We presenteren een protocol om primaire volwassen fibroblasten op een eenvoudige, snelle en betrouwbare manier te isoleren, die door beginners kunnen worden perforeerbaar (bijv. studenten). De procedure combineert enzymatische weefsel spijsvertering en mechanische opwinding met ultrasone golven om primaire fibroblasten te verkrijgen. Het protocol kan gemakkelijk worden aangepast aan specifieke experimentele eisen (bijv. menselijk weefsel).
Primaire volwassen fibroblasten zijn uitgegroeid tot een belangrijk instrument om fibrose te bestuderen, fibroblast interacties en ontsteking in alle lichaamsweefsels. Omdat primaire fibroblasten niet voor onbepaalde tijd kunnen delen door myofibroblast-differentiatie of senescentie-inductie, moeten er regelmatig nieuwe culturen worden vastgesteld. Echter, er zijn verschillende obstakels te overwinnen tijdens de processen van de ontwikkeling van een betrouwbare isolatie protocol en primaire fibroblast isolatie zelf: de moeilijkheidsgraad van de methode (vooral voor beginners), het risico van bacteriële besmetting, de vereiste tijd tot primaire fibroblasten kunnen worden gebruikt voor experimenten, en de daaropvolgende celkwaliteit en levensvatbaarheid. In deze studie, een snelle, betrouwbare en gemakkelijk te leren protocol om primaire volwassen fibroblasten te isoleren en te cultuur van muis hart, longen, lever en nieren combineren enzymatische spijsvertering en ultrasone agitatie is voorzien.
Fibroblasten zijn vlakke, spindle-vormige cellen met meerdere ecoagriturismo late processen en een uitgebreid ruw endoplasmisch reticulum1,2. Een gemiddelde fibroblast meet 30-100 μm en heeft een levensduur van 57 ± 3 dagen1,3. De gemiddelde celcyclus duur van menselijke fibroblasten varieert van 16-48 h, afhankelijk van de cultuuromstandigheden4. Er zijn aanwijzingen dat de replicatieve capaciteit en de functionele kwaliteit van gekweekte primaire fibroblasten negatief correleerden met de leeftijd van de donor, wat suggereert dat jongere donoren (dieren of patiënten) de voorkeur moeten krijgen indien mogelijk5,6 .
Fibroblasten vormen een overheersend celtype van de meeste lichaamsweefsels van zoogdieren. Ondanks hun alomtegenwoordige aanwezigheid is de moleculaire identificatie van fibroblasten nog steeds een uitdaging7. Fibroblasten migreren naar de ontwikkeling van weefsels en organen uit verschillende bronnen tijdens de embryonale ontwikkeling8. Om deze reden is er een overvloed aan marker eiwitten die kan worden gevonden in fibroblasten, terwijl unieke marker eiwitten, die aanwezig zijn in elke fibroblast populatie en exclusief voor fibroblasten, nog steeds ontbreken. Zo worden expressie patronen van verschillende herkende markers meestal gebruikt om fibroblasten te identificeren. Onder de meest erkende markers zijn vimentin, menselijk fibroblast oppervlak eiwit (hFSP), discoidin domein receptor 2 (DDR2) en alpha glad spier actine (αsma).
Fibroblasten zijn de belangrijkste extracellulaire matrix (ECM)-producerende celtype. Daardoor behouden fibroblasten een geordende weefsel architectuur en bieden ze mechanische ondersteuning voor naburige cellen1. De balans tussen ECM-synthese en afbraak is een goed gereguleerd proces. Verschuivingen in de richting van synthese markeren het begin van overmatige ECM-depositie die, indien niet beëindigd, leidt tot fibrose. Fibrose wordt gemedieerd door myofibroblasten, die afkomstig zijn van geactiveerde fibroblasten die moleculaire en fenotypische veranderingen ondergaan. Een kenmerk van myofibroblasten is verbeterde secretie van ECM en cytokinen en de uitdrukking van ordelijke gearrangeerde αSMA Microfilamenten9.
Primaire fibroblasten zijn in de schijnwerpers van recent onderzoek gericht op fibrose, weefsel ontsteking en fibroblast-kanker-celinteracties10,11. Echter, om effectief te bestuderen fibroblast eigenschappen in gezondheid en ziekte, het is noodzakelijk om te isoleren van levensvatbare primaire volwassen fibroblasten op een regelmatige basis. Er zijn verschillende methoden beschikbaar voor het isoleren van fibroblasten12,13,14. De drie belangrijkste methoden van fibroblast isolatie zijn uitgroei uit weefsel chunks12, enzymatische weefsel spijsvertering15, en enzymatische perfusie van holleorganen 9,13,16. Het voordeel van uitgroei is een zacht isolatie proces zonder enzymatische celdegradatie. Aan de andere kant vereisen de groei culturen meestal langdurige cultuur perioden totdat cellen kunnen worden gebruikt voor experimenten. Veelvoorkomende enzymatische spijsvertering is snel, maar draagt een risico van besmetting met andere celtypen (bijv. endotheel cellen) of bacteriën in het roer proces, die nodig zijn om het weefsel mechanisch te ontbinden. Bovendien, deze methoden zijn vaak uitgebreid en vereisen tijd en vaardigheid om te leren.
Wat betreft het belang van primaire fibroblasten in het onderzoek, is er nog steeds behoefte aan het optimaliseren van bestaande celisolatiebenaderingen in termen van snelheid, eenvoud en betrouwbaarheid. Hier wordt een nieuwe Ultrasone-gebaseerde enzymatische fibroblast-isolatiemethode geleverd die hoge kwaliteit cellen levert.
In vergelijking met vereeuwigd fibroblast cellijnen bieden primaire fibroblasten verschillende voordelen. Ze kunnen worden geïsoleerd kosteneffectief in hoge kwaliteit en kwantiteit. Bovendien bieden primaire culturen de mogelijkheid om cellen van meerdere individuen te bestuderen, wat de betrouwbaarheid van de verkregen resultaten verhoogt en de kans verkleint dat alleen celcultuurartefacten worden bestudeerd.
