Het protocol beschrijft een efficiënt en reproduceerbaar modelsysteem om te studeren herpes simplex virus type 1 (HSV-1) latentie en reactivatie. De test maakt gebruik van homogene sympathiek neuron culturen en maakt het mogelijk om moleculaire dissectie van virus-neuron interacties met behulp van een scala aan hulpmiddelen met inbegrip van RNA-interferentie en expressie van recombinante eiwitten.
Herpes simplex virus type 1 (HSV-1) wordt een leven lang latente infectie in de perifere neuronen. Dit latente reservoir is de bron van terugkerende reactivering gebeurtenissen te verzekeren dat de en bijdragen aan klinische ziekte. De huidige antivirale middelen hebben geen invloed op de latente reservoir en er zijn geen vaccins. Terwijl de moleculaire details van lytische replicatie zijn goed gekarakteriseerd, mechanismen die de latentie in neuronen blijven ongrijpbaar. Onze huidige kennis van latentie is afgeleid van in vivo studies met kleine diermodellen, die zijn onmisbaar voor het definiëren van virale gen-eisen en de rol van de immuunrespons. Het is echter niet te onderscheiden specifieke effecten op het virus neuron relatie van meer algemene gevolgen van infectie gemedieerd door immuun-of niet-neuronale ondersteuning cellen in levende dieren. Daarnaast dierproeven is kostbaar, tijdrovend, en beperkt in termen van beschikbare opties voor het manipuleren van gastheerprocessen. Om deze beperkingen te overwinnen, is een neuron-only systeem hard nodig dat reproduceert de in vivo eigenschappen van latentie en reactivatie, maar biedt de voordelen van weefselkweek op het gebied van homogeniteit en toegankelijkheid.
Hier presenteren we een in vitro model gebruik te maken van gekweekte primaire sympathische neuronen van de rat superieure cervicale ganglia (SCG) (figuur 1) tot HSV-1 latentie en reactivatie dat past studie zijn de meeste zo niet alle van de gewenste criteria. Na verwijderen van niet-neuronale cellen, nagenoeg homogene TrkA + neuron kweken geïnfecteerd met HSV-1 in aanwezigheid van acyclovir (ACV) tot lytische replicatie onderdrukken. Na ACV verwijdering, niet-productieve HSV-1 infecties die trouw vertonen aanvaarde kenmerken van latency op efficiënte wijze vastgesteld. Met name lytische mRNAs, eiwitten en infectieuze virus meer detecteerbaar, zelfs in de afwezigheid van selectie, maar latentie-geassocieerde transcript (LAT) expression blijft in neuronale kernen. Virale genomen worden gehandhaafd op gemiddeld aantal kopieën van 25 per neuron en kan worden geïnduceerd om productief te repliceren door te interfereren met PI3-kinase / Akt signalisatie of de eenvoudige intrekking van zenuwgroeifactor 1. Een recombinant HSV-1 codering EGFP gefuseerd met het virale eiwit US11 lytische zorgt voor een functionele, real-time marker voor replicatie voortvloeien uit de reactivering die gemakkelijk wordt gekwantificeerd. Naast chemische behandelingen kunnen genetische methoden zoals RNA-interferentie of genafgifte via lentivirale vectoren succes worden toegepast op het systeem waarmee mechanistische studies die zeer moeilijk, zo niet onmogelijk, bij dieren. Kortom, de SCG op basis van HSV-1 latency / reactivering systeem zorgt voor een krachtige, noodzakelijk instrument om de moleculaire mechanismen die HSV1 latentie en reactivatie in neuronen, van oudsher een puzzel in de virologie waarvan de oplossing kan nieuwe inzichten bieden in het ontwikkelen van nieuwe therapieën die te ontrafelen doelstelling thij latent herpesvirus reservoir.
Deze primaire neuron cultuur en infectie systeem biedt een eenvoudige en effectieve methode om de moleculaire mechanismen die ten grondslag liggen HSV-1 latentie en reactivatie te verkennen. Het systeem trouw recapituleert de algemeen aanvaarde kenmerken van vertraging in de zin van zowel menselijke infecties en in live-diermodellen.
