Here we report a protocol to measure oxidative stress in living zebrafish embryos. This procedure allows reactive oxygen species (ROS) detection in both whole embryo tissues and single-cell populations. This protocol will accomplish both qualitative and quantitative analyses.
Hoge niveaus van reactieve zuurstof species (ROS) kan een verandering van cellulaire redox toestand naar oxidatieve stress situatie veroorzaken. Deze situatie veroorzaakt oxidatie van moleculen (lipiden, DNA, eiwit) en leidt tot celdood. Oxidatieve stress ook invloed op de progressie van verscheidene pathologische aandoeningen zoals diabetes, retinopathieën, neurodegeneratie, en kanker. Derhalve is het belangrijk om instrumenten te definiëren oxidatieve stress condities te bestuderen niet alleen op het niveau van individuele cellen, maar ook in de context van hele organismen. Hier beschouwen we de zebravis embryo als een nuttig in vivo systeem om dergelijke studies uit te voeren en presenteren een protocol om in vivo oxidatieve stress te meten. Profiteren van fluorescerende probes ROS en zebravis transgene fluorescerende lijnen, ontwikkelen we twee verschillende methodes om oxidatieve stress te meten in vivo: i) een "hele embryo ROS-detectiemethode" voor de kwalitatieve meting van oxidatieve stress en ii) een "eencellige ROS detectiemethode "voor kwantitatieve metingen van oxidatieve stress. Hierin tonen we de werkzaamheid van deze procedure door het verhogen oxidatieve stress in weefsels door oxyderende agens en fysiologische of genetische methoden. Dit protocol is vatbaar voor voorwaartse genetische screens en het zal adres oorzaak-gevolg relaties van ROS helpen in diermodellen van oxidatieve stress-gerelateerde ziekten zoals neurologische aandoeningen en kanker.
Oxidatieve stress is specifiek gedefinieerd als een aandoening die het gevolg van een onevenwichtige cellulaire redox toestand. Het complex redoxreacties die routinematig gebeuren in cellen bepalen de cellulaire redox-toestand. Redoxreacties omvatten alle chemische reacties die bestaan uit de overdracht van elektronen tussen de atomen van biologische moleculen produceren reductie en oxidatie van moleculen (bijvoorbeeld redoxreacties). Deze reacties worden gekatalyseerd door elektronische sturing soorten (pro-oxidatieve deeltjes), die worden gekenmerkt door een extreme structurele instabiliteit en spontane activatie van onevenwichtige elektronen die uit met naburige biomoleculen. Deze onregelmatige reacties resulteren in DNA-schade, eiwit carboxylatie en vetverbranding, en uiteindelijk leiden tot celdood 1. Verhoogde niveaus van oxidatieve stress zijn geassocieerd met veroudering en de progressie van verschillende pathologische toestanden 2. Oxidatieve stress heeftgerapporteerd verantwoordelijk voor vasculaire veranderingen in diabetes en cardiovasculaire aandoeningen 3,4 te zijn. Het speelt ook een cruciale rol in neuronale degeneratie bij de ziekte van Alzheimer en Parkinson 5. Bovendien is oxidatieve stress is aangetoond als een kritieke factor in het bestuur van de progressie van kanker en metastatische gebeurtenissen 6,7. Bovendien kan ontsteking en immuunreacties en verdere steun oxidatieve stress 8.
In levende cellen, worden pro-oxidatieve deeltjes afkomstig uit zuurstof (ROS, reactieve zuurstofspecies) of stikstof (RNS, reactieve stikstof species). ROS zijn de hydroxylgroep, de superoxide anion (OH.) (O 2 -) en waterstofperoxide (H 2 O 2). De primaire RNS is stikstofoxide (NO.). Een reeks van secundaire reactieve species kunnen worden gegenereerd door spontane interactie between ROS en RNS of vrije metalen ionen 9. Bijvoorbeeld, het superoxide anion reageert met lachgas te peroxynitraat (ONOO -) vorm, terwijl H 2 O 2 reageren met Fe2 + genereert hydroxylradicalen. ROS en RNS, vanwege hun vermogen om te reageren met verschillende biomoleculen, worden beschouwd als een gevaarlijke bedreiging voor het onderhoud van de fysiologische redox toestand 10. Handhaving van de redox toestand cellen zijn uitgerust met een reeks ontgiftende anti-oxidant moleculen en enzymen. Het superoxide dismutase (SOD), katalase, glutathion peroxidase en Peroxiredoxins wezen vormen de anti-oxidant enzymatische-arsenaal dat cellulaire bescherming biedt tegen pro-oxidatieve soorten, waaronder H 2 O 2, OH en OONO -. 11. Ook anti-oxidant moleculen zoals vitamine C en E, polyfenolen en CoenzymeQ10 (CoQ10) zijn van cruciaal belang voor ROS en de gevaarlijke de lessenderivaten 12,13. Echter, een overmatige productie van ROS en RNS, of disfunctie in de anti-oxidant systeem, verschuift de cellulaire redox-toestand naar oxidatieve stress 14.
Naast hun negatieve connotatie, kan ROS verschillende fysiologische rol spelen in de cellen van verschillende oorsprong. Cellen normaal produceren ROS als signaalmoleculen te bemiddelen normale biologische gebeurtenissen zoals afweer en wondheling 15-17. Reactieve species worden gewoonlijk geproduceerd in cellen door intracellulaire enzymen zoals NOx (NADPH oxidase) en XO (xanthine oxidase) in reactie op signalering factoren, groeifactoren, en intracellulaire schommelingen van calcium 18,19. Er werd gemeld dat ROS differentieel kan moduleren de activiteit van belangrijke nucleaire factoren zoals p53 of cellulaire componenten zoals de ATM-kinase, de regulering van de respons op DNA-schade 20. Analoog ROS beïnvloeden sterk cellulaire signalering door te bemiddelen the oxidatie en inactivering van eiwit tyrosine fosfatasen (PTP's), die zijn gevestigd als kritische regulatoren van signaaltransductie 21. Bovendien proteomics gebaseerde methoden tonen dat RNS ook verantwoordelijk zijn voor specifieke proteïne modificaties en veranderingen van moleculaire signalering. RNS reageren met de cysteine thiolgroepen te wijzigen in S-nitrothiols (SNO) en triggering moleculaire wegen gelijktijdig met pathologische, zoals ontsteking en auto-immuunziekten 22,23.
Aangezien celcultuurexperimenten gedeeltelijk reproduceren veelheid van factoren handelen in vivo, is het van groot belang redox studies uitgevoerd in diermodellen 24,25. Hiertoe is de zebravis beschouwd als een geschikte gewervelde diermodel oxidatieve stress dynamiek 26 bestuderen. De zebravis is een nieuw model systeem dat geeft een aantal voordelen om te studeren cellulaire en genetische gebeurtenissen tijdens gewervelde development en ziekte. Grote clusters van embryo's kunnen worden gegenereerd en wekelijks beschikbaar voor experimentele behoeften. Bovendien is de buitengewone optische helderheid van zebravis embryo's, evenals hun kleine formaat, kan via een cell imaging en dynamisch volgen in een hele organismen 27. In de afgelopen tien jaar hebben een aanzienlijk aantal zebravismutanten gegenereerd te modelleren van de menselijke pathologische aandoeningen zoals kanker en genetische ziekten 28-31. Het belangrijkste is, is een veelheid van transgene lijnen geproduceerd om uitgebreide mogelijkheden van genetische en biologische manipulaties 32 mogelijk te maken. Zo worden transgene weefselspecifieke zebravislijnen regelmatig gebruikt voor in vivo studies. Deze lijnen drukken een fluorescent eiwit onder de controle van een geselecteerde promoter, die de mogelijkheid om enkele cellen in vivo te identificeren, evenals de anatomische structuur die zij bevatten.
Verschillende toxicologische studies hebben reeds gebruikt thij zebravis over het in vivo effect van chemische stoffen op redox homeostase evalueren, suggereert de geschiktheid van deze gewervelde als een diermodel voor het gebied van geneesmiddelen en oxidatieve stress 33-35. Hoewel sommige fluorescerende probes getest om te controleren oxidatieve stress in zebravis larven 36,37, er geen vaste bepalingen om de niveaus van oxidatieve stress in zebravis weefsels en levende cellen te detecteren en te meten. Hier beschrijven we een werkwijze voor in vivo kwantificatie van oxidatieve stress in levende cellen van de zebravis embryo. Imaging gereedschappen, FACS-sortering, fluorescente probes en pro-oxidatieve voorwaarden wordt gecombineerd tot een eenvoudige test voor de detectie en kwantificering van oxidatieve deeltjes in zebravis embryo's en weefsels genereren.
Kritische stappen
De procedure voor oxidatieve stress detectie in zebravis embryo hierin beschreven omvat twee verschillende methoden. De hele mount ROS-detectiemethode is vooral een kwalitatieve test voor ROS-detectie, terwijl de cel ROS-detectie methode is de specifieke kwantitatieve metingen (figuur 1).
