Here, we present a protocol to quantify the avoidance of stressed individuals. This paradigm is powerful yet user-friendly and can be used to assess the influence of genes and environment on one kind of social interaction in Drosophila melanogaster.
Drosophila melanogaster is an emerging model to study different aspects of social interactions. For example, flies avoid areas previously occupied by stressed conspecifics due to an odorant released during stress known as the Drosophila stress odorant (dSO). Through the use of the T-maze apparatus, one can quantify the avoidance of the dSO by responder flies in a very affordable and robust assay. Conditions necessary to obtain a strong performance are presented here. A stressful experience is necessary for the flies to emit dSO, as well as enough emitter flies to cause a robust avoidance response to the presence of dSO. Genetic background, but not their group size, strongly altered the avoidance of the dSO by the responder flies. Canton-S and Elwood display a higher performance in avoiding the dSO than Oregon and Samarkand strains. This behavioral assay will allow identification of mechanisms underlying this social behavior, and the assessment of the influence of genes and environmental conditions on both emission and avoidance of the dSO. Such an assay can be included in batteries of simple diagnostic tests used to identify social deficiencies of mutants or environmental conditions of interest.
Het doel van deze methode is om gemakkelijk te kwantificeren een nieuw aspect van eenvoudige sociaal gedrag in Drosophila melanogaster, onafhankelijk van verkering en agressie.
Sociale interacties zijn cruciaal voor de goede ontwikkeling en gezondheid van individuen binnen een samenleving, evenals de functionaliteit van een sociale groep als geheel. De hoge complexiteit van deze interacties noodzakelijk grote aantallen individuen en een systeem waarmee vereenvoudiging van het gedrag als de genetische en neurale grondslagen van sociaal gedrag zijn nog slecht begrepen. Drosophila melanogaster is een krachtige genetisch model dat kan worden gebruikt om de genetische identificatie en neurale grondslagen van sociale interacties. Inderdaad, D. melanogaster heeft een repertoire van complex sociaal gedrag en een aantal directe metingen van socialisatie zijn al gedaan 1-7. De meeste van deze pogingen zijn gericht op relatief complex sociaal gedrag, zoals agressive interacties 3,6, diverse aspecten van de hofmakerij 3,8-12, en hoe sociale ervaring beïnvloedt andere gedragingen zoals leren, of circadiane ritme 13-17. Bovendien zijn veel van deze assays afhankelijk analyseren van complexe interactiepatronen groepen van vliegen, met videotracking en software om de resulterende overvloed data te analyseren. Een beperking is echter de ontoegankelijkheid van deze testen aan de gemeenschap in het algemeen, en de beperkte kennis van de mechanismen die ten grondslag liggen aan erkenning van anderen. Met andere woorden, is de basis van de emissie van een signaal van een individu en de erkenning ervan door een ander nog steeds slecht begrepen 18.
Het D.: In tegenstelling, vliegen ook een eenvoudige gedrag, sociale vermijding, waar individuen te stappen van een signaal uitgezonden door benadrukte vliegen vertonen Melanogaster Stress Odorant of DSO 19. In een high-throughput assay kan dit gedrag worden gekwantificeerd als het vermijden van een spanning signaal uitgezonden door andere vliegen of sociale vermijden 19. Vliegen is in een T-doolhof apparaat geplaatst en de keuze om een flesje dat DSO voorkomen. Met dit assay, CO 2 bleek een component van de DSO en een deel van de neurale circuits noodzakelijk is om aan CO 2 werd ontleed 19 zijn.
De sociale vermijding test hier gepresenteerde is vergelijkbaar conceptueel de eenvoudige gedrags assays ontwikkeld in Seymour Benzer het laboratorium dat generaties onderzoekers nodig om complexe gedragingen 20 ontleden. Analyse van sociale vermijding kan worden uitgevoerd kosteneffectief behulp van de T-doolhof test, waardoor bredere studie van sociaal gedrag. Bijvoorbeeld met behulp van deze test we onlangs aangetoond dat verschillende genetische risico voor autisme contrasterende effecten in sociale behavior assays. Mutanten voor een kandidaat voor autisme – neurobeachin 21,22 – heden tekortkomingen zowel in de sociale ruimte (elders 23 beschreven) en sociale vermijding 24. Abnormale dopaminerge signalering wordt voorgesteld een rol spelen bij de etiologie van autisme bij mensen 25,26. In tegenstelling tot de resultaten verkregen met neurobeachin, vonden we dat sociale vermijden prestatie was niet beïnvloed door verhoogde of verlaagde niveaus van de Drosophila vesiculaire monoamine transporter (VMAT) in dopaminerge cellen, hoewel sociale ruimte rechtstreeks gecorreleerd met deze niveaus van VMAT 27. De contrasterende resultaten verkregen met neurobeachin en VMAT onderstrepen de mogelijkheid tot identificatie van verschillende vormen van asociaal gedrag en daarmee de verschillende onderliggende neurale circuits moduleren van de respons op anderen.
Dit protocol beschrijft een gedetailleerde procedure voor de sociale vermijding test. Canton-S zal alleen voorkomen dat een flacon waarin vliegt eerder mechanisch geweest benadrukt, en dat seks en het aantal responders heeft geen invloed op dat sociale vermijding prestaties. Echter, de genetische achtergrond van de respondenten heeft een grote invloed.
