A live cell fluorescent protein based method for illuminating cellular vacuoles (inclusions) containing Chlamydia is described. This strategy enables rapid, automated determination of Chlamydia infectivity in samples and can be used to quantitatively investigate inclusion growth dynamics.
The obligate intracellular bacterium Chlamydia elicits a great burden on global public health. C. trachomatis is the leading bacterial cause of sexually transmitted infection and also the primary cause of preventable blindness in the world. An essential determinant for successful infection of host cells by Chlamydia is the bacterium’s ability to manipulate host cell signaling from within a novel, vacuolar compartment called the inclusion. From within the inclusion, Chlamydia acquire nutrients required for their 2-3 day developmental growth, and they additionally secrete a panel of effector proteins onto the cytosolic face of the vacuole membrane and into the host cytosol. Gaps in our understanding of Chlamydia biology, however, present significant challenges for visualizing and analyzing this intracellular compartment. Recently, a reverse-imaging strategy for visualizing the inclusion using GFP expressing host cells was described. This approach rationally exploits the intrinsic impermeability of the inclusion membrane to large molecules such as GFP. In this work, we describe how GFP- or mCherry-expressing host cells are generated for subsequent visualization of chlamydial inclusions. Furthermore, this method is shown to effectively substitute for costly antibody-based enumeration methods, can be used in tandem with other fluorescent labels, such as GFP-expressing Chlamydia, and can be exploited to derive key quantitative data about inclusion membrane growth from a range of Chlamydia species and strains.
Infectieziekten veroorzaakt door species van de intracellulaire bacterie Chlamydia wekken grote belasting voor wereldwijde gezondheid, waaronder seksueel overgebrachte ziekte, eileiderontsteking, blindheid, longontsteking en eventueel atherosclerose 1-4. Het vermogen van Chlamydia om met de gastheercel, vanuit een vacuole (genaamd de opname), is een bepalende factor voor hun succesvolle infectie van cellen en de gastheer. De opname is een nieuwe pathogene compartiment dat chlamydia groei mogelijk maakt en wordt dynamisch aangepast gedurende de hele 2-3 dag ontwikkelings cyclus van Chlamydia 5. De obligate intracellulaire aard van chlamydiae presenteert talrijke uitdagingen voor de onderzoeksgemeenschap, in het bijzonder voor het direct bestuderen van de unieke biologie van de integratie. Een grote handicap is het onvermogen om efficiënt te visualiseren ofwel intracellulaire Chlamydia of hun opname door TL-aanpakes in levende cellen. Een recente ontdekking heeft eindelijk onthuld de middelen om GFP tot expressie C. genereren trachomatis 6; echter, is deze bevinding nog niet geleid tot de specifieke kenmerken van de integratie. Sommige technieken zijn beschreven voor labeling van bacteriën en insluitsels 7,8, maar ze vertonen tekortkomingen zoals niet-specificiteit, vergankelijkheid en gevoeligheid voor fotobleken. Een belangrijke ontdekking door onze groep werd een nieuwe strategie voor het verlichten van de opname met behulp van GFP tot expressie gastheercellen 9. Deze strategie rationeel exploiteert de intrinsieke ondoordringbaarheid van de opname membraan moleculen groter dan 520 Da 10. Wanneer cellen worden gemanipuleerd om stabiel drukken een bepaald cytosolische fluorescent eiwit (bijvoorbeeld GFP of mCherry), Chlamydia insluitsels zichtbaar met opmerkelijk heldere hun volledige uitsluiting van fluorescentie. Deze omgekeerde beeldvorming strategie maakt directe visualisatie van insluitsels voor alle Chlamydia species en kan gemakkelijk worden aangepast voor de meeste gastheercellen van belang. Als een demonstratie van het nut ervan, werd deze methode eerder gebruikt te onthullen en te definiëren de cellulaire exit routes voor Chlamydia spp 9.
Hier hebben we verder aan te tonen hoe deze methode wordt uitgevoerd, en kan worden benut om belangrijke kwantitatieve gegevens over de groei inclusie dynamiek ontlenen. Bovendien kan het effectief vervangen dure antilichamen gebaseerde inventarisatie werkwijzen en kan worden gebruikt in combinatie met andere fluorescente labels, zoals mKate2-expressie Chlamydia 11. Deze krachtige combinatie van instrumenten mogelijk maakt onderzoek van de fysische eigenschappen van de chlamydiale membraan integratie binnen levende gastheercellen.
Hier beschrijven we de experimentele strategie voor het genereren van fluorescente gastheercellen voor real time visualisatie en analyse van Chlamydia inclusies. Deze vacuole visualisatie benadering verleent het krachtige vermogen te verlichten, volgen en kwantitatief meten van de dynamische eigenschappen van Chlamydia inclusies tussen populaties van cellen of enkele cellen in de tijd. Chlamydia inclusies in fluorescent eiwit gelabelde cellen opvallend goed gedefinieerd, zodat ze gemakkelijk geïdentif…
