellen in het akoestische veld te registreren. Niet-destructieve high-throughput meting van cellen in suspensietoestand kan worden bereikt. Dit artikel introduceert het ontwerp van de microfluïdische chip, de oprichting van het systeem en de meetstappen. Meting van verschillende soorten tumorcellen is uitgevoerd om de nauwkeurigheid van de methode te verifiëren. Het toepassingsgebied van deze methode was uitgebreid tot subcellulaire structuren (zoals kern) door de resonantiefrequentie van het piëzo-elektrische keramiek en de breedte van het microkanaal aan te passen. Daarnaast werden de veranderingen in celcompressie na geneesmiddel-geïnduceerde EMT of röntgenbestraling met verschillende doses onderzocht. De resultaten tonen de brede toepasbaarheid van deze methode als een krachtig hulpmiddel voor het bestuderen van de correlatie tussen biochemische veranderingen en cellulaire mechanische eigenschappen.
Veelgebruikte celmechanica meetmethoden zijn AFM, micropipette aspiratie, microfluïdica methoden, parallel-plaat techniek, optische pincet, optische brancard en akoestische methoden20.
