Dit rapport beschrijft technieken om gesulfateerde glycosaminoglycanen (GAG’s) uit biologische monsters te isoleren en te zuiveren en een polyacrylamidegel-elektroforesebenadering om hun grootte te benaderen. GAG’s dragen bij aan de weefselstructuur en beïnvloeden signaleringsprocessen via elektrostatische interactie met eiwitten. GAG-polymeerlengte draagt bij aan hun bindingsaffiniteit voor verwante liganden.
Gesulfateerde glycosaminoglycanen (GAG’s) zoals heparansulfaat (HS) en chondroïtinesulfaat (CS) zijn alomtegenwoordig in levende organismen en spelen een cruciale rol in een verscheidenheid aan fundamentele biologische structuren en processen. Als polymeren bestaan GAG’s als een polydisperse mengsel met polysaccharideketens die kunnen variëren van 4000 Da tot ruim 40.000 Da. Binnen deze ketens bestaan domeinen van sulfatie, die een patroon van negatieve lading verlenen dat interactie met positief geladen residuen van verwante eiwitliganden vergemakkelijkt. Gesulfateerde domeinen van GAG’s moeten voldoende lang zijn om deze elektrostatische interacties mogelijk te maken. Om de functie van GAG’s in biologische weefsels te begrijpen, moet de onderzoeker in staat zijn om de grootte van GAG’s te isoleren, te zuiveren en te meten. Dit rapport beschrijft een praktische en veelzijdige polyacrylamide gel elektroforese-gebaseerde techniek die kan worden gebruikt om relatief kleine verschillen in grootte tussen GAG’s geïsoleerd uit een verscheidenheid aan biologische weefseltypen op te lossen.
Glycosaminoglycanen (GAG’s) zijn een diverse familie van lineaire polysacchariden die een alomtegenwoordig element zijn in levende organismen en bijdragen aan vele fundamentele fysiologische processen1. GAG’s zoals heparansulfaat (HS) en chondroïtinesulfaat (CS) kunnen op verschillende posities langs de polysaccharideketen worden gesulfateerd, waardoor geografische domeinen van negatieve lading worden overgedragen. Deze GAG’s, wanneer ze worden vastgebonden aan celoppervlakeiwitten die bekend staan als proteoglycanen, projecteren in de extracellulaire ruimte en binden aan verwante liganden, waardoor de regulatie van zowel cis- (ligand gehecht aan dezelfde cel) als trans- (ligand gehecht aan naburige cel) signaleringsprocessen mogelijk is2. Bovendien vervullen GAG’s ook kritieke rollen als structurele elementen in weefsels zoals het glomerulaire keldermembraan3,de vasculaire endotheelglyx4 en pulmonale epitheliale glycocalyx5, en in bindweefsels zoals kraakbeen6.
De lengte van GAG-polysaccharideketens varieert aanzienlijk afhankelijk van de biologische context en kan dynamisch worden verlengd, gesplitst en gewijzigd door een zeer complex enzymatisch regulerend systeem7. Belangrijk is dat de lengte van GAG-polymeerketens substantieel bijdraagt aan hun bindingsaffiniteit voor liganden en vervolgens aan hun biologische functie8,9. Om deze reden vereist de bepaling van de functie van een endogene GAG waardering van de grootte ervan. Helaas bestaan er, in tegenstelling tot eiwitten en nucleïnezuren, zeer weinig direct beschikbare technieken om GAG’s te detecteren en te meten, wat historisch heeft geresulteerd in relatief beperkt onderzoek naar de biologische rollen van deze diverse polysaccharidefamilie.
Dit artikel beschrijft hoe GAG’s uit de meeste biologische weefsels kunnen worden geïsoleerd en gezuiverd en beschrijft ook hoe polyacrylamidegelelektroforese (PAGE) kan worden gebruikt om de lengte van de geïsoleerde polymeren met een redelijke mate van specificiteit te evalueren. In tegenstelling tot andere, zeer complexe (en vaak op massaspectrometrie gebaseerde) glycomische benaderingen, kan deze methode worden gebruikt met behulp van standaard laboratoriumapparatuur en -technieken. Deze praktische benadering kan daarom het vermogen van onderzoekers uitbreiden om de biologische rol van inheemse GAG’s en hun interactie met contextueel belangrijke liganden te bepalen.
GAG’s spelen een centrale rol in veel verschillende biologische processen.
