Taste is an important sensory process which facilitates attraction to beneficial substances and avoidance of toxic substances. This protocol describes a simple ingestion assay for determining Drosophila gustatory preference for a given chemical compound.
Olfactory and gustatory perception of the environment is vital for animal survival. The most obvious application of these chemosenses is to be able to distinguish good food sources from potentially dangerous food sources. Gustation requires physical contact with a chemical compound which is able to signal through taste receptors that are expressed on the surface of neurons. In insects, these gustatory neurons can be located across the animal’s body allowing taste to play an important role in many different behaviors. Insects typically prefer compounds containing sugars, while compounds that are considered bitter tasting are avoided. Given the basic biological importance of taste, there is intense interest in understanding the molecular mechanisms underlying this sensory modality. We describe an adult Drosophila taste assay which reflects the preference of the animals for a given tastant compound. This assay may be applied to animals of any genetic background to examine the taste preference for a desired soluble compound.
Dieren gebruiken chemosensation om gunstige voorwaarden uit elkaar te onderscheiden van ongunstige omstandigheden. Deze waarneming kan cruciaal voor zaken als de bepaling van de beste voedselbron, het vermijden van giftige stoffen en het bepalen van de beste paring partner 1 zijn. Chemosensation is vaak verdeeld in twee zintuiglijke componenten: reukzin en smaak zintuigen. Een belangrijk kenmerk van deze zinnen is dat olfactie (reuk) wordt gebruikt om de omringende gasvormige chemische omgeving tijdens proeven gustation (smaak) fysiek contact met een niet-vluchtig substraat vereist. Zowel sensorische modaliteiten stimuleren neurologische reacties die worden verwerkt en gedecodeerd in de hersenen om de juiste aantrekkelijk of afstotende gedrag 2 produceren. Deze zintuigen zijn daarom van cruciaal belang voor de overleving van dieren.
De vrucht Drosophila melanogaster is een model organisme dat groeit in populariteit voor gebruik in begrijpening hoe insecten waarnemen geur en smaak. Fruitvliegen bieden enorme voordelen boven andere modelsystemen te wijten aan de rijkdom van genetische hulpmiddelen beschikbaar voor de dissectie van de moleculaire, cellulaire en gedrags paden. Het werk van de afgelopen 15 jaar heeft vooral instrumenteel in het karakteriseren van de specifieke cellulaire identiteiten, neuronale receptoren en signalering mechanismen die betrokken zijn bij zowel de geur en smaak zijn. Nu wordt het vermogen van Drosophila genetica worden gebruikt om verder te ontrafelen hoe deze processen worden gecodeerd op één neuron en enkel circuit niveau 3-6. Daarom testen die voorzien gemakkelijk scoorde uitlezingen van wijzigingen aan de sensorische wegen zijn van vitaal belang voor de aanhoudende opmars van deze gebieden.
Hoewel veel bekend over hoe olfactorische signalen worden gecodeerd en verwerkt in de hersenen, is veel minder begrepen land dezelfde mechanismen in de smaak route. We beschrijven hier een protocol dat kan worden gebruikt om smaak preferen vernemence in Drosophila. Drosophila, zoals zoogdieren, in het algemeen de voorkeur aan zoete smaak verbindingen in tegenstelling tot bitter smakende verbindingen. Elke combinatie van deze voedselbronnen kunnen worden gebruikt in deze proefopzet te bepalen hoe bekende genetische veranderingen beïnvloeden smaak keuze. Bovendien kunnen farmacologische interventiestrategieën eveneens worden beoordeeld op hun effect op smaak dieren voorkeur. Het gemak en flexibiliteit van deze test maakt het een nuttig model voor het begrijpen van de aard van smaak- perceptie bij Drosophila.
We hebben beschreven een eenvoudige maar effectieve protocol voor het bepalen van de smaak voorkeur in Drosophila. Versies van deze test worden routinematig gebruikt in experimenten om de bijdragen van smaak receptoren (GR) vast te stellen in het waarnemen van de verschillende kwaliteiten (bitter, zoet, zuur, zout en umami) van smaak verbindingen.
