Protein abundance reflects the rates of both protein synthesis and protein degradation. This article describes the use of cycloheximide chase followed by western blotting to analyze protein degradation in the model unicellular eukaryote, Saccharomyces cerevisiae (budding yeast).
Regulering van eiwit overvloed is cruciaal voor vrijwel elke cellulaire proces. Eiwitgehalte weerspiegelt de integratie van de snelheden van eiwitsynthese en eiwitafbraak. Veel assays rapportage over eiwit overvloed (bijvoorbeeld één tijdstip western blotting, flowcytometrie, fluorescentie microscopie, of-groei op basis reporter assays) geen discriminatie van de relatieve effecten van de vertaling en proteolyse op eiwitgehalten niet toestaan. Dit artikel beschrijft het gebruik van cycloheximide chase gevolgd door Western blotting om specifiek wat eiwitafbraak in het model eencellige eukaryoot, Saccharomyces cerevisiae (gist). In deze procedure worden gistcellen geïncubeerd in aanwezigheid van de translationele remmer cycloheximide. Hoeveelheden van cellen onmiddellijk verzameld na en op specifieke tijdstippen na toevoeging van cycloheximide. Cellen worden gelyseerd en de lysaten worden gescheiden door polyacrylamidegelelektroforese for western blot analyse van eiwitten overvloed op elk tijdstip. De cycloheximide chase werkwijze maakt visualisatie van de afbraakkinetiek van de steady state populatie van verschillende cellulaire eiwitten. De procedure kan worden gebruikt om de genetische behoefte en omgevingsinvloeden op eiwitafbraak onderzoeken.
Eiwitten uit te voeren cruciale functies in vrijwel elke cellulaire proces. Vele fysiologische processen vereisen de aanwezigheid van een specifiek eiwit (of eiwitten) voor een bepaalde periode of onder bepaalde omstandigheden. Organismen daarom controleren en reguleren eiwit overvloed aan mobiele behoeften 1 te voldoen. Bijvoorbeeld, cyclinen (eiwitten die celdeling) aanwezig op specifieke fasen van de celcyclus en het verlies van cycline gereguleerde niveau is geassocieerd met kwaadaardige tumorvorming 2. Naast het reguleren van eiwit niveaus om cellulaire behoeften te voldoen, cellen in dienst afbrekende kwaliteitscontrole mechanismen om verkeerd gevouwen, niet gemonteerd of anderszins afwijkende eiwitmoleculen 3 te elimineren. Controle van eiwit overvloed impliceert regulering van beide macromoleculaire synthese (transcriptie en vertaling) en afbraak (RNA verval en proteolyse). Gestoorde of overmatige afbraak van eiwitten bijdraagt aan meerdere pathologieën, Waaronder kanker, cystische fibrose, neurodegeneratieve aandoeningen en cardiovasculaire aandoeningen 4-8. Proteolytische mechanismen vertegenwoordigen daarom veelbelovende therapeutische doelwitten voor een reeks van ziekten 9-12.
Analyse van eiwitten op een enkel tijdstip (bijvoorbeeld door western blot 13, 14 flowcytometrie of fluorescentiemicroscopie 15) geeft een overzicht van de steady-state-eiwit overvloed zonder het openbaren van de relatieve bijdrage van de synthese of degradatie. Evenzo groei gebaseerde reporter assays tijdens steady state eiwitniveaus over een langere tijdsperiode zonder onderscheid tussen de invloeden van de synthese en degradatie 15-20. Het is mogelijk de bijdrage van afbrekende processen steady state eiwitniveaus afgeleid door vergelijking overvloed voor en na remming van specifieke onderdelen van de afbrekende mechanisme (bijvoorbeeld door het inactiveren farmacologisch proteasome 21 of het uitspelen van een gen dat de hypothese vereist zijn voor de afbraak 13). Een verandering in de steady state eiwitniveaus na remmen afbraakroutes sterk bewijs voor de bijdrage van proteolyse de controle eiwitgehalte 13. Echter, een dergelijke analyse nog steeds geen informatie verstrekken over de kinetiek van de omzet eiwit. Cycloheximide chase gevolgd door western blotting overwint deze tekortkomingen doordat onderzoekers om eiwitafbraak te visualiseren in de tijd 22-24. Omdat verder eiwitdetectie volgende cycloheximide chase typisch uitgevoerd door middel van Western blotting, radioactieve isotopen en langdurige immunoprecipitatie stappen niet vereist voor cycloheximide chase, in tegenstelling tot veel gangbare pulse chase technieken, die eveneens uitgevoerd om eiwitafbraak 25 te visualiseren.
Cycloheximide werd eerst geïdentificeerd als een verbinding met anti-schimmel juistebanden geproduceerd door de gram-positieve bacterie Streptomyces griseus 26,27. Het is een cel-permeabel molecuul dat specifiek eukaryote cytosolische (maar niet organellen) vertaling remt door afbreuk ribosomale translocatie 28-31. In een cycloheximide chase experiment wordt cycloheximide toegevoegd aan cellen en monsters van cellen onmiddellijk op specifieke tijdstippen na toevoeging van de verbinding 22 verzameld. Cellen worden gelyseerd en eiwitgehalte op elk tijdstip geanalyseerd, typisch door western blot. Afneemt eiwitgehalte na toevoeging van cycloheximide veilig kan worden toegeschreven aan eiwitafbraak. Een instabiele eiwit zal afnemen in overvloed in de tijd, terwijl een relatief stabiele eiwit weinig verandering in overvloed zal vertonen.
Mechanismen van selectieve afbraak van eiwitten zijn sterk geconserveerd in heel Eukarya. Veel van wat bekend is over eiwitafbraak werd voor het eerst geleerd inhet model eencellige eukaryoot, Saccharomyces cerevisiae (gist) 25,32-36. Studies met gist zijn waarschijnlijk blijven leveren nieuwe en belangrijke inzichten in eiwitafbraak. Werkwijze voor cycloheximide chase in gistcellen gevolgd door Western blot analyse van eiwitten overvloed wordt hier gepresenteerd.
In dit artikel wordt een methode voor het analyseren van eiwitafbraak kinetiek gepresenteerd. Deze techniek kan gemakkelijk worden toegepast op een reeks van proteïnen gedegradeerd door verschillende mechanismen. Het is belangrijk op te merken dat cycloheximide chase experimenten rapporteren afbraakkinetiek van de steady state pool van een bepaald eiwit.
