The epithelial cells of the choroid plexus (CP) form the blood-cerebrospinal fluid barrier (BCSFB). An in vitro model of the BCSFB employs human choroid plexus papilloma (HIBCPP) cells. This article describes culturing and basolateral infection of HIBCPP cells using a cell culture filter insert system.
The epithelial cells of the choroid plexus (CP), located in the ventricular system of the brain, form the blood-cerebrospinal fluid barrier (BCSFB). The BCSFB functions in separating the cerebrospinal fluid (CSF) from the blood and restricting the molecular exchange to a minimum extent. An in vitro model of the BCSFB is based on cells derived from a human choroid plexus papilloma (HIBCPP). HIBCPP cells display typical barrier functions including formation of tight junctions (TJs), development of a transepithelial electrical resistance (TEER), as well as minor permeabilities for macromolecules. There are several pathogens that can enter the central nervous system (CNS) via the BCSFB and subsequently cause severe disease like meningitis. One of these pathogens is Neisseria meningitidis (N. meningitidis), a human-specific bacterium. Employing the HIBCPP cells in an inverted cell culture filter insert system enables to study interactions of pathogens with cells of the BCSFB from the basolateral cell side, which is relevant in vivo. In this article, we describe seeding and culturing of HIBCPP cells on cell culture inserts. Further, infection of the cells with N. meningitidis along with analysis of invaded and adhered bacteria via double immunofluorescence is demonstrated. As the cells of the CP are also involved in other diseases, including neurodegenerative disorders like Alzheimer`s disease and Multiple Sclerosis, as well as during the brain metastasis of tumor cells, the model system can also be applied in other fields of research. It provides the potential to decipher molecular mechanisms and to identify novel therapeutic targets.
La barriera sangue fluido cerebrospinale (BCSFB) è uno dei tre siti barriera tra il sangue e il cervello 1. La correlazione morfologica sono le cellule epiteliali del plesso coroide (CP) 2,3, un convoluto endoteliale-epiteliale, che è fortemente vascolarizzato e situato nei ventricoli del cervello. Il CP serve per produrre il liquido cerebrospinale (CSF), nonché a separare quest'ultima dal sangue. Al fine di realizzare la funzione di barriera, le cellule epiteliali CP mostrano una bassa attività pinocitotiche, esprimono trasportatori specifici, e sono densamente collegati da una rete continua di giunzioni strette (TJS) 2,3.
Umana coroide plesso papilloma (HIBCPP), le cellule, derivate da una maligna coroide plesso papilloma di una donna giapponese a 4, sono stati usati per costruire un funzionale modello in vitro del BCSFB. cellule HIBCPP mostrano un paio di caratteristiche di un BCSFB funzionale come la formazione di TJfili, lo sviluppo di un alto potenziale di membrana transepiteliale che può essere determinato come la resistenza elettrica transepiteliale (TEER) e permeabilità minori per macromolecole. Inoltre, le cellule HIBCPP esprimono trasportatori caratteristici, che possono servire a regolare il microambiente ionico, e mostrare apicale / basolaterale 5,6,7 polarità.
Il BCSFB ha dimostrato di funzionare come un sito di ingresso per agenti patogeni (batteri, virus e funghi) nel sistema nervoso centrale (CNS) 8. L'invasione di patogeni, tra cui Neisseria meningitidis (N. meningitidis), un batterio Gram-negativo, può causare malattie gravi come la meningite. La prova che supera la barriera epiteliale protettiva del CP è supportata da osservazioni istopatologici nei pazienti con malattia meningococcica esibendo una maggiore quantità di meningococchi nei vasi e le cellule epiteliali CP 9,10. Per ottenere l'ingresso nelle cellule ospiti bacteria spesso dirottare meccanismi di endocitosi, che sono mediati o attivati da specifici recettori di superficie situati sulle cellule ospiti. Poiché interazioni di patogeni con questi recettori possono essere specie modelli 11, animali specifici possono essere consultati solo in misura limitata. La linea cellulare HIBCPP offre l'opportunità di studiare il processo di invasione nonché i meccanismi molecolari alla base in un sistema modello umano. Impiegando inserti di coltura cellulare ci permette di analizzare le interazioni di agenti patogeni con cellule ospiti da due lati delle cellule distinte. Molti batteri, tra cui N. meningitidis, sono fortemente soggetta all'impatto di gravità durante saggi di infezione. Per l'interazione ottimale dei patogeni con cellule HIBCPP durante i saggi, i batteri sono inizialmente aggiunti nel vano superiore del sistema di cartuccia del filtro di coltura cellulare. Per attivare infezione apicale o il lato della cella basolaterale rispettivamente, due varianti del sistema in vitro sono stati estafissare dei: Nel sistema standard cellule HIBCPP vengono seminate nel vano superiore dell'inserto filtrante, imitando la situazione quando i microrganismi si trovano sul CSF-lato ed entrare in contatto con il lato apicale delle cellule (Figura 1A, C). Al contrario, utilizzando le cellule HIBCPP in un sistema di cartuccia del filtro di coltura cellulare invertito riflette le condizioni quando i batteri sono entrati nel flusso sanguigno. Microrganismi diffondere nel sangue e di incontro CP cellule epiteliali dal lato basolaterale (Figura 1B, D). Degno di nota, in questo sistema modello è stato dimostrato che i batteri invadono le cellule HIBCPP in modo polare specificamente dal basolaterale 5,7 lato della cella.
In seguito alle infezioni del CP, gli agenti patogeni invasi possono essere riconosciuti dal sistema immunitario innato attraverso la legatura ai recettori pattern-recognition (PRR). i membri ben descritto dei PRRs appartengono alla famiglia dei recettori Toll-like (TLR). può TLR bind per strutture caratteristiche di microrganismi infettivi, che sono modelli patogeni associati definito molecolari (PAMPs).
